The non-receptor tyrosine phosphatase PTPN22 has a vital function in inhibiting antigen-receptor signaling in T cells, while polymorphisms in the PTPN22 gene are important risk alleles in human autoimmune diseases. We recently reported that a key physiological function of PTPN22 was to prevent naïve T cell activation and effector cell responses in response to low affinity antigens. PTPN22 also has a more general role in limiting T cell receptor-induced proliferation. Here we present new data emphasizing this dual function for PTPN22 in T cells. Furthermore, we show that T cell activation modulates the expression of PTPN22 and additional inhibitory phosphatases. We discuss the implication of these findings for our understanding of the roles of PTPN22 in regulating T cell responses and in autoimmunity. 相似文献

When human observers view dynamic random noise, such as television ''snow'', through a curved or annular aperture, they experience a compelling illusion that the noise is moving smoothly and coherently around the curve (the ''omega effect''). In several series of experiments, we have investigated the conditions under which this effect occurs and the possible mechanisms that might cause it. We contrast the omega effect with ''phi motion'', seen when an object suddenly changes position. Our conclusions are that the visual scene is first segmented into objects before a coherent velocity is assigned to the texture on each object''s surface. The omega effect arises because there are motion mechanisms that deal specifically with object rotation and these interact with pattern mechanisms sensitive to curvature. 相似文献

Following the discovery of synergistic action between oxacillin and manuka honey against methicillin-resistant Staphylococcus aureus, this study was undertaken to search for further synergistic combinations of antibiotics and honey that might have potential in treating wounds. Fifteen antibiotics were tested with and without sublethal concentrations of manuka honey against each of MRSA and Pseudomonas aeruginosa using disc diffusion, broth dilution, E strip, chequerboard titration and growth curves. Five novel antibiotic and manuka honey combinations were found that improved antibacterial effectiveness in vitro and these offer a new avenue of future topical treatments for wound infections caused by these two important pathogens. 相似文献

Middle EoceneEurotamandua from Germany has been considered to be the oldest anteater and the only known Old World xenarthran, based on numerous derived features shared with living Myrmecophagidae or Xenarthra. Many of these features are ambiguous, however, and comparison with early Tertiary Palaeanodonta reveals several detailed, derived resemblances in forelimb anatomy. Abstract— β- N -Acetyl D-galactosaminidase was studied in isolated neuronal and neuropil fractions from cerebral cortex and subcellular fractions derived from them. Although the enzyme activity evinced some latency properties, its subcellular distribution pattern was broader than that observed with other acid hydrolases. By contrast with nine other acid hydrolases, it was more active in neuropil than neuronal fractions (neuronal/neuropil activity ratio 0.63). This ratio was preserved in lysosomal subfractions derived from the isolated cell fractions. The data is taken as further evidence for the microheterogeneity of lysosomal particles from the brain. 相似文献
