Freshly fertilized ova of brown trout, Salmo trutta L., were exposed to all possible mixtures of Al (6000 nmol 11 ), Cu (80 nmol 1−1 ), Pb (50 nmol 1−1 ) and Zn (300 nmol 1 1 ). In a separate experiment, newly hatched brown trout yolk-sac fry were exposed to Mn (1500 nmol 1−1 ), Fe (2500 nmol 1 1 ), Ni (200 nmol 1−1 ) or Cd (4 nmol 1 1 ), separately, and in mixtures with either Al or Cu. Both experiments were conducted in flowing, artificial softwater media nominally at pH 5.6 [Ca] 20 μmol 1 1 and 10° C.
Mortalities were high in fry subjected to treatments which contained both Al and Cu (31–72%), and to the Cu + Fe treatment (78%) compared with those from the other trace metal mixtures (0–22%). In all the treatments tested, fry exposed to trace metal mixtures containing Al and/or Cu had reduced whole body Ca, Na and K content, and seriously impaired skeletal calcification. Whole body Mg content was variable. In trace metal mixtures which contained Cu but not Al, the effects on fry survival and whole body mineral content were in general more deleterious than the corresponding mixtures but with Al present rather than Cu. The presence of Pb and/or Zn in mixtures with Al and/or Cu had a slight ameliorative effect in terms both of fry survival and whole body mineral content. 相似文献

1. In adult rats under urethane anesthesia, a vast majority of spontaneously active neurons in the frontoparietal cortex undergo a prolonged depression of their firing rate upon microiontophoretic application of 5-HT. 2. In 5,7-DHT-deafferented cortex, this effect is of a longer duration (mean 14 min) than in controls (mean 5 min). Moreover, small ejection currents of 5-HT are sufficient to induce a prolonged depression of the firing rate. 3. In PCPA-pretreated rats, there are no changes in the responsiveness to 5-HT. 4. In control and PCPA-pretreated rats, blocking of the 5-HT reuptake with fluoxetine increases the duration of responses to 5-HT (mean 15 min), whereas small ejection currents remain without effect. 5. These data indicate that, in the cerebral cortex, denervation supersensitivity to 5-HT results primarily from the removal of 5-HT afferents, and not from depletion of their 5-HT content. The enhanced responsiveness to microiontophoresed 5-HT appears to be due to a suppression of reuptake mechanisms, mainly responsible for the prolongation of 5-HT effects, and to a modification of receptors on target cells, which accounts for their greater sensitivity to 5-HT. 相似文献

We describe a chemical proteomics approach to profile the interaction of small molecules with hundreds of endogenously expressed protein kinases and purine-binding proteins. This subproteome is captured by immobilized nonselective kinase inhibitors (kinobeads), and the bound proteins are quantified in parallel by mass spectrometry using isobaric tags for relative and absolute quantification (iTRAQ). By measuring the competition with the affinity matrix, we assess the binding of drugs to their targets in cell lysates and in cells. By mapping drug-induced changes in the phosphorylation state of the captured proteome, we also analyze signaling pathways downstream of target kinases. Quantitative profiling of the drugs imatinib (Gleevec), dasatinib (Sprycel) and bosutinib in K562 cells confirms known targets including ABL and SRC family kinases and identifies the receptor tyrosine kinase DDR1 and the oxidoreductase NQO2 as novel targets of imatinib. The data suggest that our approach is a valuable tool for drug discovery. 相似文献

Shoot regeneration after prescribed burning or following the freezing temperatures of winter was monitored for nineteen heathland species present in an Arctostaphyleto-Callunetum community in northeast Scotland. Species whose renewal buds were near the surface of the ground started to grow earlier in the spring than species with renewal buds above the surface, but grouping species according to the position of their renewal bud (i.e. their life-form) did not account for all of the interspecific variation apparent. In the case of shoot regeneration after fire, species whose renewal buds were destroyed by fire because they were above-ground started to regenerate about the same time as species with belowground buds, protected from fire, but reached their maximum frequency of occurrence later. Grouping species by life-form was of limited value as a means of interpreting this interspecific variation in the timing of shoot regeneration after fire. It would be unwise to use plant life-form as the sole basis for interpreting or predicting a species' response to temperature stress when extreme temperatures occur regularly, as they do in heathland. The possible use of other plant traits to interpret and predict interspecific variation in the regeneration rate of heathland plants is discussed.Nomenclature follows Tutin et al. To determine whether soil disturbance by digging and burrowing mammals altered community structure and the rate of succession in a midsuccessional abandoned pasture, species richness, composition and relative abundance were monitored over a two year period both on and off artificially created earth mounds (100, 900, 8100 cm2). Mean species richness increased by up to two species per small mound (100 cm2) and by up to four species per large mound (8100 cm2). However, increased species richness was evident for less than two years. Initially, up to sixteen of the twenty species present occurred more often on earth mounds than off mounds, with two of these species found only on large mounds (8100 cm2). After two years, there was little or no significant difference in species composition and relative frequency on and off earth mounds. Experimental soil disturbance temporarily altered community structure simply by increasing space available for colonization since light, nutrient and water supply did not increase significantly on mounds. Soil disturbance can increase species richness and change species' relative frequency in disturbances as small as 100 cm2 but such changes were likely too small and short lived to alter permanently the structure and rate of succession in the abandoned pasture studied here. 相似文献

The cloning of genes for complex traits in polyploid plants that possess large genomes, such as hexaploid wheat, requires an efficient strategy. We present here one such strategy focusing on the homologous pairing suppressor (Ph1) locus of wheat. This locus has been shown to affect both premeiotic and meiotic processes, possibly suggesting a complex control. The strategy combined the identification of lines carrying specific deletions using multiplex PCR screening of fast-neutron irradiated wheat populations with the approach of physically mapping the region in the rice genome equivalent to the deletion to reveal its gene content. As a result, we have located the Ph1 factor controlling the euploid-like level of homologous chromosome pairing to the region between two loci (Xrgc846 and Xpsr150A). These loci are located within 400 kb of each other in the rice genome. By sequencing this region of the rice genome, it should now be possible to define the nature of this factor. 相似文献

The interaction betweenTrybliographa rapae andAleochara bilineata, 2 parasitoids of the cabbage root fly, is discussed. Larvae ofA. bilineata could not differentiate between cabbage root fly pupae containingT. rapae in its endoparasitic state and unparasitized pupae but could recognize pupae containingT. rapae once the latter had reached its ectoparasitic state. Attack byA. bilineata whileT. rapae was still in its endoparasitic state usually resulted in the staphylinid killing the eucoilid. IfT. rapae had reached the ectoparasitic state before the host pupa was attacked byA. bilineata larvae the eucoilid survived attack by the beetle larva. Multiparasitism, however, resulted in increased levels of mortality
of both parasitoid populations.
