Repeated mild heat shock (RMHS) has beneficial hormesis-like effects on various characteristics of human skin fibroblasts undergoing replicative senescence in vitro. We have tested whether RMHS could reduce the accumulation of oxidized and glycoxidized proteins, which is a major age-related change. Levels of carbonylated proteins, furosine, N-carboxymethyl-lysine-rich proteins and advanced glycation end products increased during serial passaging of fibroblasts in culture. However, the extent of accumulation of oxidized and glycoxidized proteins was significantly reduced in RMHS cells. The basal concentration of reduced glutathione was higher and that of oxidized glutathione was lower in RMHS cells. Whereas the basal level of heat shock protein HSP27 decreased in both RMHS and control cells during serial passaging, the increase of the basal level of HSP70 with increasing passage level was significantly higher in RMHS cells. These results show that the slower accumulation of damaged proteins in fibroblasts exposed to RMHS results partly from the increased ability of these cells to cope with oxidative stress, and to synthesize HSP responsible for protein capping and refolding.  相似文献

Summary Self-diffusion coefficients of zinc and iron were determined in acid soil of Palampur and alluvial soil of Ludhiana under varying pH, CaCO3, moisture, carrier and phosphorus levels. Increase in pH caused tremendous reductions in self-diffusion coefficients (Da) of both zinc and iron in soil. The selfdiffusion coefficients of both these elements were drastically reduced as a result of CaCO3 application. The Da values of zinc and iron increased with the decrease in moisture tension and increase in carrier and phosphorus levels. The decrease in Da values were associated with increase in capacity factor.  相似文献

This research examined the application of the maximum quantum efficiency (F v/F m) and relative electron transport rate versus irradiance curves (rETR) as a rapid, sensitive assessment of Lake Erie phytoplankton nutrient status. I evaluated the potential benefits of the variable fluorescence parameters by comparing these parameters with chemical and physiological nutrient status assays. I tested the hypothesis that F v/F m and rETR curves could diagnose nutrient status in natural lake phytoplankton and be capable of discriminating which inorganic nutrient is limited temporally and spatially. F v/F m was on average highest in the more eutrophic west basin (WB) and lowest in the more oligotrophic central basin (CB). According to the chemical and physiological indicators, P deficiency was most severe in the CB during summer stratification and N deficiency was strongest in the WB during isothermal conditions. Like F v/F m, rETR at light saturation (rETRmax) and the initial slope of the rETR versus irradiance curve (α) decreased as the severity of N and P deficiency increased. Amendment with N or P stimulated increased F v/F m, rETRmax, and α in N- and P-limited samples, respectively, and abolished the photoinhibition apparent in rETR curves of nutrient-limited samples. These results supported the view that the N and P deficiency assays, and corresponding variations of variable fluorescence parameters, were valid indicators of widely variable N and P deficiency in the phytoplankton, and could be used to provide a promising tool in determining phytoplankton nutrient status. Contrary to my hopes, it did not appear that rETR–irradiance curves could discriminate between N and P deficiency. Identification of the most limiting nutrient still demanded additional information beyond the variable fluorescence measurements.  相似文献

In the present study, the effect of intracerebroventricular (icv) injection of GABA, its agonist--muscimol, and antagonist--picrotoxin, has been studied on histoenzymological alterations of acetylcholinesterase (AChE). butyrylcholinesterase (BuChE), monoamine oxidase (MAO), and succinic dehydrogenase (SDH) by cytophotometric technique. This study was conducted on medial preoptic area (mPOA), nucleus paraventricularis hypothalami (PVH), area lateralis hypothalami (LHA), nucleus dorsomedialis hypothalami (DMH), and nucleus ventromedialis hypothalami (VMH). Results showed that GABA and muscimol inhibited AChE, BuChE, MAO, and SDH in all the areas while picrotoxin stimulated these enzymes. These changes in enzyme activity by GABA, muscimol, and picrotoxin and their possible mode of action are discussed.  相似文献

A significant decline in amount of active elongation factor, EF-1 alpha, and in its catalytic activity was observed in cell-free extracts prepared from normal human diploid fibroblasts (MRC-5) and their SV40-transformed counterparts, after subjecting the cells to 60 min heat shock at different temperatures. Old MRC-5 cells which had become senescent on serial passaging were more sensitive to heat shock-related changes in activity and amounts of active EF-1 alpha than were rapidly proliferating normal and transformed cells.  相似文献

Vasoactive intestinal peptide and pituitary adenylate cyclase activating peptide have high affinity for VPAC1, VPAC2 and PAC1 receptors overexpressed on human cancer cells. Four potent analogues of these peptides, TP3939, TP3982, TP4200 and TP3805 were labeled with (64)Cu and evaluated ex vivo and in vivo to asses their biological activity and receptor specificity. The ultimate goal is to utilize (64)Cu analogues for positron emission tomography (PET) imaging of breast cancers in humans. Radiochemical purity of each analogue was >92%. The muscle relaxivity assay revealed IC(50) to be 5.3x10(-8) M, 4.4x10(-8) M, 8.1x10(-8) M, 8.1x10(-9) M and Kd values determined by receptor specific cell binding assays were 3.3 nM, 0.33 nM, 0.2 nM and 0.72 nM for TP3805, TP3939, TP3982, and TP4200 respectively. The receptor affinity, using human breast cancer tissues, was 10.93 times greater than normal breast tissues. RT-PCR confirmed increased VPAC1 receptor expression on human breast tumor cells over normal cells and corroborated with autoradiography data. The blood clearance was rapid and in vivo translocation of (64)Cu to plasma protein was <15%. Data demonstrate that these analogues are potent, have uncompromised biological activity and are worthy of further evaluation for accurate PET imaging of human breast cancers and in determining malignant and benign lesions.  相似文献

Studies were performed to determine the unknown status of PKC and RhoA/ROCK in the phorbol 12,13-dibutyrate (PDBu)-stimulated state in the human internal anal sphincter (IAS) smooth muscle cells (SMCs). We determined the effects of PDBu (10(-7) M), the PKC activator, on PKCα and RhoA and ROCK II translocation in the human IAS SMCs. We used immunocytochemistry and fluorescence microcopy in the basal state, following PDBu, and before and after PKC inhibitor calphostin C (10(-6) M), cell-permeable RhoA inhibitor C3 exoenzyme (2.5 μg/ml), and ROCK inhibitor Y 27632 (10(-6) M). We also determined changes in the SMC lengths via computerized digital micrometry. In the basal state PKCα was distributed almost uniformly throughout the cell, whereas RhoA and ROCK II were located in the higher intensities toward the periphery. PDBu caused significant translocation of PKCα, RhoA, and ROCK II. PDBu-induced translocation of PKCα was attenuated by calphostin C and not by C3 exoenzyme and Y 27632. However, PDBu-induced translocation of RhoA was blocked by C3 exoenzyme, and that of ROCK II was attenuated by both C3 exoenzyme and Y 27632. Contraction of the human IAS SMCs caused by PDBu in parallel with RhoA/ROCK II translocation was attenuated by C3 exoenzyme and Y 27632 but not by calphostin C. In human IAS SMCs RhoA/ROCK compared with PKC are constitutively active, and contractility by PDBu is associated with RhoA/ROCK activation rather than PKC. The relative contribution of RhoA/ROCK vs. PKC in the pathophysiology and potential therapy for the IAS dysfunction remains to be determined.  相似文献
