Ribosomal subunit assembly in the nucleolus is dependent on efficient targeting of ribosomal proteins (RPs) from the cytoplasm into the nucleus and nucleolus. Nuclear/nucleolar localization of a protein is generally mediated by one or more specific stretches of basic amino acids—nuclear/nucleolar localization signals (NLSs/NoLSs). Arabidopsis thaliana RPL23aA has eight putative NLSs/NoLSs (pNLSs/NoLSs). Here we mutated all eight NLS/NoLSs individually and in groups and showed, via transient expression in tobacco cells that nucleolar localization of RPL23aA was disrupted by mutation of various combinations of five or more pNLSs/NoLSs. Mutation of all eight pNLSs/NoLSs, a 50 % reduction in total basic charge of RPL23aA, resulted in a complete disruption of nucleolar localization, however, the protein can still localize to the nucleus. As no individual or specific combination of NoLSs was absolutely required for nucleolar localization, we suggest that nucleolar localization/retention of RPL23aA is dependent on the overall basic charge. In addition to the optimal basic charge conferred by these NoLSs, nucleolar localization/retention of RPL23aA also required a C-terminal putative 26S rRNA binding site. In contrast, in the RPs RPS8A and RPL15A, mutation of just two and three N-terminal pNLSs, respectively, disrupted both nuclear and nucleolar localization of these two RPs, indicating differential signal requirements for nuclear and nucleolar localization of the three Arabidopsis RPs RPL23aA, RPL15A and RPS8A. 相似文献

Summary The effect of different concentrations of CO2 on the germination of conidiospores of Aspergillus niger A 5 has been studied using Pardee's buffer mixtures which maintain constant CO2 tensions. The beneficial effect of CO2 on germination is maximum at 0.5% CO2 concentration, when 70–90% of the spores germinate within 6 hours, whereas in controls with air containing 0.03% CO2 there is only 15–20% germination at 6 hours. At higher CO2 concentrations this beneficial effect of CO2 on germination diminishes and at 3% there is a complete inhibition of spore germination.The spore density and the ph of the medium have a noticeable effect on germination rates in presence of 0.5% CO2. The objectives were to investigate the plasma lipid peroxidation and erythrocyte antioxidants status in workers exposed to nickel. The study groups comprised 69 nickel plating workers and 50 office workers residing in the same city, but away from the place of work of the study group subjects, considered as control group. Urinary nickel concentration was determined by graphite furnace atomic absorption spectrophotometry. The plasma lipid peroxidation and erythrocyte antioxidants were measured by spectrophotmetric methods. The plasma lipid peroxidation level was significantly increased in nickel-platers and their helpers as compared with controls. Erythrocyte antioxidants were significantly decreased in the nickel-platers compared with the controls. The level of plasma lipid peroxidation was positively and erythrocyte antioxidants were negatively and significantly correlated with the urine nickel levels. Multiple regression analysis assessed the oxidative stress associated with nickel and other potential confounding factors such as body mass index, the consumption of green vegetables, coffee, tea, smoking and alcohol consumption. Analysis showed that the lifestyle confounding factors: the consumption of green vegetables, smoking and alcohol, were not significantly associated with oxidative stress. The exposure to nickel, body mass index and coffee consumption were significantly associated with oxidative stress. The results show that the increased plasma lipid peroxidation and decreased erythrocyte antioxidants levels observed in nickel-exposed workers could be used as biomarkers of oxidative stress. 相似文献

Observations were made on a nest of Ropalidia cyathiformis consisting of three combs. The number of eggs, larvae, pupae and adults were monitored at about 3-day intervals for a 2-month period. The behaviour of the adults was observed with special reference to the proportion of time spent on each of the three combs, the proportion of time spent away from the nest site and the frequencies of dominance interactions and egg laying. The adults moved freely between the three combs suggesting that all of them and all the three combs belonged to one nest. However, most of the adults preferred combs 2 and 3 over comb 1. Of the 10 animals chosen for a detailed analysis of behaviour, seven spent varying periods of time away from the nest site and oRen brought back food or building material. Five of the 10 animals laid at least one egg each but two adults monopolized most of the egg-laying. The animals showed a variety of dominance interactions on the basis of which they have been arranged in a dominance hierarchy. The dominant individuals laid most of the eggs and spent little or no time foraging, while the subordinate individuals spent more time foraging and laid few eggs or none. It is argued that R. cyathiformis is different from R. marginata , the only other Indian social wasp whose behaviour has been studied, in being at a more primitive stage of social organization. 相似文献

A study was conducted with non-conventional ingredients to test their efficacy as fishmeal (FM) replacers in the diet of fringe- lipped carp. Labeo fimbriatus first feeding larvae and fry were reared for 30 and 60 days in indoor, 50 L, aerated, circular plastic tanks at 100 and 30 numbers tank−1, respectively. In the first feeding larvae to fry rearing experiment (Exp. 1), the fish were fed with either of the following isonitrogenous and isocaloric diets – live plankton, FM diet, green bottle fly (Lucilia sericata) larvae meal (GBFLM) diet and silkworm pupa (SWP) diet. The fry to fingerling rearing (Exp. 2), was also conducted using the same diets described above except live plankton. All compounded diets were formulated to contain 40% crude protein for the experiment 1 and 35% for experiment 2 and were fed ad libitum. Triplicate tanks were maintained for each treatment in both the experiments. In Exp. 1, the mean final weight of fry was higher with plankton and FM diets, while no difference (p > .05) was observed between FM and GBFLM diets. Weight of fish fed SWP diets was not statistically different from those fed GBFLM diet. No difference (p > .05) in final length, survival and condition factor was recorded. Analysis of digestive enzyme activity of whole fish revealed lower (p < .05) activity of amylase in fish fed plankton. In Exp. 2, no difference (p > .05) was observed between the different diet groups in terms of mean final weight, length, survival and condition factor. Analysis of digestive enzyme activity of whole fish revealed no difference (p > .05) in the activity of digestive enzymes between the treatments except a lower (p < .05) activity of trypsin in FM diet and lipase in FM and GBFLM diets. Since the survival and condition factors of animals is the most important aspect during nursery rearing, similar (p > .05) values recorded in different treatments indicate the possibility of incorporation of these non-conventional protein sources in the diet of L. fimbriatus during first feeding larvae to fry and fry to fingerling rearing. 相似文献

We have demonstrated that therapeutic administration of L-arginine (L-arg) (120 mg/kg) at +2 h of whole body hyperthermia (WBH) could rescue the mice from heatstroke-induced death. Studies were undertaken to elucidate the role of L-arg in the immunomodulation of the heat-stressed mice. Administration of L-arginine (L-arg), (120 mg/kg, i.p.), at +2 h of WBH, rescued the mice from heat-induced death and reduced the hypothermia. At +4 and +24 h of WBH, levels of IL-1beta, IFN-gamma, nitrite, TNF-alpha, IL-4, TGF-beta1, inducible form of nitric oxide synthase (iNOS), and corticosterone significantly increased compared to the sham group. The elevated levels of Th(1) cytokines, namely TNF-alpha, IL-1beta, IFN-gamma, nitrite, and iNOS, decreased significantly both at +4 and +24 h of WBH, following L-arg administration. However, L-arg administration did not reduce the increased levels of Th(2) cytokines, namely IL-4 and TGF-beta1, in WBH mice at +4 h of WBH. L-arg administration significantly increased the levels of Th(2) cytokines at +24 h of WBH, compared to the saline-treated WBH mice. L-arg administration significantly increased both the splenic and hepatic arginase activity at +4 and +24 h of WBH compared to the saline-treated WBH mice. L-NAME treatment at +2 h of WBH and anti-TGF-beta antibody treatment at 0 h of WBH significantly increased the mortality compared to the saline-treated WBH mice. Altered liver histopathology was attenuated following the administration of L-arg at +2 h of WBH. These results suggest that therapeutic administration of L-arg at appropriate concentration and time attenuates the acute inflammatory response, leading to the rescue of mice from heatstroke. 相似文献
