Photoaffinity labeling of rat brain membranes with [125I]ANPAA-apamin incorporated radioactivity into polypeptides of 86 and 59 kDa and occasionally a more weakly labeled component of 45 kDa. These polypeptides were immunoprecipitated with anti-apamin antibodies and treated with glycosidases. Neither the 86 nor the 59 kDa polypeptide appeared to be N-glycosylated. Partial proteolytic mapping of affinity labeled polypeptides with chymotrypsin or V8 protease generated an identical pattern. These results suggest that the 59 and 45 kDa components are not additional subunits of an oligomeric protein but result from cleavage of the 86 kDa polypeptide. 相似文献
After cell entry, HIV undergoes rapid transport toward the nucleus using microtubules and microfilaments. Neither the cellular cytoplasmic components nor the viral proteins that interact to mediate transport have yet been identified. Using a yeast two-hybrid screen, we identified four cytoskeletal components as putative interaction partners for HIV-1 p24 capsid protein: MAP1A, MAP1S, CKAP1, and WIRE. Depletion of MAP1A/MAP1S in indicator cell lines and primary human macrophages led to a profound reduction in HIV-1 infectivity as a result of impaired retrograde trafficking, demonstrated by a characteristic accumulation of capsids away from the nuclear membrane, and an overall defect in nuclear import. MAP1A/MAP1S did not impact microtubule network integrity or cell morphology but contributed to microtubule stabilization, which was shown previously to facilitate infection. In addition, we found that MAP1 proteins interact with HIV-1 cores both in vitro and in infected cells and that interaction involves MAP1 light chain LC2. Depletion of MAP1 proteins reduced the association of HIV-1 capsids with both dynamic and stable microtubules, suggesting that MAP1 proteins help tether incoming viral capsids to the microtubular network, thus promoting cytoplasmic trafficking. This work shows for the first time that following entry into target cells, HIV-1 interacts with the cytoskeleton via its p24 capsid protein. Moreover, our results support a role for MAP1 proteins in promoting efficient retrograde trafficking of HIV-1 by stimulating the formation of stable microtubules and mediating the association of HIV-1 cores with microtubules. 相似文献
Parasites can play an important role in the dynamics of host populations, but empirical evidence remains sparse. We investigated
the role of bot fly (Cuterebra spp.) parasitism in red-backed voles (Myodes gapperi) by first assessing the impacts of the parasite on the probability of vole survival under stressful conditions as well as
on the reproductive activity of females. We then identified the main factors driving both the individual risk of infection
and the abundance of bot flies inside red-backed voles. Finally, we evaluated the impacts of bot fly prevalence on the growth
rate of vole populations between mid-July and mid-August. Thirty-six populations of red-backed voles were sampled in the boreal
forest of Québec, Canada. The presence and the abundance of parasites in voles, two host life history traits (sex and body
condition), three indices of habitat complexity (tree basal area, sapling basal area, coarse woody debris volume), and vole
abundance were considered in models evaluating the effects of bot flies on host populations. We found that the probability
of survival of red-backed voles in live traps decreased with bot fly infection. Both the individual risk of infection and
the abundance of bot flies in red-backed voles were driven mainly by vole abundance rather than by the two host life history
traits or the three variables of habitat complexity. Parasitism had population consequences: bot fly prevalence was linked
to a decrease in short-term growth rate of vole populations over the summer. We found that bot flies have the potential to
reduce survival of red-backed voles, an effect that may apply to large portions of populations.

About 200 micromammal isolated teeth (rodents, lagomorphs) originating from the middle Orleanian locality of Béon 2 (Montréal-du-Gers, SW France) are described. The rodent fauna is dominated by myomorphs (7 species), including the cricetid Democricetodon aff. hispanicus Fahlbusch, the melissiodontid Melissiodon sp., the glirids Peridyromys murinus (Pomel), Pseudodryomys aff. ibericus, Pseudodryomys aff. simplicidens, and Glirudinus modestus (Dehm), and the eomyid Ligerimys aff. florancei. Sciuromorphs are represented only by Heteroxerus rubricati Crusafont, Villalta and Truyols. Within lagomorphs, 2 dental morphs referred to the ochotonid Prolagus Pomel are identified in Béon 2. They correspond to Prolagus oeningensis (König) and P. aff. vasconiensis. On a biostratigraphical point of view, this study confirms the location of Béon 2 at the early part of the MN4; this locality is older than other regional sites such as Pellecahus, Béon 1, La Romieu, and Bézian (MN4b), and it is coeval to Artenay, in the Loire basin. In particular, glirids and lagomorphs from Béon 2 testify to close relationships with micromammal localities referred to the MN3 biozone. 相似文献
Eight elasmotheriine rhinocerotid species have been described from the Middle Miocene of China and Mongolia. In this paper a revised taxonomy is presented based on direct observation and comparison of the available material. A phylogenetic analysis based on 282 morphological characters has led to the reappraisal of Procoelodonta Matthew, 1931, Caementodon Heissig, 1972 and Huaqingtherium Huang & Yan, 1983. The genus Procoelodonta is split into three subgenera: P. ( Procoelodonta ) Matthew, 1931, P. ( Begertherium ) Beliaeva, 1971, and P. ( Pasalarhinus subg. n). The genus Caementodon is split into two subgenera: C. ( Caementodon ) Heissig, 1972 and C. ( Beliajevina ) Heissig, 1974. Four species are assumed to have occurred in the Middle Miocene within the area studied: Procoelodonta ( Procoelodonta ) mongoliense (Osborn, 1924), ' P. ' ( Begertherium ) borissiaki (Beliaeva, 1971), ' Caementodon ' ( Beliajevina ) fangxianense (Yan, 1979) and Huaqingtherium lintungense (Zhai, 1978) (=' Caementodon tongxinensis ' Guan, 1988 =' Huaqingtherium qiui ' Guan, 1993 =' Hispanotherium tungurense ' Cerdeño, 1996). Shennongtherium hypsodontus Huang & Yan, 1983 is removed from the Elasmotheriina, owing to dental characters which suggest that it is a teleoceratine. The distribution of the main characters later seen in Elasmotherium is briefly discussed. The persistence and diversity of the Elasmotheriina throughout the Middle Miocene help explain how minute brachyodont animals gave rise to the mammoth-sized hypsodont Elasmotherium. 相似文献
This paper describes recent material gathered during the second fieldwork at Ma U'Oi in November 2002 by a Vietnamese–French–Japanese team. The Ma U'Oi cave, located in the province of Hoà Binh (60 km SW from Hanoi), northern Vietnam, belongs to a karstic network developed in Triassic dark-grey limestones.
The cave is filled with coarse-grained breccias containing numerous fossil remains, partially preserved at several loci inside the cave (wall, vault and ground). We describe new teeth which confirm the occurrence of mammal taxa already mentioned at Ma U'Oi (Bacon et al., 2004)[Bacon, A-M., Demeter, F., Schuster, M., Long, V.T., Thuy, N.K., Antoine, P-O., Sen, S., Nga, H.H., Huong, N.T.M., 2004. The Pleistocene Ma U'Oi cave, northern Vietnam: palaeontology, sedimentology and palaeoenvironments. Geobios 37, 305–314], while others, mainly microvertebrates, emphasize the occurrence of new species for the Pleistocene of Vietnam. We report here, for the first time, the occurrence of these microvertebrates of different groups (primates, rodents, insectivores, small reptiles and amphibians) in the faunal assemblage. Among mammal taxa, the presence of one more hominid affiliated to archaic Homo is also attested by our findings. U/Th dating carried out on 2 samples extracted from breccia speleothems confirms the biochronological estimate, with fossiliferous fillings ranging from late Middle Pleistocene to Late Pleistocene. 相似文献
The ovariectomized old cynomolgus monkey is a recognized model of human osteoporosis, and the same species can be used for the assessment of the efficacy and potential toxicity of agents intended to prevent or treat osteoporosis. Several assays have been developed that can measure the same biochemical markers of bone turnover as are used in human patients for the diagnosis and treatment follow-up of bone-related diseases, including osteoporosis. The aim of the present study was to describe the results obtained with these assays in normal control monkeys, their variations with age and sex, and their sensitivity in monitoring the bone turnover induced by ovariectomy in old skeletally mature cynomolgus monkeys. Seven old cynomolgus monkeys were bilaterally ovariectomized and 13 age-matched monkeys were sham-operated. Bone mineral density and biochemical markers were measured before and at regular intervals after surgery for up to 20 months. Total alkaline phosphatase (total ALP), bone-specific alkaline phosphatase isoenzyme (bone ALP) and osteocalcin (OC) were highly correlated to the decrease in bone mineral density (BMD) induced by ovariectomy. Deoxypyridinoline (DPD) measured by enzyme-linked immunoassay was insensitive to the bone resorption induced by ovariectomy, but cross-linked N-telopeptide (NTX-I) was higher in ovariectomized monkeys than in control monkeys. These results demonstrate that reliable biochemical parameters are available to adequately monitor and provide insight into osteoclastic bone resorption and osteoblastic bone formation, the two components of bone turnover in this animal model, and can thus be used to assess the efficacy and toxicity of potential therapeutic agents. 相似文献
