A 1.9-kilobase (kb) cDNA for a new transglutaminase protein has been cloned and sequenced from retinoic acid-induced human erythroleukemia (HEL) cells. Full-length cDNA analysis reveals an open reading frame coding for a polypeptide of 548 amino acid residues with a molecular weight of 61,740. The deduced amino acid sequence exhibited 98% identity to the human cellular transglutaminase sequence. The cysteine at position 277 in the active site and the putative Ca(2+)-binding pocket at residues 446-453 of cellular transglutaminase are conserved. Such evidence predicts that the encoded protein product is likely to be a transglutaminase homologue (TGase-H). Immunoprecipitation of the in vitro translation products from a synthetic TGase-H mRNA and from total protein of cultured erythroleukemia HEL cells revealed a protein with a molecular weight of 63,000 by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Northern blot analysis of HEL cells and normal human fibroblast cells WI-38 using a cellular TGase probe detected the 1.9- and 4.0-kb RNA species at a relative abundance of 1:3 and 1:7, respectively. The 3'-end of the human cellular transglutaminase mRNA was also cloned and sequenced to allow comparison to the 3'-end of TGase-H reported here. This new piece gives a full length of 4012 nucleotides (4.0 kb) for human cellular transglutaminase. Comparison of the 5'-end (bases 1-1747) of the 1.9- and 4.0-kb cDNA sequences revealed a very high degree of identity. Beginning with base 1748, the sequences diverge showing no homology. Congenital tremor, type A-IV is an inherited abnormality of British Saddleback piglets characterized by the presence of poorly myelinated axons throughout the CNS. This is reflected by a general lowering of the spinal cord lipid content to about half control values. In particular, cerebroside and plasmalogen levels are markedly reduced. Changes in the fatty acid composition of total lipid extracts and two isolated cerebroside fractions suggest that there is a metabolic defect in which fatty acid chain elongation, desaturation and hydroxylation are sub-optimal. Cholesterol esters accumulate, make up about 30 per cent of total sterols and contain more than 90 per cent of C16 and C18 fatty acids. These may have arisen through the breakdown of cerebrosides containing abnormal proportions of these acids. Slightly increased levels of acid hydrolase activity appear to confirm that abnormal or immature myelin is disposed of by enhanced macrophagic activity. Differences between this and type A-III, the sex-linked inherited form of cerebrospinal hypomyelinogenesis are discussed and comparisons made with two forms of murine leuko-dystrophy. 相似文献

Summary The effect of 16 weeks total starvation on the ultrastructure of the red and white myotomal muscles of the crucian carp (Carassius Carassius) has been investigated.
In the white fibres the amount of myofibrillar material fell from 89.6% to 70.7% of the total fibre volume whilst in the red
fibres the fall was from 72.2% to 70.3%. The sarcoplasmic reticulum appeared to have become swollen during starvation in both
fibre types. In the white fibres the terminal cisternae of some triads seem to have fused. The volume of the red fibres occupied
by mitochondria was reduced from 16.2 % to 5.9 %. The concentration of mitochondria in the white fibres was too low to detect
any quantitative changes. A marked reduction in the amount of euchromatin material was observed in most white fibre nuclei
and many red fibre nuclei.
Many of the ultrastructural changes noted in the present study can be correlated with biochemical changes known to occur in
the red and white myotomal muscles of fish during starvation.

A technique has been perfected for measuring the sedimentation coefficient of microgram quantities of a reduced protein in 6 M guanidine hydrochloride. The protein is sedimented through a gradient of 5-8 M guanidine-HCl in the presence of dithiothreitol in a SW 50.1 swinging-bucket rotor. Run conditions are calibrated by a simultaneous measurement using a single reference protein. Thus, the need for running a calibration curve involving several standard proteins simultaneously with a sample is eliminated. Because of the trace quantity of protein used, the technique yields an estimate of the sedimentation coefficient at zero concentration (s0) directly without extrapolation. Since s0 is a function of the molecular weight of a reduced protein in this solvent, the method also allows an estimate of the subunit molecular weight of the protein. A high correlation in the susceptibilities of 44 strains isolated from clinical material to a variety of antimicrobial agents was found between the minimum inhibitory concentration and conductivity measurements. Some discrepancy was found for vancomycin and teicoplanin when tested against 21 strains of staphylococci. Similarly, discrepancies were seen with azlocillin, chloramphenicol and gentamicin when tested against strains of aerobic and facultatively anaerobic Gram-negative bacilli. In the majority of cases in which discrepancies were noted, this reflected differences in relative, and not absolute, susceptibility. With five strains of Pseudomonas however, there was a poor correlation with three of four antibiotics studied. The advantage of this technique is that sensitivities are available within 4–6 h. 相似文献
