A biophysical characterization of the iron coordination environment in wheat germ peroxidase

 The heme protein wheat germ peroxidase (isoenzyme C₂) and its cyanide-inhibited form have been investigated by means of electronic, CD and paramagnetic NMR spectroscopy. The data indicate a protein environment of the active site distinct from that of horseradish peroxidase (HRP), with a larger solvent accessibility. The iron is pentacoordinated at neutral and low pH, whereas a hydroxyl anion may be bound at alkaline pH. The fifth axial ligand is a His residue with a partial anionic character, as found in other peroxidases. A spin equilibrium is observed at high enzyme concentrations. Liquid chromatographic assay in plasma of one of the members of a new series of anticonvulsants: -3-hydroxy-3-ethyl-3-phenylpropionamide

A method for the simultaneous determination of HEPP ( -3-hydroxy-3-ethyl-3-phenylpropionamide), a member of a new homologous series of phenylamide-derivative anticonvulsants, with six other antiepileptic drugs (ethosuximide, primidone, phenobarbital, phenytoin, carbamazepine and clonazepam) in plasma by high-performance liquid chromatography is described. These drugs are extracted from plasma by adding an equal volume of acetonitrile. An aliquot of the extract is then injected on a reversed-phase column with a acetonitrile-methanol-phosphate buffer mobile phase. The total time required for the whole analytical process, including the plasma pretreatment and chromatography, is approximately 30 min. The assay method is simple, rapid and reproducible, and therefore considered suitable for routine use in clinical investigations monitoring HEPP simultaneously with common antiepileptic drugs.     

Acute Toxicity of Ochratoxins A and B in Chicks

Ochratoxins A and B were given to 1-day-old Babcock B-300 cockerels to evaluate acute toxic effects. Two trials with ochratoxin A gave 7-day oral median lethal dose estimates of 116 mug (3.3 mg/kg) and 135 mug (3.9 mg/kg) per chick. Chicks given daily oral doses of 100 mug of ochratoxin A died on the second day. Single subcutaneous doses of 400 mug of ochratoxin A were also lethal. The 7-day oral median lethal dose of B was estimated at 1,890 mug (54 mg/kg) per chick. Chicks given oral doses of 100 mug of ochratoxin B daily for 10 days survived. Sublethal doses of both ochratoxins A and B resulted in growth suppression which was proportional to the amount of ochratoxin given. Visceral gout was the principal gross finding. Microscopic examinations revealed acute nephrosis, hepatic degeneration or focal necrosis, and enteritis. Suppression of hematopoiesis in the bone marrow and depletion of lymphoid elements from the spleen and bursa of Fabricius were frequently seen. Both ochratoxins appeared to have similar pathological effects. This is the first report on the toxicity of ochratoxin B.     

Intrauterine programming of ageing

Ageing is an unavoidable corollary to being alive; the most intuitive interpretation of ageing being that it is the consequence of progressive body degeneration. In agreement with this, current models propose that ageing occurs through a stepwise accumulation of DNA damage, which ultimately limits the regenerative capacity of tissues. On the other hand, there is increasing evidence that fetal distress can influence the development of disease in adult life, a phenomenon known as ‘intrauterine programming’. The extent to which an intrauterine exposure to DNA damage can compromise lifespan remains unclear. My group has recently generated a murine model of a human syndrome linked to defective DNA repair and observed that these animals age prematurely, but the accumulation of DNA damage is restricted mostly to the embryonic period. Here, I discuss the implications of this finding and propose that ageing can be influenced by fetal distress.     

Phase Microscopy in Bacteriology

The function and use of the phase microscope is described for controlling the contrast in the image and making visible unstained, living microorganisms and cytological details within them. The miscroscope may be used to examine unstained, growing cultures in Petri dishes, even with the oil immersion lens. Flagella are shown on the living spore of Ashbya. Since microorganisms show sharp edges under the phase microscope, measurement of unstained living cells is now possible. B. megatherium was found to average 1.0μ in width and B. cereus 1.1 μ in width with very small variation. Observations on locomotion are like those reported by Pijper. Stained preparations of low contrast may be seen with considerably enhanced contrast by phase microscopy.
