Summary This contribution deals with problems associated with the culture of a thermotolerant methylotrophic Bacillus sp. The results reported clearly demonstrate why conventional enrichment/isolation procedures have, in the past, failed to allow such microbes to assert themselves. The catastrophic effect of carbon substrate (methanol) exhaustion on such cultures is clearly evidenced, but the effects of other nutrient exhaustion or limitations are demonstrated to be markedly less stringent. The failure of such cultures to complete the sporulation process when growing on methanol has important consequences with respect to their survival characteristics. 相似文献

The purpose of this study was to select the promising biopreservation bacteriocin producer strain from goat milk and characterize the expressed bacteriocin, related to its physiological and biochemical properties and specificity of operon encoding production and expression of antimicrobial peptide. Brazilian goat milk was used as the source for the selection of bacteriocin-producing lactic acid bacteria. One strain (DF105Mi) stood out for its strong activity against several Listeria monocytogenes strains. Selected strain was identified based on the biochemical and physiological characteristics and 16s rRNA analysis. The bacteriocin production and inhibitory spectrum of strain DF105Mi were studied, together with the evaluation of the effect of temperature, pH, and chemicals on bacteriocin stability and production, activity, and adsorption to target cells as well as to the cell surface of bacteriocin producers. Physiological and bio-molecular analyses based on targeting of different genes, parts of nisin operon were performed in order to investigate the hypothesis that the studied strain can produce and express nisin. Based on biochemical, physiological, and 16s rRNA analysis, the strain DF105Mi was classified as Enterococcus hirae. The selected strain produces a bacteriocin which is stable in a wide range of pH (2.0–12.0), temperature (up to 120 °C), presence of selected chemicals and presents adsorption affinity to different test organisms, process influenced by environmental conditions. Higher bacteriocin production by Ent. hirae DF105Mi was recorded during stationary growth phase, but only when the strain was cultured at 37 °C. The strain’s genetic analysis indicated presence of the genes coding for the production of the bacteriocin nisin. This result was confirmed by cross-checking the sensitivity of the produced strain to commercial nisin A. The strong anti-Listeria activity, bacteriocin adsorption, and stability of produced bacteriocin indicate that Ent. hirae DF105Mi presents a differentiated potential application for biopreservation of fermented dairy products.
The present study evaluates the groundwater quality for drinking and agricultural purposes and determines physicochemical characteristics of groundwater in the Sistan and Baluchistan province in Iran. In order to investigate the water quality, sampling was done in 654 open dug wells, the chemical parameters were analyzed, and water quality index was determined. In this regard, Langelier saturation index (LSI), Ryznar Stability index (RSI), Puckorius scaling index (PSI), Larson–Skold index (LS), and Aggressiveness index (AI) were considered to determine water suitability for industrial purposes. Finally, the analytical results were taken to generate the numerical spatial distribution of the parameters using the geographic information system (GIS) environment. According to the results, water sources were less corrosive based on AI and PSI, low and light corrosion according to RSI, and corrosion according to the Larsson–Sckold index. The results of the drinking water quality index showed that 1.2% shared extraction wells were classified as excellent, 52.1% as good, 39% as poor, 6% as very poor, and 1.7% as unsuitable for drinking purpose classes. In addition, irrigating water quality index illustrated that 19.9% and 80.1% wells were placed in the “excellent” and “Good” classes, respectively. Parameters of the two-parameter logistic model are generally estimated via the expectation-maximization algorithm, which improves initial values for all parameters iteratively until convergence is reached. Effects of initial values are rarely discussed in item response theory (IRT), but initial values were recently found to affect item parameters when estimating the latent distribution with full non-parametric maximum likelihood. However, this method is rarely used in practice. Hence, the present study investigated effects of initial values on item parameter bias and on recovery of item characteristic curves in BILOG-MG 3, a widely used IRT software package. Results showed notable effects of initial values on item parameters. For tighter convergence criteria, effects of initial values decreased, but item parameter bias increased, and the recovery of the latent distribution worsened. For practical application, it is advised to use the BILOG default convergence criterion with appropriate initial values when estimating the latent distribution from data. 相似文献
