A Proteus-typing method based on proticin production and proticin susceptibility (c.f. Senior, 1977) has been modified to increase its sensitivity. Proticins were prepared in fluid medium and applied to agar-plates shortly before seeding the plates with indicator bacteria. A given 10 proticin producer strains, which are responsible for the susceptibility patterns of the indicator-bacteria (S-types), form the foundation for this typing method. Using this producer-set an indicator-set (28 strains) was selected which was suitable for the typing of strains with different proticin activities (P-types). Standardization of the temperature for proticin production proved to be necessary. The degree of similarity between proticins was further elaborated by testing all indicator strains for susceptibility to proticin titrations. In the group of 148 clinical Proteus-isolates (four species) used for the development of the typing system 28 S-types and 34 P-types were observed. By combining the S- and P-type parameters 86 S-P-types were obtained for the 4 species combined. Seven strains were not typable. A separate group of 100 clinical Proteus-isolates was tested in order to prove the usefulness of the method. 39 new S-P-types were found. Repeated isolations from the same patients yielded the same patterns. Proteus S-P-typing is a useful method for the typing of Proteux vulgaris and Proteus mirabilis, but proves inadequate for the typing of Proteus rettgeri and Proteus morganii.  相似文献

It is generally assumed that favourable weather conditions determine the activity levels of lizards, because of their temperature-dependent behavioural performance. Inactivity, however, might have a selective advantage over activity, as it could increase survival by reducing exposure to predators. Consequently, the effects of weather conditions on the activity patterns of lizards should be strongly influenced by the presence of predators. Using remote camera traps, we test the hypothesis that predator presence and weather conditions interact to modulate daily activity levels in two sedentary cordylid lizards, Karusasaurus polyzonus and Ouroborus cataphractus. While both species are closely related and have a fully overlapping distribution, the former is a fast-moving lightly armoured lizard, whereas the latter is a slow-moving heavily armoured lizard. The significant interspecific difference in antipredator morphology and consequently differential vulnerability to aerial and terrestrial predators, allowed us to unravel the effects of predation risk and weather conditions on activity levels. Our results demonstrate that K. polyzonus is predominantly active during summer, when ambient temperatures are favourable enough to permit activity. In contrast, a peak in activity during spring was observed in O. cataphractus, with individuals being inactive during most of summer. While favourable weather conditions had a strong effect on the activity levels of K. polyzonus, no such relationship was present in O. cataphractus. Contrary to our hypothesis, the presence of terrestrial predators does not seem to affect daily activity levels or alter the influence of weather conditions on activity levels. We conclude that inactivity in O. cataphractus appears to be related to seasonal differences in vulnerability to predators, rather than the presence of predators, and highlight the importance of additional selective pressures, such as food abundance, in determining the species’ activity levels.  相似文献

As uncontrolled cell proliferation requires nucleotide biosynthesis, inhibiting enzymes that mediate nucleotide biosynthesis constitutes a rational approach to the management of oncological diseases. In practice, however, results of this strategy are mixed and thus elucidation of the mechanisms by which cancer cells evade the effect of nucleotide biosynthesis restriction is urgently needed. Here we explored the notion that intrinsic differences in cancer cell cycle velocity are important in the resistance toward inhibition of inosine monophosphate dehydrogenase (IMPDH) by mycophenolic acid (MPA). In short-term experiments, MPA treatment of fast-growing cancer cells effectively elicited G0/G1 arrest and provoked apoptosis, thus inhibiting cell proliferation and colony formation. Forced expression of a mutated IMPDH2, lacking a binding site for MPA but retaining enzymatic activity, resulted in complete resistance of cancer cells to MPA. In nude mice subcutaneously engrafted with HeLa cells, MPA moderately delayed tumor formation by inhibiting cell proliferation and inducing apoptosis. Importantly, we developed a lentiviral vector–based Tet-on label-retaining system that enables to identify, isolate and functionally characterize slow-cycling or so-called label-retaining cells (LRCs) in vitro and in vivo. We surprisingly found the presence of LRCs in fast-growing tumors. LRCs were superior in colony formation, tumor initiation and resistance to MPA as compared with fast-cycling cells. Thus, the slow-cycling compartment of cancer seems predominantly responsible for resistance to MPA.  相似文献

Wolbachia are cytoplasmically inherited endosymbionts known to cause several reproductive alterations in insects which allow their spread in host populations. In the Uzifly Exorista sorbillans, endoparasites of silkworms, the prevalence of Wolbachia is high in the field. In the present study, we investigated Wolbachia’s effects on the Uzifly fitness traits by measuring fecundity and hatching rate in crosses involving infected and cured individuals. We found evidence for positive fitness effects associated with Wolbachia infection in females which could help promote the spread of Wolbachia in E. sorbillans populations. We tested two types of treatments for removing Wolbachia, antibiotic therapy and high temperature treatment and found an influence on the reproduction: females treated by antibiotics have a lower fecundity than females cured by high temperature which could indicate a negative effect of the antibiotherapy on females’ fitness. Furthermore, the monitoring of the Uzifly populations during 2 years revealed seasonal variations of the offspring production which may be linked to temperature.  相似文献

Sib‐mating avoidance is a pervasive behaviour that is expected to evolve in species subject to inbreeding depression. Although laboratory studies provide elegant demonstrations, small‐scaled bioassays minimize the costs of mate finding and choice, and thus may produce spurious findings. We therefore combined laboratory experiments with field observations to examine the existence of inbreeding avoidance using the parasitoid wasp Venturia canescens. In the laboratory, our approach consisted of mate‐choice experiments to assess kin discrimination in population cages with competitive interactions. A higher mating probability after sib rejections suggested that females could discriminate their sibs; however, in contrast to previous findings, sib‐mating avoidance was not observed. To compare our laboratory results to field data, we captured 241 individuals from two populations. Females laid eggs in the lab, and 226 daughters were obtained. All individuals were genotyped at 18 microsatellite loci, which allowed inference of the genotype of each female's mate and subsequently the relatedness within each mating pair. We found that the observed rate of sib‐mating did not differ from the probability that sibs encountered one another at random in the field, which is consistent with an absence of sib‐mating avoidance. In addition, we detected a weak but significant male‐biased dispersal, which could reduce encounters between sibs. We also found weak fitness costs associated with sib‐mating. As such, the sex‐biased dispersal that we found is probably sufficient to mitigate these costs. These results imply that kin discrimination has probably evolved for purposes other than mate choice, such as superparasitism avoidance.  相似文献

Background  

Homologous recombination mediated by the λ-Red genes is a common method for making chromosomal modifications in Escherichia coli. Several protocols have been developed that differ in the mechanisms by which DNA, carrying regions homologous to the chromosome, are delivered into the cell. A common technique is to electroporate linear DNA fragments into cells. Alternatively, DNA fragments are generated in vivo by digestion of a donor plasmid with a nuclease that does not cleave the host genome. In both cases the λ-Red gene products recombine homologous regions carried on the linear DNA fragments with the chromosome. We have successfully used both techniques to generate chromosomal mutations in E. coli K-12 strains. However, we have had limited success with these λ-Red based recombination techniques in pathogenic E. coli strains, which has led us to develop an enhanced protocol for recombineering in such strains.  相似文献

Triple-negative breast cancer (TNBC) represents an aggressive subtype, for which radiation and chemotherapy are the only options. Here we describe the identification of disulfiram, an FDA-approved drug used to treat alcoholism, as well as the related compound thiram, as the most potent growth inhibitors following high-throughput screens of 3185 compounds against multiple TNBC cell lines. The average IC50 for disulfiram was ~300 nM. Drug affinity responsive target stability (DARTS) analysis identified IQ motif-containing factors IQGAP1 and MYH9 as direct binding targets of disulfiram. Indeed, knockdown of these factors reduced, though did not completely abolish, cell growth. Combination treatment with 4 different drugs commonly used to treat TNBC revealed that disulfiram synergizes most effectively with doxorubicin to inhibit cell growth of TNBC cells. Disulfiram and doxorubicin cooperated to induce cell death as well as cellular senescence, and targeted the ESA+/CD24-/low/CD44+ cancer stem cell population. Our results suggest that disulfiram may be repurposed to treat TNBC in combination with doxorubicin.  相似文献
