The effects of treatment with dopamine agonists and 6-hydroxydopamine on the release of opioid peptides from the myenteric plexus of guinea-pig ileum were examined. Apomorphine or bromocriptine treatment at doses that act on dopamine autoreceptors to inhibit dopamine release resulted in a significant elevation of the release of opioid peptides. 6-hydroxydopamine treatment, which produces a lesion of catecholaminergic nerve terminals also resulted in an increase in opioid peptide release. These findings indicate that interruption of dopaminergic transmission in the myenteric plexus produces an increase in the release of opioid peptides and suggest an inhibitory modulation of opioid peptidergic neurons by dopamine systems in the myenteric plexus of the guinea-pig ileum.  相似文献

In this article, we investigate the potential for detection and characterization of sinkholes under dense forest cover by using airborne laser scanning data. Laser pulse returns from the ground provide important data for the estimation of digital elevation model (DEM), which can be used for further processing. The main objectives of this study were to map and determine the geomorphometric characteristics of a large number of sinkholes and to investigate the correlations between geomorphology and vegetation in areas with such characteristics. The selected study area has very low anthropogenic influences and is particularly suitable for studying undisturbed karst sinkholes. The information extracted from this study regarding the shapes and depths of sinkholes show significant directionality for both orientation of sinkholes and their distribution over the area. Furthermore, significant differences in vegetation diversity and composition occur inside and outside the sinkholes, which indicates their presence has important ecological impacts.  相似文献

The well-known argument against the Search for ExtraTerrestrial Intelligence (SETI) due to George Gaylord Simpson is re-analyzed almost half a century later, in the light of our improved understanding of preconditions for the emergence of life and intelligence brought about by the ongoing “astrobiological revolution”. Simpson’s argument has been enormously influential, in particular in biological circles, and it arguably fueled the most serious opposition to SETI programmes and their funding. I argue that both proponents and opponents of Simpson’s argument have occasionally mispresented its core content. Proponents often oversimplify it as just another consequence of biological contingency, thus leaving their position open to general arguments limiting the scope of contingency in evolution (such as the recent argument of Geerat Vermeij based on selection effects in the fossil record). They also tend to neglect that the argument has been presented as essentially atemporal, while referring to entities and processes that are likely to change over time; this has become even less justifiable as our astrobiological knowledge increased in recent years. Opponents have failed to see that the weaknesses in Simpson’s position could be removed by restructuring of the argument; I suggest one way of such restructuring, envisioned long ago in the fictional context by Stanislaw Lem. While no firm consensus has emerged on the validity of Simpson’s argument so far, I suggest that, contrary to the original motivation, today it is less an anti-SETI argument, and more an astrobiological research programme. In this research programme, SETI could be generalized into a platform for testing some of the deepest assumptions about evolutionary continuity and the relative role of contingency versus convergence on unprecedented spatial and temporal scales.  相似文献

Purse-web spiders Atypus spp. (Araneae: Atypidae) are the only representatives of the archaic spider infraorder Mygalomorphae in the Central European fauna. These burrow-dwelling spiders are red-listed in all Central European countries. Atypus muralis occurs on relics of continental steppes, Atypus piceus on dry submediterranean grassland and dry herb-rich forest fringes, while Atypus affinis inhabits sparse dwarf dry forests, usually those dominated by oaks. Long-term sustainability of their habitats in the relatively humid Central European climate was maintained by grazing. Since the 1950s, pasturing was terminated across most of Central Europe, which led to immediate overgrowth of pastures by aggressive grasses and woody plants. Grazing was later reinstated in some reserves, but in cases of poorly controlled grazing intensity, footsteps had immediate detrimental effects on the residual populations of purse-web spiders and other invertebrates. Therefore, grazing intensity needs to be regulated, considering its double-edge effects on the biota of xerothermophilic habitats. Nests of European Atypus species have never been found in habitats subject to recent or even regular distribution. Therefore, they may serve as reliable indicators of well preserved xerothermic habitats. Their nests are easy to detect, and the European Atypus spiders can be utilized as model species for monitoring impacts of conservation management. Monitoring population dynamics of Atypus requires establishing permanent study plots of uniform size, documentation of vegetation by phytocenological relevé, counting their density (number of purse-webs found in the plot) and age structure (diameter of purse webs).  相似文献

Ischemic acute renal failure (ARF) is a highly complex disorder involving renal vasoconstriction, filtration failure, tubular obstruction, tubular backleak and generation of reactive oxygen species. Due to this complexity, the aim of our study was to explore effects of Angiotensin II type 1 receptor (AT1R) blockade on kidney structure and function, as well as oxidative stress in spontaneously hypertensive rats (SHR) after renal ischemia reperfusion injury. Experiments were performed on anaesthetized adult male SHR in the model of ARF with 40 minutes clamping the left renal artery. The right kidney was removed and 40 minutes renal ischemia was performed. Experimental groups received AT1R antagonist (Losartan) or vehicle (saline) in the femoral vein 5 minutes before, during and 175 minutes after the period of ischemia. Biochemical parameters were measured and kidney specimens were collected 24h after reperfusion. ARF significantly decreased creatinine and urea clearance, increased LDL and lipid peroxidation in plasma. Treatment with losartan induced a significant increase of creatinine and urea clearance, as well as HDL. Lipid peroxidation in plasma was decreased and catalase enzyme activity in erythrocytes was increased after losartan treatment. Losartan reduced cortico-medullary necrosis and tubular dilatation in the kidney. High expression of pro-apoptotic Bax protein in the injured kidney was downregulated after losartan treatment. Our results reveal that angiotensin II (via AT1R) mediates the most postischemic injuries in hypertensive kidney through oxidative stress enhancement. Therefore, blockade of AT1R may have beneficial effects in hypertensive patients who have developed ARF.  相似文献

Zusammenfassung Die Protoplasten der obligat aeroben Hefe Rhodotorula gracilis wurden hinsichtlich ihrer charakteristischen physiologischen und Transporteigenschaften mit intakten Zellen verglichen. Folgende Ergebnisse wurden gewonnen: 1. Endogene und durch d-Glucose stimulierte Atmung entsprach den Werten von intakten Hefezellen. 2. d-Glucose wurde von Protoplasten aus dem Medium aufgenommen und abgebaut. 3. Die Aufnahme von d-Xylose führte zu vielfacher Akkumulation der Pentose im Zellinnern. Nach 50 min wurde ein für den Xyloseabbau induziertes System wirksam. 4. Bei Zugabe im Gemisch wurde die Aufnahme von d-Xylose durch d-Glucose unterbunden. 5. Akkumulierte d-Xylose wurde bei Zugabe von d-Glucose im Austauschtransport durch den mobilen Träger aus der Zelle heraus befördert. 6. Der Zuckertransport, gemessen an der d-Xyloseaufnahme, war streng stoffwechselenergieabhängig und wurde durch Entkoppler vollständig gehemmt.Diese Ergebnisse zeigen, daß die Stoffwechsel- und Transportfunktionen der intakten Hefezellen in ihren Protoplasten vollstädig erhalten bleiben. Die Anwendung von R. gracilis-Protoplasten zur Klärung spezieller Fragestellungen ergab: 1. Der Transport von d-Trehalose erfolgte nach extracellulärer Spaltung des Disaccharides durch Aufnahme der entstandenen Glucose. 2. Densitometrische Messungen an Protoplastensuspensionen zeigten sich geeignet zur kontinuierlichen Aufzeichnung von Zuckeraufnahmevorgängen.
Protoplasts from the yeast Rhodotorula gracilis II. Physiological and transport properties
The protoplasts of the obligatory aerobic yeast Rhodotorula gracilis (5/Fres/Harrison) were compared with the intact yeast cells with respect to the identity of their physiological and transport properties. It was found: 1. The rates of endogenous and glucose-stimulated respiration of protoplasts were similar to those of the whole cells. 2. d-glucose was taken up from the medium with constant velocity; no free glucose could be detected inside the protoplasts. 3. The uptake of d-xylose led to manifold accumulation of the pentose intracellularly. Within 50 min incubation an enzyme system for the degradation of d-xylose became effective. 4. In a mixture of d-xylose and d-glucose the latter blocked the uptake of the pentose. 5. d-xylose once accumulated was exchanged by the mobile membrane carrier for d-glucose after its addition to the protoplast suspension. 6. Addition of NaN3 or CCCP resulted in an inhibition of d-xylose uptake. The transport process is tightly coupled to cell metabolism.It is concluded that the metabolic and transport functions of R. gracilis protoplasts equal those of the intact yeast cells. The application of the protoplasts to study some special transport problems revealed: 1. In the course of d-trehalose uptake the disaccharide was cleaved to glucose, which was actually transported across the cell membrane. 2. Densitometry of protoplasts suspensions was found suitable for the continuous recording of sugar uptake processes. This observation is of special importance for further investigations of the oscillations in sugar transport observed earlier (Heller and Höfer, 1973).
