This article considers the objection to abortion that a woman who voluntarily engages in sexual activity is responsible for her fetus and so cannot have an abortion. The conclusion argued for is that the conceptions of responsibility that can ground the objection that are considered do not necessitate a requirement on the part of a pregnant woman to carry her pregnancy to term. Thus, the iterations of the responsibility objection presented cannot be used to curtail reproductive choice.  相似文献

The two enantiomers of 12-hydroxy-(5,8,10,14)-eicosatetraenoic acid (12-HETE) are products of different biosynthetic pathways and have distinct biologic actions. Conventional methods of stereochemical analysis of 12-HETE require multimicrogram amounts of material and cannot be applied to systems where the availability of tissue is limited and only trace quantities of 12-HETE are generated. We have developed a method capable of measuring subnanogram amounts of 12-HETE enantiomers which involves addition of racemic. 18O2-labeled 12-HETE as an internal standard, chiral phase HPLC of the pentafluorobenzyl ester derivative of 12-HETE, and stable isotope dilution gas chromatographic-negative ion chemical ionization mass spectrometric quantitation of the resolved stereoisomers. This method has been employed to determine the stereochemical composition of 12-HETE produced by isolated pancreatic islets.  相似文献

Summary A matrix program to predict short term genetic gain from single trait selection for milk yield was developed. Rate of genetic gain was calculated as the annual change in the mean breeding value of all producing females. Several parameters sets representing various selection policies were used to examine situations pertinent to dairy populations of the United States. Approach to the asymptotic rates of genetic gain within the model varied with the choice of parameters, but even with consistent selection policies, predicted total genetic gain in the first 10 years was only half of the expected from classical theory. Considerable year to year variation in the rate of gain occurred. Early gains were more dependent on female selection decisions than gains during the steady state. In a two-phase model, the approach to the linear rate of gain in the second phase was accelerated by starting with an ongoing improvement program, but considerable delays still existed. Selection for sex- limited traits such as milk yield, which require pedigree selection and a waiting time for progeny test results reached asymptotic rates more slowly than previously assumed.  相似文献

We measured the electrophoretic mobility of multilamellar phospholipid vesicles, the 31P NMR spectra of both sonicated and multilamellar vesicles, and the conductance of planar bilayer membranes to study the binding of spermine and gentamicin to membranes. Spermine and gentamicin do not bind significantly to the zwitterionic lipid phosphatidylcholine. We measured the concentrations of gentamicin and spermine that reverse the charge on vesicles formed from a mixture of phosphatidylcholine and either phosphatidylserine or phosphatidylinositol. From these measurements, we determined that the intrinsic association constants of the cations with these negative lipids are all about 10 M-1. This value is orders of magnitude lower than the apparent binding constants reported in the literature by other groups because the negative electrostatic surface potential of the membranes and the resultant accumulation of these cations in the aqueous diffuse double layer adjacent to the membranes have not been explicitly considered in previous studies. Our main conclusion is that the Gouy-Chapman-Stern theory of the aqueous diffuse double layer can describe surprisingly well the interaction of gentamicin and spermine with bilayer membranes formed in a 0.1 M NaCl solution if the negative phospholipids constitute less than 50% of the membrane. Thus, the theory should be useful for describing the interactions of these cations with the bilayer component of biological membranes, which typically contain less than 50% negative lipids. For example, our results support the suggestion of Sastrasinh et al. [Sastrasinh, M., Krauss, T. C., Weinberg, J. M., & Humes, H. D. (1982) J. Pharmacol. Exp. Ther. 222, 350-358] that phosphatidylinositol is the major binding site for gentamicin in renal brush border membranes.  相似文献

A peptide corresponding to residues 26–41 of α-bungarotoxin, and closed by a disulfide bond between two cysteine residues at the amino and C terminal ends of the peptide, was synthesized and the monomeric form was purified. The peptide, which represents the exposed part of the long central loop of the toxin molecule, was examined for binding to acetylcholine receptor. The peptide was shown by radiometric titrations to bind radiolabeled receptor, and radiolabeled peptide was bound by receptor. The specificity of the binding was confirmed by inhibition with the parent toxin. A synthetic analog of the peptide in which Trp-28 was replaced by glycine had very little (10%) of the original activity. Succinylation of the amino groups of the peptide resulted in virtually complete (98%) loss of the binding activity. These results indicate that a shortened loop peptide corresponding to the region 26–41 of α-bungarotoxin exhibits binding activities mimicking those of the parent molecule. In this region, Trp-28, and one or both of Lys-26 and Lys-38, are essential contact residues in the binding to receptor.  相似文献

Employing genotypes of day-neutral tobacco that exhibit a wide range in the number of nodes produced, it has been established that node number, and plant size, in tobacco is regulated, in large part, by two endogenous signals and one developmental state, competence. All genotypes have the same level of a root signal that maintains a vegetative pattern during early growth. The number of nodes produced before the formation of the terminal flower, as well as plant size, is a function of the strength of the floral stimulus from the leaves and the competence of the terminal meristem to respond to the floral stimulus by initiating the terminal flower.  相似文献

Studies were undertaken to determine whether the effect of alloxan to inactivate a membrane-bound calcium- and calmodulin-dependent protein kinase was specific for the pancreatic islets and whether inactivation of the kinase occurred also after injection of a diabetogenic dose of alloxan into rats. The effect of alloxan was also examined on similar particulate calcium- and calmodulin-dependent kinases present in two other secretory tissues, mammary acini and forebrain. Exposure of alloxan to cell-free preparations of all secretory tissues examined inhibited the calcium- and calmodulin-dependent kinase activities, suggesting that the specificity of alloxan action was not due to the presence in islets of a kinase uniquely sensitive to alloxan. To determine whether the selective effect of alloxan action was mediated at the cellular level, experiments were performed with alloxan presented to intact cells. Whereas alloxan exposure to viable cell preparations of islets and brain decreased the subsequently measured calcium- and calmodulin-dependent protein kinase activity, the activity measured in mammary acini exposed to these alloxan concentrations was unaffected. Injection (i.v.) of a diabetogenic dose of alloxan (50 mg/kg) produced an immediate (10 min) and selective inactivation of the calcium- and calmodulin-dependent protein kinase in pancreatic islests but had no effect on the similar kinases measured in brain and mammary acini. These results indicate that the unique sensitivity of islets to alloxan may result from the ability of alloxan to rapidly gain intracellular access and then inactivate this kinase activity. The selective effect of alloxan injection on this islet protein kinase is consistent with the hypothesis that inactivation of the kinase by alloxan is related to its diabetogenic effect in vivo.  相似文献
