Casein kinase 1 (CK1) is a pleiotropic protein kinase implicated in several fundamental processes of eukaryotic cell biology. Plasmodium falciparum encodes a single CK1 isoform, PfCK1, that is expressed at all stages of the parasite’s life cycle. We have previously shown that the pfck1 gene cannot be disrupted, but that the locus can be modified if no loss-of-function is incurred, suggesting an important role for this kinase in intra-erythrocytic asexual proliferation. Here, we report on the use of parasite lines expressing GFP- or His-tagged PfCK1 from the endogenous locus to investigate (i) the dynamics of PfCK1 localisation during the asexual cycle in red blood cells, and (ii) potential interactors of PfCK1, so as to gain insight into the involvement of the enzyme in specific cellular processes. Immunofluorescence analysis reveals a dynamic localisation of PfCK1, with evidence for a pool of the enzyme being directed to the membrane of the host erythrocyte in the early stages of infection, followed by a predominantly intra-parasite localisation in trophozoites and schizonts and association with micronemes in merozoites. Furthermore, we present strong evidence that a pool of enzymatically active PfCK1 is secreted into the culture supernatant, demonstrating that PfCK1 is an ectokinase. Our interactome experiments and ensuing kinase assays using recombinant PfCK1 to phosphorylate putative interactors in vitro suggest an involvement of PfCK1 in many cellular processes such as mRNA splicing, protein trafficking, ribosomal, and host cell invasion. 相似文献

In this study, we re-describe the advertisement and territorial calls of Boana pardalis, carry out an acoustic comparison between the studied species and the other congeners of the B. faber group, and report for the first time the tympanic amplexus for the studied species. We investigated the phylogeography of the salt water rotifer Brachionus plicatilis, a cyclical parthenogen with passive dispersal mechanisms, using resting eggs recovered from saline lake sediments. Individual resting eggs were obtained from a large selection of lakes which were representative of five endorheic basins and the chain of coastal ponds in the Iberian Peninsula. The novel use of resting eggs allows the integration of seasonal and annual variations as well as the impact of stochastic effects such as drift and local extinction. A 653 bp fragment of the mitochondrial cytochrome oxidase subunit I (COI) gene was sequenced from 98 eggs. Our results revealed a deep phylogeographical structure in this species, with a division into two main lineages with distinct geographical distributions, which probably diverged at the beginning of the Pleistocene period. Most of the mitochondrial DNA haplotypes were restricted to single lakes. Nested clade analysis supported Early Pleistocene fragmentation of populations, low gene flow and some long-distance colonization. These conclusions contrast strongly with previous ideas on rotifer biogeography and this pattern is consistent with a recolonization of the Iberian Peninsula from two glacial refugia. The results provide new insights into the processes responsible for the genetic diversification of passive dispersers, a life-history trait typical of zooplanktonic biotas. 相似文献

Between December 1971 and December 1977, 41 patients with long-standing boutonnière deformities were treated by Matev's operation. The average follow-up time was 13 months (range 8 to 18 months). The duration of their lesions varied from 32 to 176 days. The digits most affected were the long finger (14) and the ring finger (11). Postoperatively, all patients showed excellent flexion of the fingers with only a very slight deficit in extension of the PIP joint. Eight of them also had some deficit in extension of the DIP joint, with flexion there of 10 degrees to 15 degrees, but little real disability. The results indicate that when the Matev operation is clearly indicated and properly performed, it is an excellent one for the treatment of this condition. 相似文献

In this work we report biochemical ex vivo studies with a vanadium compound containing a pyridinone ligand, the bis(1,2-dimethyl-3-hydroxy-4-pyridinonate)oxovanadium (IV), VIVO(dmpp)2, which has shown to have promising antidiabetic activity. The experiments were carried out on primary adipocytes of 6-8 week old Wistar rats. Insulin-stimulated glucose uptake studies were performed using a radioactive assay by measuring the (U)-14C-glucose taken up by the isolated adipocytes for 30 min. Adipocytes were incubated with and without insulin and in the presence and absence of different concentrations of VIVO(dmpp)2 (100-500 μM) for 45 min. We observed that in a nontoxic concentration, as demonstrated by the Alamar Blue test, VIVO(dmpp)2 significantly increases glucose uptake, in the absence of insulin, by 5-folds higher than basal, and it has a significant inhibitory effect of 78% on free fatty acid release in isolated adipocytes from normal rats. We also demonstrated that it promotes the phosphorylation of Akt1, a key protein in the insulin signaling cascade. These results were compared with those obtained with another vanadium compound reported in the literature, with a similar structure, the bis(maltolato)oxovanadium (IV) (BMOV), which is now in clinical trials. Our ex vivo results clearly indicate that VIVO(dmpp)2 is a good candidate to be a promising drug for the treatment of diabetes and other metabolic disorders. 相似文献

In a study of 450 Amazonian anurans, we isolated yeasts and yeast-like fungi from 54 animals (Bufo granulosus, B. marinus, Dendrophrynyscus sp., Hyla geographica, H. lanciformes, Ololygon rubra, Adenomera hylaedactyla, Eleutherodactylus fenestratus, Leptodactylus fuscus, L. ocellatus, L. pentadactylus). The internal organs of these animals did not show any macroscopic anomaly nor histopathology. We recovered 105 fungal isolates from the anuran liver, lung, kidney, spleen, heart and gonad. The isolates were made up of 30 fungal species, 9 of which (48 isolates, 46%) were fungi with known pathogenic potentials, namely: Candida guilliermondii, C. parapsilosis, C. tropicalis, C. glabrata, Geotrichum candidum, Aureobasidium pullulans, Wangiella dermatitidis, Trichosporon cutaneum and Exophiala werneckii. Eleven animals harbored identical fungi in more than one of their internal organs; seven animals had more than one fungal species colonizing a single organ. Our findings indicated probable natural subclinical infections of candidiasis, geotrichosis or phaeohyphomycosis, and also symbiotic presence of non-pathogenic fungi among neotropical anurans. 相似文献
