A prokaryotic biotin acceptor domain was fused to the carboxy terminal end of the Chlorella hexose—proton sym- porter. The plant symporter is biotinylated in vivo when expressed in Schizosaccharomyces pombe. The extended biotinylated transport protein is fully active, catalyzes accumulation of d -glucose analogs and restores growth of a glucose-uptake-deficient yeast strain. Crude membranes were solubilized with octyl-β-d -glucoside in the presence of Escherichia colil -α-phosphatidylethanolamine. Biotinylated symporter was purified to homogeneity by biotinavidin affinity chromatography. The symporter protein was reconstituted together with cytochrome-c oxidase prepared from beef heart mitochondria into proteo-liposomes. Cytochrome-c oxidase is a redox-driven H+-pump generating a proton motive force (inside negative and alkaline) while transferring electrons from cytochrome-c to oxygen; this energy is used by the symporter to accumulate d -glucose at least 30-fold. In the absence of the driving force the transport protein facilitates diffusion of d -glucose until the concentration equilibrium is reached. It was shown that maximal transport activity depends highly on the amount of co-reconstituted cytochrome-c oxidase and that the symporter possesses 10% of its in vivo turnover number under optimized in vitro transport conditions. 相似文献

Aquatic macrophytes are one of the biological quality elements in the Water Framework Directive (WFD) for which status assessments
must be defined. We tested two methods to classify macrophyte species and their response to eutrophication pressure: one based
on percentiles of occurrence along a phosphorous gradient and another based on trophic ranking of species using Canonical
Correspondence Analyses in the ranking procedure. The methods were tested at Europe-wide, regional and national scale as well
as by alkalinity category, using 1,147 lakes from 12 European states. The grouping of species as sensitive, tolerant or indifferent
to eutrophication was evaluated for some taxa, such as the sensitive Chara spp. and the large isoetids, by analysing the (non-linear) response curve along a phosphorous gradient. These thresholds
revealed in these response curves can be used to set boundaries among different ecological status classes. In total 48 taxa
out of 114 taxa were classified identically regardless of dataset or classification method. These taxa can be considered the
most consistent and reliable indicators of sensitivity or tolerance to eutrophication at European scale. Although the general
response of well known indicator species seems to hold, there are many species that were evaluated differently according to
the database selection and classification methods. This hampers a Europe-wide comparison of classified species lists as used

Is it better to design a classifier and estimate its error on the full sample or to design a classifier on a training subset and estimate its error on the holdout test subset? Full-sample design provides the better classifier; nevertheless, one might choose holdout with the hope of better error estimation. A conservative criterion to decide the best course is to aim at a classifier whose error is less than a given bound. Then the choice between full-sample and holdout designs depends on which possesses the smaller expected bound. Using this criterion, we examine the choice between holdout and several full-sample error estimators using covariance models and a patient-data model. Full-sample design consistently outperforms holdout design. The relation between the two designs is revealed via a decomposition of the expected bound into the sum of the expected true error and the expected conditional standard deviation of the true error. 相似文献

Incubation of human serum or high density lipoprotein (HDL) at 37 degrees C in the presence of Fe2+, Fe2+/Fe3+, or Mn2+ results in the increased immunoreactivity (up to 12-, 40-, and 80-fold, respectively) of specific apoA-I epitopes identified as 3D4 and 6B8, while Mg2+, Ca2+, or Cu2+ have minimal or nonsignificant effects. The effect of Mn2+ on the 3D4 epitope requires a specific association with lipids since it can be observed with HDL but not with apoHDL, even in the presence of other lipoproteins. The increase in immunoreactivity noted with Fe2+/Fe3+ or Mn2+ can be blocked with either EDTA or antioxidants (GSH and ascorbic acid), suggesting that it takes place during a peroxidative reaction of the lipids. The peroxidation of lipids which accompanies the increase in immunoreactivity does cross-link apoA-I both with itself and with apoA-II but does not cleave the molecule. The apoA-I-containing lipoproteins which float between 1.18 and 1.22 g/ml and have a pre B-electrophoretic migration are characterized by a very low immunoreactivity with monoclonal antibody 3D4 but are 10-fold or more responsive to Mn2+ treatment than other lipoprotein subfractions, thus demonstrating heterogeneity under oxidative conditions. Proteoliposomes containing apoA-I, cholesterol, and dilinoleyl-lecithin are sensitive to Mn2+ treatment, but not those made with dioleyl- or dimyristoyl-lecithins. However, the increase in 3D4 immunoreactivity is weak and transient and is followed by the disappearance of the epitope caused by cross-linking. We measured the amount of apoA-I in serum by isotope dilution, finding 1.33 mg/ml (standard deviation 0.177) in six normolipidemic, healthy subjects. We developed this method by adapting published techniques to purify apoA-I from 3 ml of serum in two steps: density gradient ultracentrifugation and high performance liquid chromatography gel filtration. The 125I-labeled apoA-I tracer was first screened, by incubation with serum, to select labeled apoA-I which retained the ability to exchange with native apoA-I and bind to HDL. A known amount of 125I-labeled apoA-I-labeled HDL was added to unknown serum samples; apoA-I was reisolated from the serum and its specific radioactivity was used to calculate the dilution of the added, labeled apoA-I by the unlabeled apoA-I in the unknown serum. By not relying on immunochemical techniques, the isotope dilution assay provided results that are independent of the expression of individual apoA-I antigenic sites. Therefore, sera that have been assayed by isotope dilution can serve as standards to evaluate the accuracy of immunoassays for serum apoA-I and provide primary standards for such immunoassays. 相似文献

Angola is one of the most neglected African countries in terms of botanical research, in respect of both native and naturalized species. We conducted a rapid assessment of invasive plant species in western Angola during August 2014. In thirteen primary vegetation types, we recorded populations of 44 naturalized plant species, nineteen of which are conclusively invasive (spreading far from introduction sites). Dense invasive populations of Chromolaena odorata, Inga vera and Opuntia stricta pose the greatest environmental and economic threats. Some species with known taxonomic and/or biogeographic uncertainties (e.g. Chromolaena odorata and Ageratina adenophora) or which lacked key characteristics for identification such as flowers during our survey (e.g. Eucalyptus spp.) were subjected to DNA barcoding for comparisons with available genetic data from other studies. This approach allowed us to confirm the identity of taxonomically challenging taxa such as Inga vera, Opuntia stricta and Prosopis chilensis, to conclusively differentiate Chromolaena odorata from Ageratina adenophora, and identify the subspecific identity of Acacia saligna. Canonical correspondence analysis was used to assess the presence and abundance of invasive plant species with respect to the major abiotic factors and vegetation types. Three fairly distinct groups of species emerge from this analysis: (i) species of dry lowland habitats (Calotropis gigantea, Leucaena leucocephala and Opuntia stricta); (ii) species of relatively wet habitats at mid elevations (Ageratum conyzoides, Bidens pilosa, Cardiospermum grandiflorum, Chromolaena odorata, Solanum mauritianum and Tithonia diversifolia); and (iii) upland species (Ageratina adenophora, Galinsoga parviflora and Tagetes minuta). Several invasive species that are widespread in other tropical and subtropical African countries are currently either missing (e.g. many Australian Acacia species, Azolla filiculoides, Broussonetia papyrifera, Clidemia hirta, Parthenium hysterophorus, Rubus rosaefolius, Salvinia molesta), have only very localized populations in Angola (e.g. Lantana camara, Prosopis chilensis) or exist only as planted individuals (e.g. Acacia mearnsii and A. saligna subsp. saligna). 相似文献

The long-term role of photorespiration was investigated by comparing growth, development, gas exchange characteristics and mineral nutrition of a wheat crop ( Triticum aestivum L. cv. Courtot) cultivated in a culture chamber during a life cycle, either in 4% O2 or in normal O2 Low O2 pressure reduced photorespiration, but CO2 was controlled so that net photosynthesis remained the same as in the control crop. The growth and development of the low O2 crop was slowed down. Ear appearance was 16 days late, but the rate of tillering was the same as in the control and was maintained longer so that the final number of tillers was doubled. Pigment, ribulose bisphosphate carboxylase (EC 4.1.1.39) and soluble sugar contents were similar. The response of photosynthesis to CO2 and O2 was not appreciably changed by the low O2 treatment. There was almost no seed formation, and the senescence of the leaves was delayed. It appears that in non-stress conditions most of the photorespiration can be suppressed without damage to the photosynthetic apparatus.
