In the primitive eukaryotic parasite, Trypanosoma brucei, most of the enzymes of glycolysis are located within microbody organelles called glycosomes. Proteins destined for the glycosome are synthesized on free ribosomes and post-translationally translocated into the organelle. The gene, gPGK, encoding the glycosomal isozyme of phosphoglycerate kinase (gPGK), was cloned adjacent to a T7 promoter and cotransformed with a plasmid encoding T7 RNA polymerase into Escherichia coli Pgk-cells. Functional complementation occurred, but only after the creation of a ribosome-binding site by mutagenesis. This represents the first example of complementation of an E. coli mutant with a gene encoding a microbody protein. Enzymatically active recombinant gPGK was purified to near homogeneity by ion exchange chromatography from highly expressing E. coli. The recombinant protein will aid in studies of glycosomal biogenesis.  相似文献

Further characterization of the free alpha subunit immunoreactive material, not combined with beta subunit in extracts of bovine pituitaries, shows that the only significant modifications, relative to alpha subunits themselves, are the oligosaccharide O-linked to threonine-43, and heterogeneity of the carboxyl terminus. Removal of the O-linked carbohydrate with a mixture of glycosidases from Streptococcus pneumoniae results in an alpha-like material capable of combining with lutropin beta subunit and, thus, the presence of the oligosaccharide is responsible for the inability of the free alpha-like material to combine with beta subunits. Amino acid compositions of tryptic peptides spanning the entire sequence indicate no change in amino acid sequence of the free alpha-like material as compared to lutropin alpha. Further, based on the similar behavior reverse phase high performance liquid chromatography of the tryptic peptides as compared to their lutropin alpha counterparts, it is concluded that no additional post-translational modifications are present. The N-linked oligosaccharides of the free alpha-like material most likely contain terminal O-sulfated N-acetylhexosamines (as do the asparagine-linked carbohydrates from the pituitary hormones) as indicated by the presence of 3 mol of sulfate/mol of free alpha-like material and the resistance of these oligosaccharides to enzymatic deglycosylation. The O-linked oligosaccharide does not contain sulfated residues.  相似文献

Human mononuclear phagocyte (M phi) populations were compared to adult human endothelial cells (HEC) for their respective abilities to influence the proliferative responses of purified human T lymphocytes to the mitogenic agents Na-m-periodate (IO-4), soybean agglutinin (SBA), or allogeneic cells. HEC and M phi were both capable of inducing proliferative responses of allogeneic T lymphocytes in mixed-lymphocyte culture. Under low cell density culture conditions, purified T-lymphocyte proliferative responses to IO-4 or SBA could be restored by addition of syngeneic M phi or HEC. At higher cell density culture conditions, proliferation of T cells to IO-4 could be amplified more by HEC than M phi. T-lymphocyte proliferative responses to SBA were amplified by addition of HEC but were suppressed by addition of M phi. These findings indicate that human adult HEC are unique and potent accessory cells for T lymphocytes. Furthermore, these findings demonstrate that accessory cell functions of HEC can be discriminated from those of M phi.  相似文献

The technique of limited necropsy by histological examination of needle tissue cores obtained percutaneously is indicated when a full necropsy is not justified owing to the risk of infection or when tissue for special investigations is needed soon after death without recourse to full necropsy facilities. The method is ideal for detecting conditions producing diffuse changes in an organ. Because the cadaver is preserved essentially intact relatives who refuse permission for a standard necropsy might consent to a needle necropsy if this option is offered to them.  相似文献

Summary Adult tolerance of ethanol vapour in a closed system containing 12% ethanol in solution, decreases in a cline from southern to northern Australia. However a Darwin population is more tolerant than predicted from its latitude. Ethanol tolerance races in Australia have almost certainly evolved within the last 100–150 years, because of resource unavailability prior to that time. Within populations, variation among isofemale strains is lowest in the climatically extreme southern Melbourne (37°S) and northern Darwin and Melville I. (11–12°S) populations. This suggests low resource diversity within extreme populations compared with the climatically less extreme Brisbane (28°S) and especially Townsville (19°S) populations. For desiccation resistance, the population rankings are: Darwin Melbourne > Townsville > Brisbane Melville I. and for development time, rankings are similar: Darwin Melbourne < Townsville < Brisbane Melville I.Therefore resource utilization heterogeneity is greatest in populations not greatly stressed by desiccation and where development times are extended. In total therefore, the utilization of a diversity of resources is a feature of populations tending somewhat towards a K-strategy; this is emphasized by the relative heterogeneity among isofemale strains of these populations for desiccation resistance and to a lesser extent development times.The D. melanogaster gene pool can be viewed as made up of climate-associated races. Since the ethanol tolerances of adjacent (and climatically similar) Darwin and Melville I. are very different, resource utilization races may occur within climatic races. Such a mosaic of resource utilization races are more likely in climatically extreme than in optimal habitats.  相似文献
