A simple and sensitive high-performance liquid chromatography method with electrochemical detector is described for the determination of free 3-methoxy-4-hydroxyphenylacetic acid (HVA) in human plasma. The method does not involve any extraction, is specific and reproducible, and has the potential to measure serotonin (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) simultaneously. The plasma concentration of free HVA in eight normal, healthy adult volunteers was 10.9 ± 4.6 (mean ± SD). In a preliminary study, in one schizophrenic patient the plasma HVA increased twofold after neuroleptic treatment.  相似文献

Bei Rückführung in Meerwasser wird die Assimilationsrate ebenso schnell wieder erhöht, die Ausgangswerte werden allerdings innerhalb der Versuchszeit — einige 20-min-Perioden — meist nicht in voller Höhe wieder erreicht.Folgt der Wechsel zwischen Meer- und Süßwasser im Wechsel von 20 min, so reagieren die Versuchspflanzen mit der gleichen Präzision wie auf das Aus- und Einschalten einer Lichtquelle.Wird anPosidonia die photosynthetische Rate in verschiedenen Stufen verdünnten Meerwassers (Verdünnung nicht mit dest. Wasser, sondern mit Leitungswasser von bekanntem Bicarbonatgehalt und pH) bestimmt und in Prozenten der Leistung in Meerwasser ausgedrückt, so ergibt sich die in Abb. 3 dargestellte Kurve. Innerhalb der Versuchszeit scheint jeder Meerwasserkonzentration ein bestimmter Photosynthesewert zugeordnet zu sein, ein Verhalten, das auf das Vorhandensein von Gleichgewichtszuständen hindeutet, die zwischen dem photosynthetischen System und dem Zustand des Außenmediums bestehen.Infolge der überaus raschen Reaktion der Pflanzen auf Änderungen im Außenmedium wird als Arbeitshypothese die Vermutung ausgesprochen, daß an der Änderung der photosynthetischen Rate bei Übertragung in und Rückführung aus Süßwasser in Meerwasser nicht Zustandsveränderungen imZellinneren, sondern Zustände der Membranen verantwortlich sind, welche das Ausmaß der CO2-Permeation bestimmen und damit einen begrenzenden Faktor der Photosynthese darstellen.Mit 4 Textabbildungen  相似文献

The provision of healthcare in rural African communities is a highly complex and largely unsolved problem. Two main difficulties are the identification of individuals that are most likely affected by disease and the prediction of responses to health interventions. Social networks have been shown to capture health outcomes in a variety of contexts. Yet, it is an open question as to what extent social network analysis can identify and distinguish among households that are most likely to report poor health and those most likely to respond to positive behavioural influences. We use data from seven highly remote, post-conflict villages in Liberia and compare two prominent network measures: in-degree and betweenness. We define in-degree as the frequency in which members from one household are named by another household as a friends. Betweenness is defined as the proportion of shortest friendship paths between any two households in a network that traverses a particular household. We find that in-degree explains the number of ill family members, whereas betweenness explains engagement in preventative health. In-degree and betweenness independently explained self-reported health and behaviour, respectively. Further, we find that betweenness predicts susceptibility to, instead of influence over, good health behaviours. The results suggest that targeting households based on network measures rather than health status may be effective for promoting the uptake of health interventions in rural poor villages.  相似文献

Recent studies have implicated the dying cell as a potential reservoir of modified autoantigens that might initiate and drive systemic autoimmunity in susceptible hosts. A number of subunits of the exosome, a complex of 3'→5' exoribonucleases that functions in a variety of cellular processes, are recognized by the so-called anti-PM/Scl autoantibodies, found predominantly in patients suffering from an overlap syndrome of myositis and scleroderma. Here we show that one of these subunits, PM/Scl-75, is cleaved during apoptosis. PM/Scl-75 cleavage is inhibited by several different caspase inhibitors. The analysis of PM/Scl-75 cleavage by recombinant caspase proteins shows that PM/Scl-75 is efficiently cleaved by caspase-1, to a smaller extent by caspase-8, and relatively inefficiently by caspase-3 and caspase-7. Cleavage of the PM/Scl-75 protein occurs in the C-terminal part of the protein at Asp369 (IILD369↓G), and at least a fraction of the resulting N-terminal fragments of PM/Scl-75 remains associated with the exosome. Finally, the implications of PM/Scl-75 cleavage for exosome function and the generation of anti-PM/Scl-75 autoantibodies are discussed.  相似文献

New Zealand Obese (NZO) male mice develop a polygenic juvenile-onset obesity and maturity onset hyperinsulinemia. Approximately 50% transit to chronic hyperglycemia. Here we report on the proliferation of beta cells in relation to both the individual's metabolic status and structural parameters of the endocrine pancreas. Proliferating beta cells were quantified in pancreas sections by immunoenzymatic double staining of Ki-67 protein, as a marker for proliferating cells, and endocrine non-beta cells in order to distinguish them from beta cells. In normoglycemic NZO/Hl males Ki-67 labelling indices (IKi-67) of beta cells varied between 0.14 and 1.5%, and correlated significantly with both serum insulin levels and beta cell size. There was no correlation with the glycemic status. In diabetic males, beta cell size was increased. IKi-67 varied between 1 and 3%. The data suggest that the secretory activity of beta cells triggered by glucose, entailed changes in both beta cell hypertrophy and proliferation. As shown by morphometric measurements, beta cell expansion in diabetic mice was limited, in spite of high IKi-67 values. This suggested increased death rates of beta cells.  相似文献

In southern Bahia, Brazil, cabrucas are the traditional agroforests in which cacao trees are planted under thinned-out native forests. To analyze the role of cabrucas in tree species conservation, we inventoried the non-cocoa trees in 1.0 ha plots of cabruca in 16 cocoa farms and compared our results with a similar survey undertaken in the early 1960s in the same region to analyze the long term changes. We also interviewed 160 cocoa farmers to investigate their preferences for species and the main practices used in managing shade trees. The cabrucas showed high levels of tree diversity for an agroforestry system (Shannon index ranging from 2.21 to 3.52) and also high variation in structure and composition among the different farms. Forest specialist trees accounted for most species (63.9%) in the survey and were among the species most preferred by the farmers, although we found evidence that some of these trees are gradually being replaced by other species. Our results indicate that cabrucas are poor substitutes for undisturbed forests in terms of tree species richness, but their presence in human-altered landscapes is of utmost importance to the conservation of forest tree species as they increase overall heterogeneity and may serve as ecological corridors, additional habitats, and buffer zones.  相似文献
