To develop a mathematical model of the distribution and metabolism of rat corticotropin-releasing factor (rCRF), the time course of 125I-labelled rCRF in plasma was measured in male Sprague-Dawley rats (i) following a rapid injection of 24 ng rCRF/100 g body weight (BW), or (ii) following a rapid injection of 424 ng rCRF/100 g BW, or (iii) during an infusion at a rate ranging from 0.28 to 0.73 ng rCRF X min-1 X 100 g BW-1. The comparison of the one-, two-, and three-compartment models shows that the two-pool structure fits better to the dynamics of CRF in plasma as measured in each rat. Following a rapid injection the decay curve occurs in a biphasic manner; the early phase of disappearance is 25 times faster than the late one. There is no significant difference between the estimates of the metabolic clearance rate following both amplitudes of injection (0.40 +/- 0.06 and 0.48 +/- 0.05 mL X min-1 X 100 g BW-1). The volume of the first pool, 16.8 +/- 1.1 mL/100 g BW, is four times larger than the plasma volume. It would thus appear that CRF is rapidly distributed from plasma into several tissues which are represented in the first pool of the model. The mean residence time of every CRF molecule in the second compartment, from the moment of secretion to its elimination, is from three to four times longer than in the first one. The American southwest and northern Mexico has a great degree of endemic diversity compared with the rest of North America. The Pleistocene glaciations and the dispersal of species from glacial refuges in this region have been important engines for the production of biodiversity in the region. The New World Junonia are a recent radiation of butterflies that are thought to have spent time in these refuges during periods of glacial advancement. We have reconstructed the plausible movements and the contemporary geographic distributions of the five taxa (J. coenia, J. grisea, J. litoralis, J. nigrosuffusa and J. zonalis) that occur in the American southwest and northern Mexico using phenotypic and genotypic information primarily from specimens preserved in museum collections, supplemented with additional contemporary collections. Evidence of cryptic species and hybridization events were observed using mitochondrial haplotypes, genotypic variation at the nuclear wingless locus, multilocus DNA fingerprinting, patterns of larval host plant use, variation in life‐history traits and morphological characteristics. Based on these lines of evidence, and in spite of low levels of hybridization between them, we argue that all five Junonia taxa are independent evolutionary lineages. Junonia grisea and J. coenia are morphologically very similar, but differences in morphology, life‐history traits, nuclear genotypes and mitochondrial haplotypes suggest that they are a cryptic species pair, thus elevating J. grisea comb.n. to a full species when it had previously been considered to be a subspecies of J. coenia. 相似文献

Two filamentous, branched, and septate actinomycetes were isolated from field-collected and from axenic in vitro produced root nodules of Alnus crispa var. mollis Fern. host plant. After their transfer to a chemically defined medium, these nodule isolates could not be distinguished from each other on the basis of morphology, cultural reactions, and whole cell composition and were considered to be the same species. They were morphologically similar to the root nodule endophyte, but were incapable of nodulating aseptic host plants growing in a nitrogen-deficient substrate. Whole cells of the nodule isolates were used for the production of rabbit antibodies. The resulting specific antiisolate antibodies were conjugated with fluorescein isothiocyanate and used in staining tests of the nodule endophyte. The immunofluorescence reactions demonstrated the homology of the nodule isolates with the nodule endophyte. After pectinase degradation of the endophyte capsule, the indirect immunoferritin method corroborated the fluorescent anti-body (FA) staining reactions. There was no antigenic relationship between the nodule isolates and 13 known strains of actinomycetes as determined by the FA techique. Fluorescent antibody reactions of adsorbed conjugates suggested that endophytes of both Alnus crispa var. mollis Fern. and Alnus rugosa (DuRoi) Spreng. root nodules belong to a common serotype. The LL and mesoisomers of diaminopimelic acid were present in similar proportions in the nodule endophyte and in the nodule isolates. Glucose, mannose, and an unknown sugar were the predominant whole cell sugars in the nodule isolates, although trace amounts of arabinose and rhamnose were also displayed. Siliceous sinters that precipitate around modern hot spring systems are able to fossilize the indigenous microbial communities, forming molds that accurately outline the shape of the microorganisms. Over time, the biomass decays, and only silica molds or their infill may remain as evidence of the former living cells. However, little is known regarding the fidelity of such silica molds in terms of size and morphology, and the preservation of critical parameters for the identification of ancient silicified microorganisms by silica molds remains untested. Here we report experiments examining the formation of microbial molds of the cyanobacterium Synechococcus elongatus in silica gel. We demonstrate that post-depositional processes, primarily desiccation, are crucial for obtaining accurate and robust molds, and that initial desiccation acts to strengthen cell molds against further alteration. However, all silica gel treatments systematically created preservational biases (changes in size, additional structures) that may be misleading and may complicate the identification of fossil microorganisms. 相似文献

Bovine diarrhea virus (BVDV) causes a variety of economically important enteric and infertility problems in cattle. For that reason, several countries have eradicated the disease, and some others have schemes in progress to achieve freedom. Although there is a considerable amount of information about the risk of BVDV transmission through contaminated semen used for artificial insemination (AI), there is no evidence to indicate whether the resulting embryos, when used for embryo transfer, can lead to the transmission of BVDV to recipients or offspring. For this experiment, semen from a bull persistently infected with BVDV (105 50% tissue culture infective doses/mL NY strain) was used for insemination (two times at estrus) of BVDV-seronegative, superovulated cows (N = 35). Embryos were collected 7 days after insemination and subsequently were washed according to the International Embryo Transfer Society recommendations or left unwashed. Out of 302 collected oocytes and embryos, 173 (57%) were fertilized and the remaining 129 (43%) had degenerated. Infectious BVDV was detected in 24% (17/71) of unwashed and 10% (8/77) of washed embryos, and in all (N = 11) follicular fluid samples, oviductal epithelial cells, endometrium, and corpora lutea tissues as determined by the virus isolation test. After transfer of 39 washed embryos to 27 BVDV-seronegative recipients, 12 (44%) cows became pregnant and 17 calves free of BVDV and BVDV antibodies, including five sets of twins, were born. After embryo transfer, all pregnant and nonpregnant recipients remained free of BVDV and antibodies. In conclusion, results herein suggest that BVDV can be transmitted by AI resulting in the production of some proportion of contaminated embryos. Neuropsychiatric signs are critical in primary caregiving of Alzheimer patients and yet have been relatively ignored in murine models. In the present study, APPswe/PS1 bigenic mice had higher levels of irritability than non-transgenic controls as measured in the touch escape test. Moreover, APPswe/PS1 mice showed poorer nest building than controls and a higher duration of immobility in the forced swimming assay. These results are concordant with the hypothesis of increased apathy and depression-like behavior in an Alzheimer's disease model. In addition, APPswe/PS1 bigenic mice were deficient in retention of passive avoidance learning and left–right discrimination learning, concordant with previous findings in other Alzheimer-like models. 相似文献
