Blockage of the progress of a DNA replication fork in Escherichia coli can be ascribed to an inhibition of helicase action at the orientation-specific binding of a termination sequence (ter) by the ter-binding protein (Lee, E.H., Kornberg, A., Hidaka, M., Kobayashi, T., and Horiuchi, T. (1989) Proc. Natl. Acad. Sci. U.S.A. 86, 9104-9108). These observations have been extended to include the PriA helicase, thus confirming that blockage is general for helicases. The site of arrest of synthesis by a replication fork is at the very first nucleotide of the 22-base pair E. coli-terB sequence. Strand displacement by DNA polymerases is also inhibited, but is less profound and is orientation-specific. Background
Domestication of chicken is believed to have occurred in Southeast Asia, especially in Indus valley. However, non-inclusion of Indian red jungle fowl (RJF), Gallus gallus murghi in previous studies has left a big gap in understanding the relationship of this major group of birds. In the present study, we addressed this issue by analyzing 76 Indian birds that included 56 G. g. murghi (RJF), 16 G. g. domesticus (domestic chicken) and 4 G. sonneratii (Grey JF) using both microsatellite markers and mitochondrial D-loop sequences. The gene encoding protein n of the Escherichia coli primosome has been discovered in the rpsF-rpsR-rplI ribosomal protein operon and designated priB. The low copy number of PriB protein and the distinctive codon usage of its gene argue against its being a ribosomal protein. The regulatory protein AlgR2 in Pseudomonas aeruginosa positively regulates nucleoside diphosphate kinase (Ndk) and succinyl-CoA synthetase, enzymes critical in nucleoside triphosphate (NTP) formation. AlgR2 positively regulates the production of alginate, GTP, ppGpp and inorganic polyphosphate (poly P). An algR2 mutant with low levels of these metabolites has them restored by introducing and overexpressing either the algR2 or the ndk gene into the algR2 mutant. Thus, Ndk is involved in the formation of these compounds and largely prevents the death of the algR2 mutant, which occurs early in the stationary phase. We demonstrate that the 12 kDa Ndk–pyruvate kinase (Pk) complex, previously shown to generate predominantly GTP instead of all the NTPs, has a low affinity for the deoxynucleoside diphosphates and cannot generate the dNTPs needed for DNA replication and cell division; this complex may thus be involved in regulating the levels of both NTPs and dNTPs that modulate cell division and survival in the stationary phase. 相似文献

Rat lymphoma cells (Nb2) are exquisitely sensitive to lactogenic hormones and are an ideal system to study receptor-mediated signal transduction. The effect of human growth hormone (hGH) on macromolecular synthesis, intracellular cAMP concentrations and protein phosphorylation was investigated in Nb2 cells maintained in serum-free medium. hGH stimulated the incorporation of radiolabeled precursors into protein, RNA and DNA in a time-dependent manner. The concentration of hGH inducing half-maximal DNA synthesis was 11 pM, indicating that Nb2 cells cultured in serum-free medium maintain the same sensitivity to lactogen as cells in horse serum-containing medium. hGH over a period of 4 h had no effect on intracellular cAMP regardless of the presence or absence of isobutylmethylxanthine (IBMX). IBMX (250 microM), increased intracellular cAMP levels 2-fold indicating that the cAMP assay was sufficiently sensitive to detect relatively small changes in intracellular cAMP. Cyclic AMP had no effect on protein phosphorylation. However, hGH, prolactin and placental lactogen enhanced phosphorylation of many protein targets, as well as that of a specific protein (Mr = 29,000). Rat growth hormone, which is not mitogenic, had no effect on protein phosphorylation. These results suggest that lactogen-mediated Nb2 mitogenesis does not involve modulation of intracellular cAMP concentration and that cAMP-independent protein phosphorylation may play a role. 相似文献
