We had earlier identified a 110/120-kDa protein specific to nuclear matrix of rat pachytene spermatocytes (Behal, A., Prakash, K., and Rao, M.R.S. (1987) J. Biol. Chem. 262, 10898-10902). This protein is now shown to be a disulfide-linked homodimer of a 60-kDa polypeptide. Indirect immunofluorescence and Western blot analyses using anti-120-kDa polyclonal antibodies have shown that this protein is a component of the pore-complex lamina structure of spermatogonia. As germ cells enter meiotic prophase and the lamina structure disassembles, this polypeptide is redistributed in the nucleus and can be isolated as a component of synaptonemal complexes. Following meiotic division, this 60-kDa protein is relocalized in the lamina, then representing the sole major component of the lamina structure of round spermatids. The identity of the 60-kDa protein in the pore-complex lamina fraction and synaptonemal complexes was further confirmed by two-dimensional analysis of iodinated tryptic peptides. Such an analysis has also shown that the germ cell-specific 60-kDa protein is related but not identical to somatic lamin B.  相似文献

Urease was immobilized onto gelatin-poly (HEMA) copolymer by covalent linkage. Maximum amount of urease was immobilized onto the support at a pH of 8.5. The optimal pH of the immobilized urease was similar to that of free urease; the optimal temperature showed an increase of 10 °C over the free enzyme. The stability of the immobilized urease for a range of pH, temperature and shelf life was greater than the corresponding values for the free enzyme. Enzyme activities of N-acetylglucosaminyltransferase (GlcNAc-Tase) I-V involved in N-linked complex-type carbohydrate synthesis were determined in a non-metastatic hormone-dependent rat prostate tumor (R3327-H) and a related, hormone-independent variant metastasizing to lymph nodes and lungs (R3327-MatLyLu). In the metastasizing variant a significantly increased activity of both GlcNAc-Tase III and GlcNAc-Tase V was observed, whereas the activities of GlcNAc-Tase I and II were essentially unchanged. The increase in activity of GlcNAc-Tase III is particularly noteworthy since it indicates that elevated expression of this enzyme cannot be considered as an exclusive marker of hepatic malignancy.  相似文献

Hysterothylacium pseudotumbili n.sp. from the stomach and body cavity of marine fishes. Chrorinemus tol Couvier. Saurida undosquamis (Richardson), Lutianus russelli (Bleeker), Rastrilliger kanagurta (Cuvier) and Saurida pseudotumbili Dutt and Vidyasagar collected from Visakhapatnam (Bay of Bengal) is described and figured. A detailed examination has allowed us to erect a new species of Hysterothylacium to accommodate the worm. It shows remarkable differences from other known species of Hysterothylacium in respect of body measurements, the papillae on the lips arrangement, location of nerve ring and excretory pore, long intestinal caecum, position of vulva, number and arrangement of caudal papillae and size of the spicules. The new species is designated as Hysterothylacium pseudotumbili. Saurida pseudotumbili is the new host record. This is the first report of the genus in this host. Visakhapatnam coast is the new locality record.  相似文献

Bacopa monniera Wettst. (BM, syn. Herpestis monniera L; Scrophulariaceae), is an Ayurvedic drug used as a rasayana. Its fresh juice was earlier reported to have significant antiulcerogenic activity. In continuation, methanolic extract of BM (BME) standardized to bacoside-A content (percentage-38.0 ± 0.9), when given in the dose of 10–50 mg/kg, twice daily for 5 days, showed dose-dependent anti-ulcerogenic on various gastric ulcer models induced by ethanol, aspirin, 2 h cold restraint stress and 4 h pylorus ligation. BME in the dose of 20 mg/kg, given for 10 days, twice daily showed healing effects against 50% acetic acid-induced gastric ulcers. Further work was done to investigate the possible mechanisms of its action by studying its effect on various mucosal offensive acid-pepsin secretion and defensive factors like mucin secretion, mucosal cell shedding, cell proliferation and antioxidant activity in rats. BME 20 mg/kg showed no effect on acid-pepsin secretion, increased mucin secretion, while it decreased cell shedding with no effect on cell proliferation. BME showed significant antioxidant effect per se and in stressed animals. Thus, the gastric prophylactic and curative effects of BME may be due to its predominant effect on mucosal defensive factors.  相似文献

A rapid, simple and sensitive reversed-phase high-performance liquid chromatographic (HPLC) method has been developed for the measurement of acyclovir concentrations in human plasma and its use in bioavailability studies is evaluated. Unchanged acyclovir has been quantified without the introduction of an internal standard using the present method. Human plasma proteins were selectively precipitated by the addition of 7% perchloric acid to spiked plasma samples or to the plasma samples obtained after acyclovir administration to human volunteers and the mixture was spun at 1000 g for 10 min. The supernatant was directly injected into a Novaflex C18 column and detected at 254 nm. The mobile phase consisted of octane sulfonic acid buffer (pH 2.5) and methanol (92:08). The limit of quantitation for acyclovir in plasma was 20 ng/ml, which enabled the determination of the area under the curve (AUC) more precisely, that is, it is much closer to its extrapolated value. The present method has been successfully applied to samples from bioavailability studies.  相似文献

Anthers of Vitis latifolia L. (wild grape) cultured on Nitsch and Nitsch medium supplemented with 20 μM 2,4-D and 9 μM BAP produced callus after 4–6 weeks. Subculture of callus onto Nitsch and Nitsch medium containing 10 μM NAA produced somatic embryos within 6 weeks. On growth regulator-free Nitsch and Nitsch basal medium somatic embryos converted to plantlets in 6–8 weeks. One gram of callus produced more than 400 somatic embryos with 13.7% being converted to complete plantlets, which were subsequently established in soil.
