Summary

The site of yolk protein synthesis in crustaceans has long been a subject of controversy. A portion of the vitellogenin gene structure was reported recently in a freshwater giant prawn (Macrobrachium rosenbergii) and black tiger shrimp (Penaeus monodori), in which the hepatopancreas was confirmed to be the extraovarian site of vitellogenin synthesis. The ovary is also frequently reported to be the site of yolk protein synthesis in penaeid shrimp. The same PCR product was obtained using cDNA from the hepatopancreas or the ovary as a template. The deduced amino acid sequence of Vg in P. vannamei showed high identities of 57% and 78% with those from M. rosenbergii and P. monodon, respectively. The same location of the intron in the sequenced region of genomic DNA was also found between these three species. We therefore concluded that the hepatopancreas and ovary are sites of vitellogenin synthesis in P. vannamei. The partial structure of the vitellogenin gene is further presented.  相似文献

IN spite of the apparent simplicity of RNA bacteriophage, several proteins, both phage and bacterial, are required for the synthesis of Qβ RNA in vitro. The polymerase complex alone contains one phage-coded and three host proteins1,2. The specific role of these proteins in Qβ RNA replication is unknown, but because they demonstrate an associative interaction and are always found with active enzyme, it has been suggested that all four contribute to polymerase activity1.  相似文献

Chlorella emersonii (211/11n) was grown at external NaCl concentrationsranging between 1.0 and 335 mM (0.08–1.64 MPa). Previousstudies showed that there was no significant change in the internalconcentrations of Na+ or Cl over this range, the concentrationsremaining below 35 mM. Relative growth rates of C. emersoniiwere 30–45% lower in 335 mM NaCl than in 1.0 mM NaCl.Turgor pressure varied with the osmotic pressure of the growthmedium. Plots of cell volume versus (external osmotic pressure)–1indicated that cells grown in 1.0 mM NaCl (0.08 MPa) had turgorpressures ranging from 0.5 to 0.8 MPa, while cells in 335 mMNaCl (1.64 MPa) had turgor pressures of 0.0–0.14 MPa.Estimates of turgor pressure derived from the osmotic pressureof cell sap had a mean value of 0.6 MPa for cells in 1.0 mMNaCl, and 0.3 MPa for cells in 335 mM NaCl. The volumetric elasticmodulus () depended on the osmotic pressure of the growth medium: was 8.5 ± 1.7 MPa for cells grown in 1.0 mM NaCl, and0.9 ± 0.6 for cells in 335 mM NaCl. was measured bychanging turgor pressures over the range 0.0–0.5 MPa,and was found to be independent of turgor. Electron micrographsshowed that the walls of cells grown in 335 mM NaCl were 70%thicker than those grown in 1.0 mM NaCl. Other changes in cellularstructure were small, however, the area occupied by vacuolesincreased from 7% in cells grown in 1.0 mM NaCl to 14% in cellsin 335 mM. The percent osmotic volume of cells grown in 1.0–335mM NaCl (61 ± 17%, v/v) was similar to the percent watercontent (59 ± 13%, w/w). The vascular system in the rachis of ears of wheat (Triticumaestivum L. cvs Gamenya, Olympic and Bungulla) was examinedon material grown in the field and in a growth cabinet. In theinternodes, central and peripheral bundles were observed andtheir mean number and size were determined. A significant 1:1 relationship between the number of spikelets on the ear andthe number of central bundles at the base of the rachis wasestablished. The number of both central and peripheral bundlesdeclined acropetally along thé length of the rachis.The decline in peripheral bundles occurred mainly between internodes1 and 6, numbered from the base. The decline in central bundlesoccurred at a rate of less than one bundle per internode betweeninternodes 1 and 4, though in some ears, there was no decline;in larger ears, central bundles declined at a rate of one totwo bundles per internode between internodes 5 and 11. Aboveinternode 11, the rate of decline varied with ear size. Threecentral bundles consistently reached the terminal spikelet.The number and cross-sectional surface area of xylem vesselsand sieve tubes and the total vascular size also declined acropetallyalong the rachis. The decline in total vascular size was dueto (a) some bundles branching and to reductions in size, (b)the diversion or dropping of bundles into spikelets, or (c)a combination of (a) and (b). Gross morphology and internal structure of haustoria of Olaxphyllanthi are described in parasitism with a range of hosts,including roots of woody and herbaceous dicotyledons and certainmonocotyledons, and occasional instances of autoparasitism andhaustorial formation on monocotyledon rhizomes. Successful penetrationto xylem occurs on virtually all hosts across broad diameters,ages and anatomies of host root, but anatomical impedimentsto haustorial establishment and penetration are recorded forcertain host taxa. Each haustorium is a comparatively simpleand ephemeral structure. Its developing sucker (endophytic regionof the haustorium) spreads laterally around the surface of thehost xylem, yet never completely encircles the host stele. Damageto hosts is minimal and secondary thickening (of hosts) continueson the side of a host root opposite to a haustorium. The haustorialsucker lacks phloem and its interface with host xylem is comprisedalmost entirely (more than 98.7%) of parenchyma. The few terminatingtracheids at an interface lie in very close proximity to oroccasionally directly against exposed xylem vessels, but lumento lumen continuity between tracheary elements of the partnersis not achieved. Three dimensional reconstructions based onserial transverse sectioning indicate that well defined filesof tracheids connect back from an interface to the core of graniferoustracheary elements in the external body of the haustorium, andthence to the xylem of the parent parasite root. Olax phyllanthi was found to parasitize a wide range of taxain the native habitat in coastal heath, South-West Australia.All major life and growth forms were regularly exploited, includingmost woody dicotyledons (except members of the Myrtaceae), afew monocotyledons and cohabiting root hemiparasites. Initiationof haustoria occurred mostly in autumn (southern hemisphere),with some senescing the following summer, and others survivinguntil replaced by a new generation of haustoria the followingautumn. Seedlings increased in dry matter and contents of N,P, K, Mg and Ca during preparasitic development, but did notsurvive beyond 6 months if failing to establish haustoria ona suitable host. Plant dry matter, mineral content and haustorialnumber increased exponentially during subsequent parasitic development.Mean shoot:root d. wt ratios of 1st to 3rd-year plants lay withinthe range 1.2–1.4. Root systems were laterally extensiveand restricted to the top 40 cm of rooting substrate. Mean totalroot lengths of 2nd- and 3rd-year plants were 7.1m (n = 5) and60.9 m (n = 5), respectively. Haustoria comprised 0.7–3.5%of plant d. wt, with a mean of 7.9 haustoria (n = 10, 2nd- and3rd-year plants) per metre of root length. Comparisons of mineralconcentrations in dry matter of O. phyllanthi and of a rangeof commonly parasitized hosts showed the parasite to be muchricher on average than its hosts in K, P, and to a lesser extentin N, but not noticeably different in Mg and Ca. Climate and litter quality are primary drivers of terrestrial decomposition and, based on evidence from multisite experiments at regional and global scales, are universally factored into global decomposition models. In contrast, soil animals are considered key regulators of decomposition at local scales but their role at larger scales is unresolved. Soil animals are consequently excluded from global models of organic mineralization processes. Incomplete assessment of the roles of soil animals stems from the difficulties of manipulating invertebrate animals experimentally across large geographic gradients. This is compounded by deficient or inconsistent taxonomy. We report a global decomposition experiment to assess the importance of soil animals in C mineralization, in which a common grass litter substrate was exposed to natural decomposition in either control or reduced animal treatments across 30 sites distributed from 43°S to 68°N on six continents. Animals in the mesofaunal size range were recovered from the litter by Tullgren extraction and identified to common specifications, mostly at the ordinal level. The design of the trials enabled faunal contribution to be evaluated against abiotic parameters between sites. Soil animals increase decomposition rates in temperate and wet tropical climates, but have neutral effects where temperature or moisture constrain biological activity. Our findings highlight that faunal influences on decomposition are dependent on prevailing climatic conditions. We conclude that (1) inclusion of soil animals will improve the predictive capabilities of region‐ or biome‐scale decomposition models, (2) soil animal influences on decomposition are important at the regional scale when attempting to predict global change scenarios, and (3) the statistical relationship between decomposition rates and climate, at the global scale, is robust against changes in soil faunal abundance and diversity.  相似文献
