We analyzed the pattern of correlations among fitness components, herbivory, and resin characteristics in a natural all-aged stand of ponderosa pine, to infer the strength and mechanism of natural selection on plant chemistry. Male and female cone production were monitored yearly for 15 years, and levels of herbivory for 9 years in 165 trees. Resin flow rate and monoterpene composition were determined for these same trees. Multiple regression of fitness components on resin characteristics showed significant associations consistent with directional selection for increased resin flow rates and increased proportions of α- and β-pinene, myrcene and terpinolene. However, negative correlations among monoterpene fractions of the resin constrained the overall selection. Selective herbivory by aphids approached statistical significance and monoterpenes showed some (non-significant) effect as deterrents against deer browse. Resin characteristics were not correlated with attack by cone insects or porcupines. However, the association between resin characteristics and fitness is significantly different from that predicted by the path coefficients involving herbivores. Therefore the hypothesis that these herbivores mediate selection on the resin is not supported by the observed pattern of correlations among resin characteristics, herbivory, growth and fecundity. In this population, most of the association between resin characteristics and fitness appears to be mediated by some other factor independent of attack by herbivore species present. The red yeast Rhodotorula mucilaginosa produced an esterase that accumulated in the culture supernatant on induction with triacetin. The enzyme was specific for substrates bearing an O-acetyl group, but was relatively nonspecific for the rest of the molecule, which could consist of a phenol, a monosaccharide, a polysaccharide, or an aliphatic alcohol. The esterase was more active against acetylxylan and glucose beta-d-pentaacetate than were a number of esterases from plant and animal sources, when activities on 4-nitrophenyl acetate were compared. The enzyme exhibited Michaelis-Menten kinetics and was active over a broad pH range (5.5 to 9.2), with an optimum between pH 8 and 10. In addition, the enzyme retained its activity for 2 h at 55 degrees C. The yeast that produced the enzyme did not produce xylanase and, hence, is of interest for the production of acetylxylan esterase that is free of xylanolytic activity.  相似文献

Many theoretical studies in biological and physical sciences consider the dynamical behavior of ann-dimensional ordinary differential equation that contains a large number of independent parameters. A frequently asked question is, are there permissible parameter sets that result in periodic or chaotic behavior? The large number of distinct parameters often limits the feasibility of trial and error calculations. The large dimension and nonlinearity of the system make application of analytic methods at best difficult and at worst effectively impossible. It is shown here that a computational search for parameter-dependent transitions of attractor topology can be effected by constrained optimization of quantitative measures of dynamical behavior (Hurwitz polynomials, Floquet coefficients, Lyapunov exponents and correlation dimension). As an example, we examine a three-dimensional nonlinear ordinary differential equation containing seven parameters that was constructed by Goldbeter and Segel to model periodic synthesis of cyclic AMP inDictyostelium. A search for bifurcations to periodic solutions is made by minimizing Hurwitz coefficients subject to parameter constraints. By comparing four optimization algorithms, the defects and advantages of the procedure are identified. It is also argued that it may be possible to use this characterization of dynamics to construct optimal responses to dynamical diseases (those disorders that result from parameter-dependent bifurcations in physiological control systems).  相似文献

Summary A general method is presented for the formulation and numerical evaluation of mathematical models describing epithelial transport. The method is based on the principles of conservation of mass, and maintenance of electroneutrality within the cells and bathing solutions. It is therefore independent of the specific membrane transport mechanisms, and can be used to evaluate different models describing arbitrary transport processes (including passive, active and cotransport processes). Detailed numerical methods are presented that allow computation of steady-state and transient responses under open-circuit, current-clamp and voltage-clamp conditions, using a general-purpose laboratory minicomputer. To evaluate the utility of this approach, a specific model is presented that is consistent with the Koefoed-Johnson and Ussing hypothesis of sodium transport in tight epithelia (Acta Physiol. Scand. 42:298–308, 1958). This model considers passive transport of an arbitrary number of permeant solutes, active transport of sodium and potassium, and osmotically induced water transport across the apical and basolateral membranes. Results of the model are compared to published experimental measurements in rabbit urinary bladder epithelium.  相似文献

Expression plasmids carrying the coding sequence of mature human interleukin 1 beta (IL 1 beta) linked either to a Met start codon, or fused to different efficient Escherichia coli secretion signal sequences, have been constructed. In the latter case, we used signal peptides derived either from an outer membrane protein (OmpA) or from a periplasmic protein (PhoA). The synthesis of IL1 beta from these fusions was investigated in an otherwise strictly isogenic context using identical conditions of derepression and culture media. The Met-IL1 beta fusion produced a soluble cytoplasmic protein which could be released from the cells by osmotic shock whereas the OmpA and PhoA fusions were always insoluble. The extent of sOmpA-IL1 beta maturation was found to vary from 50 to 100%, mainly depending on the medium used, whereas no significant maturation of the signal peptide could be detected in the case of the sPhoA-IL1 beta fusion. Immuno-electron microscopy revealed that the sOmpA-IL1 beta fusion was targeted to the inner membrane, whereas the sPhoA-IL1 beta fusion remained within the cytoplasm and thus did not appear to enter the secretion pathway. Amplifying the E. coli signal peptidase lep gene on a multicopy plasmid did not improve signal peptide removal from sOmpA-IL1 beta. Moreover, these E. coli secretion vectors allowed us to produce, in high levels, IL1 beta fragments which otherwise could not be stably accumulated within the cytoplasmic compartment.  相似文献

Social behavior was studied in four (one male and three females) adult, captivebred Matschie's tree kangaroos (Dendrolagus matschiei) over a 124 day span in a large, naturalistic exhibit at the Woodland Park Zoological Gardens, Seattle, WA. A relatively high rate of social interaction occurred (15.2 per hour), over half of which consisted of approaches and nose contacts. The male initiated 54.1% of all social behaviors. Females tended to respond aggressively toward the male (avoid, bite, cuff, or swipe). The male initiated more affiliative behaviors and the females initiated more agonistic behaviors toward both the male and other females. A clear-cut dominance hierarchy could not be determined; reversals (i.e., cases in which the “subordinate” supplanted the “dominant”) occurred 43% of the time overall. Non-aggressive contact behavior consisted primarily of olfactory examination (nose contact). Allogrooming was extremely rare. The high level of agonism and lack of consistent association among individuals suggests that this species may be solitary in the wild. Two joeys were found dead on the floor during the course of the study, and based on previous findings, infanticide or behavioral stress was suspected to be the cause. Successful reproduction did not occur until females were isolated by removing other conspecifics from the enclosure.  相似文献

The role of the C terminus of Escherichia coli DNA helicase II (UvrD), a region outside the conserved helicase motifs, was investigated by using three mutants: UvrDDelta107C (deletion of the last 107 C-terminal amino acids), UvrDDelta102C, and UvrDDelta40C. This region, which lacks sequence similarity with other helicases, may function to tailor UvrD for its specific in vivo roles. Genetic complementation assays demonstrated that mutant proteins UvrDDelta107C and UvrDDelta102C failed to substitute for the wild-type protein in methyl-directed mismatch repair and nucleotide excision repair. UvrDDelta40C protein fully complemented the loss of helicase II in both repair pathways. UvrDDelta102C and UvrDDelta40C were purified to apparent homogeneity and characterized biochemically. UvrDDelta102C was unable to bind single-stranded DNA and exhibited a greatly reduced single-stranded DNA-stimulated ATPase activity in comparison to the wild-type protein (kcat = 0.01% of the wild-type level). UvrDDelta40C was slightly defective for DNA binding and was essentially indistinguishable from wild-type UvrD when single-stranded DNA-stimulated ATP hydrolysis and helicase activities were measured. These results suggest a role for a region near the C terminus of helicase II in binding to single-stranded DNA.  相似文献

Proteus mirabilis is a common opportunistic Gram-negative uropathogen that infects the upper urinary tract. We have examined the role of the nonagglutinating fimbriae (NAF) of P. mirabilis in mediating bacterial adhesion to cell surface receptors. Purified NAF of P. mirabilis were demonstrated to bind to a number of glycolipids, including asialo-GM1, asialo-GM2, and lactosyl ceramide (LacCer) in solid-phase binding assays and in thin layer chromatography (TLC) overlay assays. Furthermore, preincubation of the biotinylated NAF (Bt-NAF) with anti-NAF monoclonal antibodies resulted in inhibition of NAF binding to immobilized asialo-GM1, asialo-GM2, and LacCer. In adherence assays, P. mirabilis binding to Madin-Darby canine kidney (MDCK) cells was inhibited by murine anti-asialo-GM1 monoclonal antibodies H2G10 to about 50% of the binding level in the absence of the antibody, specific for the terminal beta-galactopyranosyl residue of asialo-GM1 (Harrison et al. 1998). The results of this study suggest that NAF of P. mirabilis recognize a GalNAc beta 1-4Gal moiety present in the ganglio-series of asialoglycolipids, and that the terminal beta-galactopyranosyl-containing glycoconjugates play a role in NAF-mediated adherence of P. mirabilis to MDCK cells. Similarly to other bacteria, P. mirabilis NAF was also shown to express the LacCer specificity.  相似文献
