Nastiness of cancer does not only reside in the corruption of cancer cells by genetic aberrations that drive their sustained proliferative power—the roots of malignancy—but also in its aptitude to reciprocally sculpt its surrounding environment and cellular stromal ecosystem, in such a way that the corrupted tumor microenvironment becomes a full pro-tumorigenic entity. Such a contribution had been appreciated three decades ago already, with the discovery of tumor angiogenesis and extracellular matrix remodeling. Nevertheless, the recent emergence of the tumor microenvironment as the critical determinant in cancer biology is paralleled by the promising therapeutic potential it carries, opening alternate routes to fight cancer. The study of the tumor microenvironment recruited numerous lead-scientists over the years, with distinct perspectives, and some of them have kindly accepted to contribute to the elaboration of this special issue entitled Tumor microenvironment indoctrination: An emerging hallmark of cancer.  相似文献

Aim We investigated the phylogeography, geographical variation in leaf morphology, freezing tolerance and climatic niches of two widespread evergreen sister oak species (Quercus) in the series Virentes. Location South‐eastern USA, Mexico and Central America. Methods Nuclear microsatellites and non‐recombining nuclear and chloroplast DNA sequences were obtained from trees throughout the range of two sister lineages of live oaks, represented by Quercus virginiana in the temperate zone and Q. oleoides in the tropics. Divergence times were estimated for the two major geographical and genetic breaks. Differentiation in leaf morphology, analysed from field specimens, was compared with the molecular data. Freezing sensitivities of Q. virginiana and Q. oleoides populations were assessed in common garden experiments. Results The geographical break between Q. virginiana and Q. oleoides was associated with strong genetic differentiation of possible early Pleistocene origin and with differentiation in freezing sensitivity, climatic envelopes and leaf morphology. A second important geographical and genetic break within Q. oleoides between Costa Rica and the rest of Central America showed a mid‐Pleistocene divergence time and no differentiation in leaf morphology. Population genetic differentiation was greater but genetic diversity was lower within the temperate Q. virginiana than within the tropical Q. oleoides, and genetic breaks largely corresponded to breaks in leaf morphology. Main conclusions Two major breaks, one between Mexico and the USA at the boundary of the two species, and a more recent one within Q. oleoides between Honduras and Costa Rica, implicate climatic changes as potential causes. The latter break may be associated with the formation of the Cordillera de Guanacaste, which was followed by seasonal changes in precipitation. In the former case, an ‘out of the tropics’ scenario is hypothesized, in which the acquisition of freezing tolerance in Q. virginiana permitted colonization of and expansion in the temperate zone, while differences in climatic tolerances between the species limited secondary contact. More pronounced Pleistocene changes in climate and sea level in the south‐eastern USA relative to coastal Mexico and Central America may explain the greater population differentiation within temperate Q. virginiana and greater genetic diversity in tropical Q. oleoides. These patterns are predicted to hold for other taxa that span temperate and tropical zones of North and Central America.  相似文献

Technological and theoretical advances over the past two decades have allowed researchers to quantify eggshell color in ways that were not previously possible. However, differences among studies in the timing of color measurements during laying or incubation and inclusion of data from inviable eggs may affect the results of these studies as well as conclusions based on comparisons among studies. To determine the effect of the timing of color measurements, we compared the color of the eggs of Mountain Bluebirds (Sialia currucoides) during both laying/early incubation and late incubation. We also assessed the influence of egg viability on eggshell color by comparing viable and inviable eggs from the same clutch. We found that all color metrics investigated (blue‐green and UV chroma, brightness, and hue) were significantly different between early and late incubation, and viable and inviable eggs. However, color metrics of eggs measured during early and late incubation and of viable and inviable eggs in the same clutch were correlated. Our results suggest that the timing of color measurements and the viability of eggs have important effects on eggshell color and, therefore, in future studies, investigators should always provide information about the timing of measurements and the viability of eggs measured. Our results also suggest that comparisons among studies where eggshell color was measured at different times and/or the viability of eggs was not determined or reported are possible, given that the color metrics of viable and inviable eggs and of eggs measured at different times in our study were correlated, but those comparisons should be interpreted with caution.  相似文献

Recent studies investigating the evolution of genome size diversity in ferns have shown that they have a distinctive genome profile compared with other land plants. Ferns are typically characterized by possessing medium‐sized genomes, although a few lineages have evolved very large genomes. Ferns are different from other vascular plant lineages as they are the only group to show evidence for a correlation between genome size and chromosome number. In this study, we aim to explore whether the evolution of fern genome sizes is not only shaped by chromosome number changes arising from polyploidy but also by constraints on the average amount of DNA per chromosome. We selected the genus Asplenium L. as a model genus to study the question because of the unique combination of a highly conserved base chromosome number and a high frequency of polyploidy. New genome size data for Asplenium taxa were combined with existing data and analyzed within a phylogenetic framework. Genome size varied substantially between diploid species, resulting in overlapping genome sizes among diploid and tetraploid spleenworts. The observed additive pattern indicates the absence of genome downsizing following polyploidy. The genome size of diploids varied non‐randomly and we found evidence for clade‐specific trends towards larger or smaller genomes. The 578‐fold range of fern genome sizes have arisen not only from repeated cycles of polyploidy but also through clade‐specific constraints governing accumulation and/or elimination of DNA.  相似文献

Although genetic and plastic responses are sometimes considered as unrelated processes, their phenotypic effects may often align because genetic adaptation is expected to mirror phenotypic plasticity if adaptive, but run counter to it when maladaptive. Because the magnitude and direction of this alignment has further consequences for both the tempo and mode of adaptation, they are relevant for predicting an organisms’ reaction to environmental change. To better understand the interplay between phenotypic plasticity and genetic change in mediating adaptive phenotypic variation to climate variability, we here quantified genetic latitudinal variation and thermal plasticity in wing loading and wing shape in two closely related and widespread sepsid flies. Common garden rearing of 16 geographical populations reared across multiple temperatures revealed that wing loading decreases with latitude in both species. This pattern could be driven by selection for increased dispersal capacity in the cold. However, although allometry, sexual dimorphism, thermal plasticity and latitudinal differentiation in wing shape all show similar patterns in the two species, the relationship between the plastic and genetic responses differed between them. Although latitudinal differentiation (south to north) mirrored thermal plasticity (hot to cold) in Sepsis punctum, there was no relationship in Sepsis fulgens. While this suggests that thermal plasticity may have helped to mediate local adaptation in S. punctum, it also demonstrates that genetic wing shape differentiation and its relation to thermal plasticity may be complex and idiosyncratic, even among ecologically similar and closely related species. Hence, genetic responses can, but do not necessarily, align with phenotypic plasticity induced by changing environmental selection pressures.  相似文献

Background

Quercus oleoides Cham. and Schlect., tropical live oak, is a species of conservation importance in its southern range limit of northwestern Costa Rica. It occurs in high-density stands across a fragmented landscape spanning a contrasting elevation and precipitation gradient. We examined genetic diversity and spatial genetic structure in this geographically isolated and genetically distinct population. We characterized population genetic diversity at 11 nuclear microsatellite loci in 260 individuals from 13 sites. We monitored flowering time at 10 sites, and characterized the local environment in order to compare observed spatial genetic structure to hypotheses of isolation-by-distance and isolation-by-environment. Finally, we quantified pollen dispersal distances and tested for local adaptation through a reciprocal transplant experiment in order to experimentally address these hypotheses.

Results

High genetic diversity is maintained in the population and the genetic variation is significantly structured among sampled sites. We identified 5 distinct genetic clusters and average pollen dispersal predominately occurred over short distances. Differences among sites in flowering phenology and environmental factors, however, were not strictly associated with genetic differentiation. Growth and survival of upland and lowland progeny in their native and foreign environments was expected to exhibit evidence of local adaptation due to the more extreme dry season in the lowlands. Seedlings planted in the lowland garden experienced much higher mortality than seedlings in the upland garden, but we did not identify evidence for local adaptation.

Conclusion

Overall, this study indicates that the Costa Rican Q. oleoides population has a rich population genetic history. Despite environmental heterogeneity and habitat fragmentation, isolation-by-distance and isolation-by-environment alone do not explain spatial genetic structure. These results add to studies of genetic structure by examining a common, tropical tree over multiple habitats and provide information for managers of a successional forest in a protected area.  相似文献

Climate warming is affecting the Arctic in multiple ways, including via increased dominance of deciduous shrubs. Although many studies have focused on how this vegetation shift is altering nutrient cycling and energy balance, few have explicitly considered effects on tundra fauna, such as the millions of migratory songbirds that breed in northern regions every year. To understand how increasing deciduous shrub dominance may alter breeding songbird habitat, we quantified vegetation and arthropod community characteristics in both graminoid and shrub dominated tundra. We combined measurements of preferred nest site characteristics for Lapland longspurs (Calcarius lapponicus) and Gambel's White‐crowned sparrows (Zonotrichia leucophrys gambelii) with modeled predictions for the distribution of plant community types in the Alaskan arctic foothills region for the year 2050. Lapland longspur nests were found in sedge‐dominated tussock tundra where shrub height does not exceed 20 cm, whereas White‐crowned sparrows nested only under shrubs between 20 cm and 1 m in height, with no preference for shrub species. Shrub canopies had higher canopy‐dwelling arthropod availability (i.e. small flies and spiders) but lower ground‐dwelling arthropod availability (i.e. large spiders and beetles). Since flies are the birds' preferred prey, increasing shrubs may result in a net enhancement in preferred prey availability. Acknowledging the coarse resolution of existing tundra vegetation models, we predict that by 2050 there will be a northward shift in current White‐crowned sparrow habitat range and a 20–60% increase in their preferred habitat extent, while Lapland longspur habitat extent will be equivalently reduced. Our findings can be used to make first approximations of future habitat change for species with similar nesting requirements. However, we contend that as exemplified by this study's findings, existing tundra modeling tools cannot yet simulate the fine‐scale habitat characteristics that are critical to accurately predicting future habitat extent for many wildlife species.  相似文献
