Genotypes of Solanum tuberosum, assessed for their differential response to a protoplast regeneration protocol, were identified. This enabled comparisons to be made in relation to their abilities to regenerate into dividing cells and colonies. Protoplasts from genotypes with contrasting regeneration responses were plated as single or mixed genotype cultures at various plating densities in replicated, randomised experiments, and the effect on the regeneration of particular cell types observed. Protoplasts from single genotype cultures showed intergenotypic variation in the extent of cell regeneration and there were significant effects of genotype mixing and density × genotype mixing interactions. Overall, the effect of mixing was beneficial to a regenerating culture but significant density genotype mixing interactions showed only a positive benefit at the lowest plating density. The protoplast mixing phenomenon was not correlated with the behaviour of the same genotypes at the plantlet level in experiments with in vitro meristem cultures.  相似文献

Artificial nest experiments have been used in approximately 80 recent studies to identify a wide range of factors influencing reproductive success of birds. Despite the same factors being examined in different studies, few consistent patterns have emerged from these experiments. Analysis of reproductive success at artificial and natural nests reveals that reproductive success measured with use of artificial nests is frequently underestimated. In addition, several studies suggest that different species of predators may be differentially attracted to the two types of nest. These results call the utility of artificial nests into question as a tool for elucidating factors influencing the success of natural nests. Analysis of the methodology used in artificial nest experiments reveals that in general there is poor correspondence between the appearance of experimental setups and the natural systems they attempt to model. If future experiments with artificial nests are to contribute to theory, the quality of experiments must improve and the assumptions underlying the artificial approach must be tested. Collecting this information will be relatively simple compared with the time-consuming task of experimenting with natural nests. Until these data are collected, conclusions about nesting ecology derived from the use of artificial nest experiments should be treated as preliminary.  相似文献

The higher‐level phylogeny of the order Hemiptera remains a contentious topic in insect systematics. The controversy is chiefly centred on the unresolved question of whether or not the hemipteran suborder Auchenorrhyncha (including the extant superfamilies Fulgoroidea, Membracoidea, Cicadoidea and Cercopoidea) is a monophyletic lineage. Presented here are the results of a multilocus molecular phylogenetic investigation of relationships among the major hemipteran lineages, designed specifically to address the question of Auchenorrhyncha monophyly in the context of broad taxonomic sampling across Hemiptera. Phylogenetic analyses (maximum parsimony, maximum likelihood and Bayesian inference) were based on DNA nucleotide sequence data from seven gene regions (18S rDNA, 28S rDNA, histone H3, histone 2A, wingless, cytochrome c oxidase I and NADH dehydrogenase subunit 4) generated from 86 in‐group exemplars representing all major lineages of Hemiptera (plus seven out‐group taxa). All combined analyses of these data recover the monophyly of Auchenorrhyncha, and also support the monophyly of each of the following lineages: Hemiptera, Sternorrhyncha, Heteropterodea, Heteroptera, Fulgoroidea, Cicadomorpha, Membracoidea, Cercopoidea and Cicadoidea. Also presented is a review of the major lines of morphological and molecular evidence for and against the monophyly of Auchenorrhyncha.  相似文献

Soil is the largest reservoir of organic carbon (C) in the terrestrial biosphere and soil C has a relatively long mean residence time. Rising atmospheric carbon dioxide (CO2) concentrations generally increase plant growth and C input to soil, suggesting that soil might help mitigate atmospheric CO2 rise and global warming. But to what extent mitigation will occur is unclear. The large size of the soil C pool not only makes it a potential buffer against rising atmospheric CO2, but also makes it difficult to measure changes amid the existing background. Meta‐analysis is one tool that can overcome the limited power of single studies. Four recent meta‐analyses addressed this issue but reached somewhat different conclusions about the effect of elevated CO2 on soil C accumulation, especially regarding the role of nitrogen (N) inputs. Here, we assess the extent of differences between these conclusions and propose a new analysis of the data. The four meta‐analyses included different studies, derived different effect size estimates from common studies, used different weighting functions and metrics of effect size, and used different approaches to address nonindependence of effect sizes. Although all factors influenced the mean effect size estimates and subsequent inferences, the approach to independence had the largest influence. We recommend that meta‐analysts critically assess and report choices about effect size metrics and weighting functions, and criteria for study selection and independence. Such decisions need to be justified carefully because they affect the basis for inference. Our new analysis, with a combined data set, confirms that the effect of elevated CO2 on net soil C accumulation increases with the addition of N fertilizers. Although the effect at low N inputs was not significant, statistical power to detect biogeochemically important effect sizes at low N is limited, even with meta‐analysis, suggesting the continued need for long‐term experiments.  相似文献

Cryptic belowground organisms are difficult to observe and their responses to global changes are not well understood. Nevertheless, there is reason to believe that interactions among above- and belowground communities may mediate ecosystem responses to global change. We used grassland mesocosms to manipulate the abundance of one important group of soil organisms, arbuscular mycorrhizal (AM) fungi, and to study community and ecosystem responses to CO2 and N enrichment. Responses of plants, AM fungi, phospholipid fatty acids and community-level physiological profiles were measured after two growing seasons. Ecosystem responses were examined by measuring net primary production (NPP), evapotranspiration, total soil organic matter (SOM), and extractable mineral N. Structural equation modeling was used to examine the causal relationships among treatments and response variables. We found that while CO2 and N tended to directly impact ecosystem functions (evapotranspiration and NPP, respectively), AM fungi indirectly impacted ecosystem functions by influencing the community composition of plants and other root fungi, soil fungi and soil bacteria. We found that the mycotrophic status of the dominant plant species in the mesocosms determined whether the presence of AM fungi increased or decreased NPP. Mycotrophic grasses dominated the mesocosm communities during the first growing season, and the mycorrhizal treatments had the highest NPP. In contrast, nonmycotrophic forbs were dominant during the second growing season and the mycorrhizal treatments had the lowest NPP. The composition of the plant community strongly influenced soil N, and the community composition of soil organisms strongly influenced SOM accumulation in the mesocosms. These results show how linkages between above- and belowground communities can determine ecosystem responses to global change.  相似文献

ABSTRACT The Trivers-Willard (1973) model suggests maternal control of offspring sex, in utero or by the end of parental investment, may be an adaptive advantage in some species. We tested for differential sex allocation using 11,408 known-sex fetal elk (Cervus elaphus) from biological collections and hunter harvest returns from 2 southwestern Montana, USA, elk populations (1961–2007). We included maternal and environmental condition covariates measured pre- and postconception and throughout pregnancy. Results suggested that adult female elk in southwest Montana did not differentially invest in male offspring when conditions were beneficial. We found evidence that, when the Northern Yellowstone elk herd was at low density, beneficial spring (May-Jun) growing conditions, as indexed by a local precipitation measure and a regional drought indicator, correlated with production of more female fetuses (1 SD increase in precipitation and 1 SD decrease in drought resulted in 6% and 5% more F fetuses, respectively). In the same herd, we found evidence that improved maternal condition, as indexed by kidney fat mass and heart fat mass, also correlated with production of more female fetuses (1 SD increase in kidney fat mass and heart fat mass resulted in 8% more F fetuses). When the same elk herd reached higher densities under different ecological conditions, no covariate was associated with a deviation in the 50:50 female-to-male sex ratio. Similarly, there was no association between covariates and fetal sex ratios in a nearby elk herd at high population density. In modeling, wildlife managers should consider factors that could alter sex ratios at birth, and also how biased sex ratios postpartum could affect population models.  相似文献

Many hypotheses attempt to explain why younger, less experienced birds have a relatively low reproductive output. We evaluated reproductive patterns of marked American Kestrels Falco sparverius nesting in boxes in southwestern Idaho from 1992 to 2006 to test predictions of these hypotheses. Results were consistent with the selection (differential mortality) hypothesis and did not support the constraint, restraint or recruitment hypotheses. Most known-age Kestrels nested in their first year of life, and there was no apparent short-term or long-term reproductive advantage to delayed breeding. The number of years that Kestrels nested in study area boxes ranged from 1 to 6 years, with most Kestrels nesting in only 1 year. Reproductive rates were higher for birds with at least 1 year of nesting experience than for birds nesting in boxes for the first time. After 2 years of nesting, reproductive rates levelled off; there was no evidence for additional improvement or for senescence. Differences in reproductive output with experience/age were due to variation among and not within individuals. Individuals that nested in more than 1 year had similar reproductive rates in their first and second years. Poor producers either died or dispersed after 1 year of nesting in study area boxes. Successful females that nested early in the season and successful males that had been produced locally had the highest probability of returning to nest in a subsequent year.  相似文献

Abstract The absence of small birds from many suburban areas may be due to adverse garden characteristics, interspecific aggression or human behaviour such as supplementary food provisioning that encourages predators. We investigated the relationship between these factors and the presence of seven small bird species in Sydney through a community‐based survey. The survey was conducted by participants over a 7‐day period between 7 am and 10 am in November and early December 2000. Three dominant species, the noisy miner (Manorina melanocephala), pied currawong (Strepera graculina) and common myna (Acridotheres tristis) were each present in over 59% of gardens. Each small bird species was present in less than 40% of gardens. All small birds were negatively associated with noisy miners, but only the silvereye (Zosterops lateralis) was negatively associated with pied currawongs. None of the species of small birds was negatively associated with common mynas. Four species of small birds were associated with at least one habitat variable, notably the proportion of native vegetation. Although more birds were recorded in gardens in which meat was provided, there were significantly fewer small birds in these gardens. There were also more birds recorded in gardens where seed was provided, with red‐browed finches (Neochmia temporalis) positively associated with seed provisioning in most regions of Sydney. The presence of dogs and cats was not related to the total abundance of birds overall or small birds in gardens. While garden characteristics may influence the presence of small birds to some degree, the presence of noisy miners, a species that are thought to aggressively exclude other species from their territories, is likely to be an important influence on these species in suburban areas. Furthermore, supplementary feeding by people is likely to negatively influence some small birds. The presence of carnivorous pets does not seem to influence the presence of small birds at the scale of the individual garden.  相似文献

Legumes form a symbiotic interaction with Rhizobiaceae bacteria, which differentiate into nitrogen‐fixing bacteroids within nodules. Here, we investigated in vivo the pH of the peribacteroid space (PBS) surrounding the bacteroid and pH variation throughout symbiosis. In vivo confocal microscopy investigations, using acidotropic probes, demonstrated the acidic state of the PBS. In planta analysis of nodule senescence induced by distinct biological processes drastically increased PBS pH in the N2‐fixing zone (zone III). Therefore, the PBS acidification observed in mature bacteroids can be considered as a marker of bacteroid N2 fixation. Using a pH‐sensitive ratiometric probe, PBS pH was measured in vivo during the whole symbiotic process. We showed a progressive acidification of the PBS from the bacteroid release up to the onset of N2 fixation. Genetic and pharmacological approaches were conducted and led to disruption of the PBS acidification. Altogether, our findings shed light on the role of PBS pH of mature bacteroids in nodule functioning, providing new tools to monitor in vivo bacteroid physiology.  相似文献

In primates, females typically drive the evolution of the social system and present a wide diversity of social structures. To understand this diversity, it is necessary to document the consistency and/or flexibility of female social structures across and within species, contexts, and environments. Macaques (Macaca sp.) are an ideal taxon for such comparative study, showing both consistency and variation in their social relations. Their social styles, constituting robust sets of social traits, can be classified in four grades, from despotic to tolerant. However, tolerant species are still understudied, especially in the wild. To foster our understanding of tolerant societies and to assess the validity of the concept of social style, we studied female crested macaques, Macaca nigra, under entirely natural conditions. We assessed their degree of social tolerance by analyzing the frequency, intensity, and distribution of agonistic and affiliative behaviors, their dominance gradient, their bared‐teeth display, and their level of conciliatory tendency. We also analyzed previously undocumented behavioral patterns in grade 4 macaques: reaction upon approach and distribution of affiliative behavior across partners. We compared the observed patterns to data from other populations of grade 4 macaques and from species of other grades. Overall, female crested macaques expressed a tolerant social style, with low intensity, frequently bidirectional, and reconciled conflicts. Dominance asymmetry was moderate, associated with an affiliative bared‐teeth display. Females greatly tolerated one another in close proximity. The observed patterns matched the profile of other tolerant macaques and were outside the range of patterns of more despotic species. This study is the first comprehensive analysis of females’ social behavior in a tolerant macaque species under natural conditions and as such, contributes to a better understanding of macaque societies. It also highlights the relevance of the social style concept in the assessment of the degree of tolerance/despotism in social systems. Am. J.
