Responses of individual roots of the widely cultivated cactus Opuntia ficus-indica to salinity stress were evaluated using a split-root system. Three roots from a plant with at least 20 roots were isolated from the remainder of the root system and exposed to 0, 30 or 100 mol m-3 NaCl for up to 28 d. Cortical cells became shorter and lateral root development was substantially reduced as salinity increased. Compared with the control, the increase in dry weight for the isolated roots was reduced 40% by 30 mol m-3 NaCl and 93% by 100mol m-3 NaCl. The sodium content of roots increased only two-fold with increasing salinity. Respiration rates of roots exposed to 30 or 100 mol m-3 NaCl were higher than those of the control. Carbon accumulation in roots measured 2 d after exposing shoots to 14CO2 was not initially affected by 30 mol m-3 NaCl but was substantially reduced at 100 mol m-3 NaCl. Thus, roots exposed to short periods of moderate salinity stress maintained sufficient carbon sink strength for continued growth of the roots. Moreover, increased salinity led to decreased efficiency of carbon usage for the expansion of root surface area.  相似文献

Snails were collected from seven chalk grassland sites, in orderto assess variation in snail faunas with respect to location,vegetation, and management practices. Four of the sites weregrazed, intermittently to continuously, and three had been irregularlyburned and mown. Snails that are characteristic of the dry,open conditions of chalk grasslands made up 28% to 94% of allindividuals, and 33% to 75% of all species at the differentsites. Open-country snails tended to be relatively more commonat sites that had been consistently grazed than at sites thatwere mown or burned, although some heavily grazed sites showedlow overall snail abundance. Sites varied with respect to turfheight, the size of gaps in the turf, soil moisture, and higherplant species richness, but none of these site characteristicswere good predictors of the proportion of open-country snails.Comparisons of fresh and old shells at any given site can indicatewhether particular species have changed in relative abundanceover time. No changes were evident at two sites, Wylye Downand Galley Hill, but three of nine species changed at AstonRowant South, and four of ten species at Castle Hill. As a group,open-country snails declined relative to other snails at justone site, Castle Hill, which had been consistently grazed. Thisstudy confirms site-to-site variability in chalk grassland snailfaunas, while finding faunal composition to be generally constantthrough time within each site. Microsatellite data are widely used to test ecological and evolutionary hypotheses in wild populations. In this paper, we consider three typical sources of scoring errors capable of biasing biological conclusions: stuttering, large‐allele dropout and null alleles. We describe methods to detect errors and propose conventions to mitigate scoring errors and report error rates in studies of wild populations. Finally, we discuss potential bias in ecological or evolutionary conclusions based on data sets containing these scoring errors.  相似文献

Eastern chipmunks are small forest rodents for which ecological data at multiple spatial scales exist, but no molecular markers have been developed that provide adequate resolution to examine genetic structure at fine scales in this species. Thus, we developed 12 variable microsatellite loci, which amplified in three multiplexed reactions, and screened them in 27 individuals from northern Indiana, USA. Observed heterozygosities across loci ranged from 0.26 to 0.91 and number of alleles per locus ranged from three to 10. We will use these markers along with ecological data to understand population dynamics of eastern chipmunks at small spatial scales.  相似文献

In order to complement ecological information with genetic data we isolated and characterized 14 polymorphic microsatellite markers from raccoons (Procyon lotor). Three multiplexed panels comprising the loci were developed and 29 individuals from a contiguous habitat patch in northern Indiana, USA were genotyped. The number of alleles per locus ranged from four to 18, and overall heterozygosities ranged from 0.31 to 1.00. One locus was identified as possibly being X‐linked, since males appeared to be hemizygous. Data generated using these markers will be used to further our understanding of small‐scale raccoon population dynamics in a highly fragmented landscape.  相似文献

CHLOROPLASTS are more permeable to amino-acids1,2 than are cell membranes3 and also discriminate between their optical isomers2. This has led to the suggestion that the passage of amino-acids through the limiting membranes of chloroplasts is facilitated by carriers2. Polyploidy (whole‐genome duplication) has contributed significantly to angiosperm evolution and diversification. To date, it has been found that most polyploids are the result of multiple formation events, which may contribute to genetic diversity and affect interfertility among polyploid lineages of independent origin. A recently discovered allotetraploid derivative of Mimulus guttatus and M. nasutus, Mimulus sookensis, is found throughout the valleys of western Oregon and Vancouver Island. Here, we analyse the patterns of nucleotide diversity at three chloroplast and six nuclear loci in M. guttatus, M. nasutus and M. sookensis, to gain insight into the formation of M. sookensis. By analysing the patterns of genetic variation seen in the diploid progenitors in comparison with the variation seen in M. sookensis, we are able to show that M. sookensis has recurrently formed. We also observed that most M. sookensis individuals are fixed heterozygotes at all of the nuclear loci examined, suggesting that duplicate gene loss is not extensive in M. sookensis. To assess the possibility that hybridization among M. sookensis has contributed to genetic diversity, we conducted crossing experiments within M. sookensis. We found that M. sookensis of independent origin are highly interfertile, suggesting that crossing barriers do not exist within M. sookensis, and that hybridization among M. sookensis may result in new recombinant genotypes. Together, the data suggest that although recurrent origins may be common, they can contribute to genetic diversity without contributing to reproductive isolation among independently arisen polyploid lineages.  相似文献
