Rabies pseudotyped lentiviral vectors have great potential in gene therapy, not least because of their ability to transduce neurons following their distal axonal application. However, very little is known about the molecular processes that underlie their retrograde transport and cell transduction. Using multiple labeling techniques and confocal microscopy, we demonstrated that pseudotyping with rabies virus envelope glycoprotein (RV-G) enabled the axonal retrograde transport of two distinct subtypes of lentiviral vector in motor neuron cultures. Analysis of this process revealed that these vectors trafficked through Rab5-positive endosomes and accumulated within a non-acidic Rab7 compartment. RV-G pseudotyped vectors were co-transported with both the tetanus neurotoxin-binding fragment and the membrane proteins thought to mediate rabies virus endocytosis (neural cell adhesion molecule, nicotinic acetylcholine receptor, and p75 neurotrophin receptor), thus demonstrating that pseudotyping with RV-G targets lentiviral vectors for transport along the same pathway exploited by several toxins and viruses. Using motor neurons cultured in compartmentalized chambers, we demonstrated that axonal retrograde transport of these vectors was rapid and efficient; however, it was not able to transduce the targeted neurons efficiently, suggesting that impairment in processes occurring after arrival of the viral vector in the soma is responsible for the low transduction efficiency seen in vivo, which suggests a novel area for improvement of gene therapy vectors.  相似文献

Zwitterions, a class of materials that contain covalently bonded cations and anions, have been extensively studied in the past decades owing to their special features, such as excellent solubility in polar solvents, for solution processing and dipole formation for the transfer of carriers and ions. Recently, zwitterions have been developed as electrode modifiers for organic solar cells (OSCs), perovskite solar cells (PVSCs), and organic light‐emitting devices (OLEDs), as well as electrolyte additives for lithium ion batteries (LIBs). With the rapid advances of zwitterionic materials, high‐performance devices have been constructed with enhanced efficiencies by introducing them as interface layers and electrolyte additives. In this review, recent progress in OSCs, PVSCs, OLEDs, and LIBs by using zwitterions is highlighted. The authors also elaborate the role of various zwitterionic materials as interfacial layers and additives for highly efficient OSCs, PVSCs, OLEDs, and LIBs. This article presents an overview of device performance of zwitterionic materials. The structure–property relationship is also discussed. Finally, the prospects of zwitterion materials are also addressed.  相似文献

Summary The morphology and life-cycle of Trichobilharzia arcuata n. sp. from the nasal blood vessels of Dendrocygna arcuata from northern Australia are described. T. arcuata is distinguished from all species of Trichobilharzia except T. australis and African nasal schistosomes by its location in the nasal blood vessels and by the cercariae having 16 flame cells with a formula 2[(3)+(4+1)] against 14 with a formula 2[(3)+(3+1)], or 12 with a formula 2[(3)+(2+1)] in the case of T. corvi. T. arcuata differs from the five African nasal schistosomes in the nature of the tegument, the position of the male genital papilla and the shape of the eggs and from T. australis in the position of the caecal reunion, the number of testes, the absence of spines on the ventral tegument between the ovary and the seminal receptacle, the shape of the eggs and the presence of two germinal masses in the miracidium. Lymnaea lessoni exposed to miracidia of T. arcuata developed patent infection in 22 to 41 days. Intramolluscan development of T. arcuata is similar to T. stagnicolae and T. physellae. Domestic Muscovy ducks and pigeons developed patent infections in 22 and 12 days respectively. Chickens, a black duck and a grey teal did not become infected. ac]19840616  相似文献

1. The Qilian Mountains represent one of the key livestock‐raising grasslands in China. The two main herbivore species raised in this area – yaks and sheep – are of critical economical value. Grasshoppers compete with these animals for available nutrients, creating multifaceted relationships between livestock, grasshoppers and plants. A clear understanding of such relationships is lacking and is urgently needed to guide conservation efforts. 2. This study aims to document the effects of yak and sheep grazing on grasshopper assemblages and to elucidate the underlying mechanisms of such effects. 3. It is shown here that yaks and sheep impact grasshopper assemblages differently. Grasshopper assemblages exhibited lower density, biodiversity, richness, and evenness of distribution in yak‐grazed pastures than in grazing‐free grasslands. Sheep‐grazed pastures exhibited a dramatically divergent picture, with elevated density, biodiversity and richness, and a slightly decreased evenness of distribution. Grasshoppers were generally larger in grazed pastures than in grazing‐free grasslands, especially in yak‐grazed plots. 4. The present study suggests that differences between yak and sheep pastures in plant assemblage structure and plant traits are probably the underlying forces driving the differences in grasshopper assemblage structure and grasshopper traits, respectively. 5. The study shows that the grasshopper habitat indicator species differ between yak and sheep pastures, raising the possibility that such indicators can be used to monitor grassland usage and degradation in the Qilian Mountains. 6. These results provide novel insights into the dynamic interactions of common domesticated herbivore species, grasshoppers and plants in Qilian Mountains, which augment current knowledge and may ultimately lead to better conservation practices.  相似文献

Summary The effects of oxygen tension and dilution rate on expression and stability of killer toxin chimeric plasmid (pADH-10A) in transformants of wine yeast (Montrachet 522) were investigated using a glucose-limited chemostat culture. Killer toxin activity increased as a function of dilution rate when transformants were grown in N2 or air, but not in 100% O2. Killer transformants grown in the presence of N2 had 2–4 fold and 10–15 fold higher toxin expression than that in the presence of air and 100% O2, respectively. Plasmid stability increased as a function of dilution rate, regardless of the oxygen tension used.
