The involvement of platelet glycoprotein (GP) IIb-IIIa complex in calcium channel activity on the plasma membrane was investigated using an electrophysiological approach. Plasma membrane vesicles were prepared from thrombin-stimulated platelets and incorporated into planar lipid bilayers. Voltage-independent Ca2+ channel currents with a conductance of about 10 pS (in 53 mM Ba2+) were observed, in membranes derived from thrombin-stimulated, but not unstimulated platelet membranes. These channel activities were markedly reduced by exposure of membranes to EGTA at 37 degrees C. This reduction was specifically related to the dissociation of the GPIIb-IIIa complex since preincubation of the membranes with a monoclonal antibody to the GPIIb-IIIa complex (AP-2) could protect the channel activities from the effect of EGTA. Thrombasthenic platelets, which lack the GPIIb-IIIa complex, showed impaired channel activities characterized by decreased open probability and lowered conductance states. Furthermore, when platelets were stimulated by thrombin in the presence of EGTA, AP2, or the synthetic peptide RGDS, to prevent fibrinogen binding to the GPIIb-IIIa complex, open probabilities of the channel currents in these membrane vesicles were also decreased. These results suggest that the GPIIb-IIIa complex is involved in platelet Ca2+ channel activation and that ligand binding to the complex during platelet activation may modify the activation of Ca2+ channels.  相似文献

Reactions of a single-cubane cluster [{Rh(cod)}2{MoCl(dtc)}23-S)4] (cod = 1,5-cyclooctadiene, dtc = diethyldithiocarbamate) with 1 equiv. of L (L = pyrazine, 4,4′-bipyridyl, trans-1,2-bis(4-pyridyl)ethylene) in the presence of 2 equiv. of AgBF4 in CH2Cl2 gave doubly bridged double-cubane clusters [({Rh(cod)}2{Mo(dtc)}23-S)4)2(μ-L)2][BF4]4, whose structures were determined by the single-crystal X-ray analysis.  相似文献

Background

The relationships between facial mimicry and subsequent psychological processes remain unclear. We hypothesized that the congruent facial muscle activity would elicit emotional experiences and that the experienced emotion would induce emotion recognition.

Methodology/Principal Findings

To test this hypothesis, we re-analyzed data collected in two previous studies. We recorded facial electromyography (EMG) from the corrugator supercilii and zygomatic major and obtained ratings on scales of valence and arousal for experienced emotions (Study 1) and for experienced and recognized emotions (Study 2) while participants viewed dynamic and static facial expressions of negative and positive emotions. Path analyses showed that the facial EMG activity consistently predicted the valence ratings for the emotions experienced in response to dynamic facial expressions. The experienced valence ratings in turn predicted the recognized valence ratings in Study 2.

Conclusion

These results suggest that facial mimicry influences the sharing and recognition of emotional valence in response to others'' dynamic facial expressions.  相似文献

In recent decades, the extent of Sasa palmata-dominant communities has increased in Sarobetsu Mire in northern Hokkaido, Japan, replacing the original Sphagnum bog vegetation. However, this marked increase in distribution of Sasa in the mire has not been formally documented or investigated in detail. Using aerial photo-interpretation, the present study updated the distribution maps of Sasa communities, showing the changes that have occurred to these communities between 1977 and 2003. The results revealed that the extent of Sasa communities has increased by 15.8 % from 6.60 km2 in 1977 to 7.64 km2 in 2003. The most marked increase occurred on the ground associated with drainage channels, although the oldest channels were constructed more than half a century ago, suggesting that some responses to the drainage of peat bog ecosystems may take a considerable period of time before becoming particularly evident.  相似文献

Freezing of gait in patients with Parkinson’s disease is associated with several factors, including interlimb incoordination and impaired gait cycle regulation. Gait analysis in patients with Parkinson’s disease is confounded by parkinsonian symptoms such as rigidity. To understand the mechanisms underlying freezing of gait, we compared gait patterns during straight walking between 9 patients with freezing of gait but little to no parkinsonism (freezing patients) and 11 patients with Parkinson’s disease (non-freezing patients). Wireless sensors were used to detect foot contact and toe-off events, and the step phase of each foot contact was calculated by defining one stride cycle of the other leg as 360°. Phase-resetting analysis was performed, whereby the relation between the step phase of one leg and the subsequent phase change in the following step of the other leg was quantified using regression analysis. A small slope of the regression line indicates a forceful correction (phase reset) at every step of the deviation of step phase from the equilibrium phase, usually at around 180°. The slope of this relation was smaller in freezing patients than in non-freezing patients, but the slope exhibited larger step-to-step variability. This indicates that freezing patients executed a forceful but noisy correction of the deviation of step phase, whereas non-freezing patients made a gradual correction of the deviation. Moreover, freezing patients tended to show more variable step phase and stride time than non-freezing patients. Dynamics of a model of two coupled oscillators interacting through a phase resetting mechanism were examined, and indicated that the deterioration of phase reset by noise provoked variability in step phase and stride time. That is, interlimb coordination can affect regulation of the gait cycle. These results suggest that noisy interlimb coordination, which probably caused forceful corrections of step phase deviation, can be a cause of freezing of gait.  相似文献

Abstract

This article discusses the production of new “postgenomic” knowledges that aim to be more ecological and “wholistic” than the reductionist genetics of the last forty years. It examines systems biology and, briefly, developmental systems theory, which are two approaches that attempt to model complexities in biology. System biological metaphors and languages have been in part taken from engineering models of automobiles, airplanes and robots and then applied to complex living systems. Systems biology is only the most recent example and perhaps an excellent case in which to study this movement back and forth across the machine-living organism border in Euro-American biology to track how what we know to be nature and machine is constituted. This article argues for a careful analysis of this historical production specifically around the question of what is lost in translation at these border crossings and their potential consequences.  相似文献
