Summary Transepithelial electrogenic Na+ transport (INa) was investigated in the coprodeum of 20-days-old chicken embryos in Ussing chambers. Short circuit current (Isc) and transepithelial resistance (Rt) were 14.7±4.8 A · cm-2 (n=12) and 0.53±0.09 k · cm-2 (n=12), respectively. INa was calculated from changes in Isc by substitution of mucosal Na+ by (N-methyl-d-glucamine) (NMDG). Isc inversed during Na+ removal, and INa was found to be 27.8±4.7 A · cm-2 (n=12). Amiloride (100 mol · l-1) inhibited only about 60% of INa. Analysis of Isc fluctuations revealed a Lorentzian component in the power density spectrum with a corner frequency of about 57 Hz. This component was not correlated to INa, and its origin is still unclear. Removal of mucosal Ca2+ increased INa about 2.5-fold due to an increase of the amiloride-insensitive component of INa in additionally investigated adult tissues. The results clearly show that this is due to a non-selective cation channel with an apparent order of selectivity Cs+>Na+=K+>Rb+>Li+. The Ca2+ concentration required to block 50% of the Isc was about 18 mol · l-1. The I sc Ca could also be supressed by other divalent cations such as Mg2+ and Ba2+. Additionally, an INa-linked Lorentzian component occurred which dominated the control spectrum with a significantly higher corner frequency (about 88 Hz). The results indicate that Na+ absorption in the coprodeum of the chicken embryo is more complex than in adult hens. However, the Ca2+ sensitivity of INa is similar to comparable effects described for other epithelia. The Neuroplastins Np65 and Np55 are neuronal and synapse‐enriched immunoglobulin superfamily molecules that play important roles in a number of key neuronal and synaptic functions including, for Np65, cell adhesion. In this review we focus on the physiological roles of the Neuroplastins in promoting neurite outgrowth, regulating the structure and function of both inhibitory and excitatory synapses in brain, and in neuronal and synaptic plasticity. We discuss the underlying molecular and cellular mechanisms by which the Neuroplastins exert their physiological effects and how these are dependent upon the structural features of Np65 and Np55, which enable them to bind to a diverse range of protein partners. In turn this enables the Neuroplastins to interact with a number of key neuronal signalling cascades. These include: binding to and activation of the fibroblast growth factor receptor; Np65 trans‐homophilic binding leading to activation of p38 MAPK and internalization of glutamate (GluR1) receptor subunits; acting as accessory proteins for monocarboxylate transporters, thus affecting neuronal energy supply, and binding to GABAA α1, 2 and 5 subunits, thus regulating the composition and localization of GABAA receptors. An emerging theme is the role of the Neuroplastins in regulating the trafficking and subcellular localization of specific binding partners. We also discuss the involvement of Neuroplastins in a number of pathophysiological conditions, including ischaemia, schizophrenia and breast cancer and the role of a single nucleotide polymorphism in the human Neuroplastin (NPTN) gene locus in impairment of cortical development and cognitive functions.

We develop a method to predict and validate gene models using PacBio single-molecule, real-time (SMRT) cDNA reads. Ninety-eight percent of full-insert SMRT reads span complete open reading frames. Gene model validation using SMRT reads is developed as automated process. Optimized training and prediction settings and mRNA-seq noise reduction of assisting Illumina reads results in increased gene prediction sensitivity and precision. Additionally, we present an improved gene set for sugar beet (Beta vulgaris) and the first genome-wide gene set for spinach (Spinacia oleracea). The workflow and guidelines are a valuable resource to obtain comprehensive gene sets for newly sequenced genomes of non-model eukaryotes.

A 1330 base-pair fragment of a 16S rRNA gene has been amplified, cloned and sequenced. Comparison to other 16S rRNA sequences of eubacteria showed that P. niger represents a deep branch within the subdivision "Gram-positive with Gram-negative cell walls". It is not related to peptostreptococci, representatives of this genus studied so far are more closely related to clostridia.  相似文献

Neutrophils represent the major fraction of circulating immune cells and are rapidly recruited to sites of infection and inflammation. The inflammasome is a multiprotein complex that regulates the generation of IL-1 family proteins. The precise subcellular localization and functionality of the inflammasome in human neutrophils are poorly defined. Here we demonstrate that highly purified human neutrophils express key components of the NOD-like receptor family, pyrin domain containing 3 (NLRP3), and absent in melanoma 2 (AIM2) inflammasomes, particularly apoptosis-associated speck-like protein containing a CARD (ASC), AIM2, and caspase-1. Subcellular fractionation and microscopic analyses further showed that inflammasome components were localized in the cytoplasm and also noncanonically in secretory vesicle and tertiary granule compartments. Whereas IL-1β and IL-18 were expressed at the mRNA level and released as protein, highly purified neutrophils neither expressed nor released IL-1α at baseline or upon stimulation. Upon inflammasome activation, highly purified neutrophils released substantially lower levels of IL-1β protein compared with partially purified neutrophils. Serine proteases and caspases were differentially involved in IL-1β release, depending on the stimulus. Spontaneous activation of the NLRP3 inflammasome in neutrophils in vivo affected IL-1β, but not IL-18 release. In summary, these studies show that human neutrophils express key components of the inflammasome machinery in distinct intracellular compartments and release IL-1β and IL-18, but not IL-1α or IL-33 protein. Targeting the neutrophil inflammasome may represent a future therapeutic strategy to modulate neutrophilic inflammatory diseases, such as cystic fibrosis, rheumatoid arthritis, or sepsis.  相似文献

In the flowers ofApios americana the vexillum remains ± in its bud position. In this way, a dark cavity is formed displaying a bright window at its base. Insect visitors are attracted to the hidden inflorescences by scent. In their attempt to reach the window they trigger an explosive release of stigma and pollen. Stigmatic fluid glues pollen to the visitor. Zusammenfassung Brandentenküken verfügen über drei verschiedene Taucharten, die sich nach Ausführung und Tauchdauer deutlich unterscheiden lassen. Während das Nahrungstauchen in der 6. Lebenswoche eingestellt wird, bleiben Tauchbaden und Fluchttauchen lebenslang erhalten. Neben dem Nahrungstauchen werden weitere Techniken des Nahrungserwerbs in Abhängigkeit vom Nahrungsangebot und dem Entwicklungsstand unterschiedlich häufig angewendet. Der zeitliche Aufwand für die Nahrungsaufnahme ist in den ersten 2 Lebenswochen maximal. Anschließend nimmt er, bei schneller als bei kontinuierlich ab. Kälteperioden führen zu einer weiteren Reduzierung. Die Schlafhäufigkeit sinkt nach einer anfänglichen Zunahme (bis 10. Lebenswoche) ab. Kälteperioden bewirken eine Zunahme. Unter 3 verschiedenen Schlafhaltungen wird bis zur 12. Lebenswoche das Schlafen im Liegen bevorzugt. Danach nimmt es zugunsten des einbeinigen Schlafens im Stehen, das erstmals in der 7. Lebenswoche auftritt, ab. Kälteperioden stellen die ursprünglichen Häufigkeitsverhältnisse zwischen beiden Schlafhaltungen wieder her. Brandentenküken bilden in den ersten Lebenswochen Schlafhaufen. Ihre Häufigkeit sinkt bis zur 12. Lebenswoche kontinuierlich auf Null ab.
Ontogenetic changes in sleeping, diving and foraging behaviour in Shelduck (Tadorna tadorna) ducklings
Summary Shelduck ducklings show different types of diving behaviour: diving for food, flight-diving and dashing and diving in comfort situations. These types differ by diving duration and the behaviour of the ducks. In diving for food diving duration and frequency decrease during the first 6 weeks. After this, no food diving was observed. Other feeding methods include sieving, upending, pecking on the surface of water, feeding on plants and foot-trampling. Except for feeding on plants, which was first observed in the 10th week, all feeding methods are present in one day old ducklings. The share of foraging time is highest during the first 2 weeks of life. Afterwards it decreases (in females faster than in males) from about 50 to 20 %. During cold weather, it was still lower whereas resting and sleeping behaviour increased. After an increase during the first 10 weeks of life, the share of resting decreases to 25–40 % of observation time. Shelduck ducklings have 3 different sleeping postures: lying, standing on one leg, standing on two legs. During the first weeks of life ducklings only sleep standing on two legs or lying on the ground, the latter is being clearly preferred. Standing on one leg appeared in the 7th week. Its frequency increased and it became the most prominent resting pattern. Only during cold spells shelducks do return to sleep by lying on the ground. Under such conditions also sleeping in contact with one another reoccurs, which is typical for ducklings during their first weeks of life.

The fat-storing endosperm of Ricinus communis L. was found tocontain an ascorbate peroxidase (EC 1.11.1.11 [EC] ), which is nearlyas active as catalase (EC 1.11.1.6 [EC] ) in degradation of hydrogenperoxide (H2O2) at its physiological concentrations. This ascorbateperoxidase probably functions together with monodehydroascorbatereductase (EC 1.6.5.4 [EC] ) or dehydroascorbate reductase (EC 1.8.5.1 [EC] )and glutathione reductase (EC 1.6.4.2 [EC] ) to remove the H2O2 producedduring the transformation of fat to carbohydrate in the glyoxysomes.The activities of these enzymes as well as the content of ascorbateand glutathione increase parallel to the activities of glyoxysomalmarker enzymes during the course of germination. Inhibitionof catalase by aminotriazole results in increases of the ascorbateperoxidase activity and of the glutathione content. All fourenzymes are predominantly localized in the cytosol of the Ricinusendosperm with low activities found in the plastids and themitochondria. The results suggest, that the ascorbate-dependentH2O2 scavenging pathway, which has been shown to be responsiblefor the reduction of photosynthetically derived H2O2 in thechloroplasts, operates also in the Ricinus endosperm.
