Fed and 3-day fasted inland (average mass: 6.97 g) and anadromous (average mass: 6.54 g) striped bass Morone saxatilis fingerlings were held in dipnets above water for 5 min in groups of six. Severity of the response to this handling was measured by whole-body glucose, glycogen, and lactic acid in non-handled bass (considered control level), and then at 30 min, 1, 6, 12, 24 and 48 h recovery. At resting levels, both fed and fasted inland bass showed significantly higher concentrations of the whole body variables than anadromous bass. All four groups of bass showed an increase in lactic acid and glucose immediately after handling, with a concomitant decrease in glycogen. Peak levels of glucose and lactic acid were similar in the four groups. Fasting did not have an effect on the glucose and lactic acid responses, but did affect the glycogen response. The two fasted groups did not return to control glycogen concentrations during the 48-h recovery period. By 48 h, both glucose and lactic acid had returned to control levels. It is concluded that inland and anadromous strains of fingerling striped bass do not differ in their sensitivity to an acute handling stress. Recovery of glycogen energy stores following handling is much better if fish are not fasted before handling.  相似文献

Background  

Populations of the Oriental White-backed Vulture (Gyps bengalensis) have declined by over 95% within the past decade. This decline is largely due to incidental consumption of the non-steroidal anti-inflammatory veterinary pharmaceutical diclofenac, commonly used to treat domestic livestock. The conservation status of other Gyps vultures in southern Asia is also of immediate concern, given the lack of knowledge regarding status of their populations and the continuing existence of taxonomic uncertainties. In this study, we assess phylogenetic relationships for all recognized species and the majority of subspecies within the genus Gyps. The continuing veterinary use of diclofenac is an unknown but potential risk to related species with similar feeding habits to Gyps bengalensis. Therefore, an accurate assessment of the phylogenetic relationships among Gyps vultures should aid in their conservation by clarifying taxonomic uncertainties, and enabling inference of their respective relatedness to susceptible G. bengalensis.  相似文献

We have used the binding of liposomes conjugated with antifluorescein antibody specific for fluorescein isothiocyanate-modified erythrocytes as a model for multivalent antigen-antibody interactions. We examined a series of liposome preparations which were conjugated to between 0 and 332 active antibodies per liposome. The antigen binding capacity and mean intrinsic affinity of the soluble and conjugated antibody were determined by fluorescence quenching of carboxyfluorescein. Liposome-cell interaction data were fitted with a Scatchard-type equation. Functional affinity of liposomes for cells was up to 1000-fold greater than the intrinsic affinity of the antibody for soluble ligand. Analysis for binding at high cell concentrations revealed that liposome-induced cell agglutination reduces the number of available binding sites per cell.  相似文献

Activities of Escherichia coli DNA polymerase-I were examined in the presence of the anti-tumor drug cis-diaminedichloroplatinum(II) and its inactive geometric isomer trans-diaminedichloroplatinum(II). The trans-isomer did not inhibit the enzyme activity. The anti-tumor drug, on the other hand, retarded the enzyme in its ability to extend the primer strand of DNA. Two alternative mechanisms of inhibition, covalent binding of cis-diaminedichloroplatinum(II) to the polymerase and to the template DNA, were explored. Selective preincubations of the platinum drug with the polymerase and DNA reveal that the inhibition is primarily due to covalent binding to the enzyme. The rates of inhibition were found to be first order in enzyme and zeroth order in platinum in the concentration range 0.05-3.0 mM. A mechanism that deals with the formation of an initial platinum-polymerase-I complex with a binding constant > 10(5) M(-1) followed by a further reaction to form an inhibitory complex is consistent with the kinetic data. The rate limiting first order rate constant for the formation of the inhibitory complex is comparable to that observed for the thiol coordination of peptides containing cysteine residues. Analyses of known structures and functions of catalytic domains of various polymerases point to the direction that the inhibition is perhaps due to the distortion of the DNA binding domain of the enzyme due to platinum coordination.  相似文献

Anthropogenic changes can influence mutualism evolution; however, the genomic regions underpinning mutualism that are most affected by environmental change are generally unknown, even in well-studied model mutualisms like the interaction between legumes and their nitrogen (N)-fixing rhizobia. Such genomic information can shed light on the agents and targets of selection maintaining cooperation in nature. We recently demonstrated that N-fertilization has caused an evolutionary decline in mutualistic partner quality in the rhizobia that form symbiosis with clover. Here, population genomic analyses of N-fertilized versus control rhizobium populations indicate that evolutionary differentiation at a key symbiosis gene region on the symbiotic plasmid (pSym) contributes to partner quality decline. Moreover, patterns of genetic variation at selected loci were consistent with recent positive selection within N-fertilized environments, suggesting that N-rich environments might select for less beneficial rhizobia. By studying the molecular population genomics of a natural bacterial population within a long-term ecological field experiment, we find that: (i) the N environment is indeed a potent selective force mediating mutualism evolution in this symbiosis, (ii) natural variation in rhizobium partner quality is mediated in part by key symbiosis genes on the symbiotic plasmid, and (iii) differentiation at selected genes occurred in the context of otherwise recombining genomes, resembling eukaryotic models of adaptation.  相似文献

The proportion of the primary electron transport acceptor forphotosystem II(Q) in the oxidized state was estimated usingin vivo fluorescence from populations of the marine diatom Thalassiosirapseudonana. Under nutrient replete conditions, the transitionfrom light-dependent to light-independent development duringthe cell cycle was correlated with a 20% decrease in the amountof Q in the oxidized state. As a consequence, mean redox statuswas correlated with the fraction of the population in the pre-commitmentor light-dependent period of the cycle. Comparison with theresults of other investigators suggests that de-coupling ofphotosynthetic units from the electron transport system linkingphotosystems II and I during the committed or light-independentperiod of the cycle, may be the mechanism controlling the redoxstate of Q.  相似文献
