Abundance and length of the highly-targeted snapper Chysophrys auratus were compared between sites in ''no take'' areas (Sanctuary Zones: SZ), partial protected areas which are fished (Habitat Protection Zones: HPZ), and areas outside (Outside) the Solitary Islands Marine Park (SIMP), Australia. Baited Remote Underwater Video (BRUV) sampling on shallow rocky reef (15 - 25 m) was conducted annually from 2002 until 2014 in the Austral-winter, covering the decade after these marine park zones were established (2002). Additional deeper sites (25 - 40 m) were sampled in 2010-2011 to assess if findings were more-broadly applicable. Lengths were measured using stereo-BRUVs from 2011-2014. Snapper were significantly more abundant in SZ overall and in most years compared with the other two management types, which did not significantly differ. Snapper rapidly increased after 2 - 3 years protection in all management types, especially SZ. Snapper were present on more SZ deployments than HPZ and Outside after the same period. The positive SZ response in snapper abundance on shallower reef was also found at a broader spatial scale on deeper sites. Again the two fished management types did not show significant differences among each other. There was considerable variation in snapper abundance between years, with strong peaks in 2005, 2009 and 2014 especially in SZ. Abundances remained higher in SZ in the year or two following a strong peak, but decreased to similar abundances to fished areas before the next peak. Snapper length frequency distribution significantly differed between SZ and both fished management types, with more larger snapper within SZ including a higher proportion (58%) that were legal-sized (>25.7 cm FL). HPZ and Outside did not significantly differ from each other, and were dominated by individuals below legal size. Overall, SZ''s have positively influenced abundance and length of snapper on these subtropical rocky reefs.  相似文献

The removal of 3-methyladenine and 7-methylguanine from nuclear DNA was determined following exposure of Chlamydomonas reinhardi to methyl methanesulfonate (MMS). The amount of 3-methyladenine in DNA was determined using an extract from Micrococcus luteus that has a 3-methyladenine-DNA glycosylase. The amount of 7-methylguanine was estimated by heating the DNA for 30 min at 70° followed by alkaline hydrolysis of the resulting apurinic sites. The molecular weight of the DNA was determined using alkaline sucrose gradients. The 3-methyladenine is removed with a half-life of 2–3 h whereas the 7-methylaguanine is removed with a half-life of 10–12 h. The rate of removal of the 7-methylguanine is more than an order of magnitude faster than the estimated non-enzymatic hydrolysis rate indicating the probability of enzymatic repair. Addition of cycloheximide immediately after MMS treatment inhibits the removal of 3-methyladenine and 7-methylguanine from DNA. If cycloheximide is added 1.5 h after treatment with MMS, there is much less inhibition of the removal of 3-methyladenine. These results are interpreted to mean that MMS induces the synthesis of 1 or more proteins that are required for the repair of 3-methyladenine from Chlamydomonas DNA.  相似文献

Background  

South America's western coastline, extending in a near-straight line across some 35 latitudinal degrees, presents an elegant setting for assessing both contemporary and historic influences on cladogenesis in the marine environment. Southern bull-kelp (Durvillaea antarctica) has a broad distribution along much of the Chilean coast. This species represents an ideal model taxon for studies of coastal marine connectivity and of palaeoclimatic effects, as it grows only on exposed rocky coasts and is absent from beaches and ice-affected shores. We expected that, along the central Chilean coast, D. antarctica would show considerable phylogeographic structure as a consequence of the isolating effects of distance and habitat discontinuities. In contrast, we hypothesised that further south - throughout the region affected by the Patagonian Ice Sheet at the Last Glacial Maximum (LGM) - D. antarctica would show relatively little genetic structure, reflecting postglacial recolonisation.  相似文献

Moment index displacement automatically corrects a number of significant nonrandom instrumental errors in fluorescence time-decay measurements. Three-component data, obtained by measuring the fluorescence decay of three different species mixed in the same solution, were used as a test sample. It was shown, as predicted by theory, that moment index displacement corrects three nonrandom instrumental errors: (1) the presence of scatter in the data; (2) time origin shifts between lamp and fluorescence data; and (3) lamp drift, or time-dependent changes in the shape of the excitation curve. The data clearly show that the use of the method of moments with moment index displacement to analyze fluorescence decay data is not a curve-fitting procedure. This procedure will accurately obtain decay parameters for multiple-exponential decays from certain badly distorted data, yielding a calculated curve very different from the actual data.  相似文献
