The fossil burrow Dolopichnus, capped by an ash layer, was collected ~0.6 m below the Permian–Triassic Boundary at the Lung Cam section in North Vietnam. The burrow occurs at the top of Bed 9, directly below the stepped extinction in foraminifera that marks initiation of the global perturbations related to the end-Permian extinction event. Dolopichnus is a plug-shaped burrow, possessing lining and fills. This specimen was found capped by an undisrupted bentonite layer. The main chamber has internal structure throughout indicative of benthic organisms overcoming living pressures of repeated ash falls, which can be applied to paleoenvironmental research across different regions and times, including the recent.  相似文献

Apolipoprotein E2 (apoE2) and apoE3-Leiden cause chylomicron remnant accumulation (type III hyperlipidemia). However, the degree of dyslipidemia and its penetrance are different in humans and mice. Remnant uptake by isolated liver from apoE-/- mice transgenic for human apoE2, apoE3-Leiden, or apoE3 was measured. In the presence of both LDL receptor (LDLR) and LDL receptor-related protein (LRP), remnant uptake was apoE3>E3-Leiden>E2 mice. Absence of LDLR reduced uptake in apoE3 and apoE3-Leiden-secreting livers but not in apoE2-secreting livers. LRP inhibition with receptor-associated protein reduced uptake in apoE3- and apoE2-secreting livers, but not in apoE3-Leiden-secreting livers, regardless of the presence of LDLR. Fluorescently labeled remnants clustered with LRP in apoE3-secreting livers only in the absence of LDLR, but clustered in livers that expressed apoE2 even in the presence of LDLR, and did not cluster with LRP in livers of apoE3-Leiden even in the absence of LDLR. Remnants were reconstituted with the three human apoE isoforms. Removal by liver of mApoe-/-/mldlr-/- mice expressing the human LDLR was slightly greater than removal in the previous experiments with apoE3>E2> E3-Leiden. Thus, in vivo, human apoE2 is cleared primarily by LRP, apoE3-Leiden is cleared only by the LDLR, and apoE3 is cleared by both.  相似文献

Anoxic sediments from Rotsee (Switzerland) were analyzed for the presence and diversity of methanogens by using molecular tools and for methanogenic activity by using radiotracer techniques, in addition to the measurement of chemical profiles. After PCR-assisted sequence retrieval of the 16S rRNA genes (16S rDNA) from the anoxic sediment of Rotsee, cloning, and sequencing, a phylogenetic analysis identified two clusters of sequences and four separated clones. The sequences in cluster 1 grouped with those of Methanosaeta spp., whereas the sequences in cluster 2 comprised the methanogenic endosymbiont of Plagiopyla nasuta. Discriminative oligonucleotide probes were constructed against both clusters and two of the separated clones. These probes were used subsequently for the analysis of indigenous methanogens in a core of the sediment, in addition to domain-specific probes against members of the domains Bacteria and Archaea and the fluorescent stain 4', 6-diamidino-2-phenylindole (DAPI), by fluorescent in situ hybridization. After DAPI staining, the highest microbial density was obtained in the upper sediment layer; this density decreased with depth from (1.01 +/- 0.25) x 10(10) to (2.62 +/- 0.58) x 10(10) cells per g of sediment (dry weight). This zone corresponded to that of highest metabolic activity, as indicated by the ammonia, alkalinity, and pH profiles, whereas the methane profile was constant. Probes Eub338 and Arch915 detected on average 16 and 6% of the DAPI-stained cells as members of the domains Bacteria and Archaea, respectively. Probe Rotcl1 identified on average 4% of the DAPI-stained cells as Methanosaeta spp., which were present throughout the whole core. In contrast, probe Rotcl2 identified only 0.7% of the DAPI-stained cells as relatives of the methanogenic endosymbiont of P. nasuta, which was present exclusively in the upper 2 cm of the sediment. Probes Rotp13 and Rotp17 did not detect any cells. The spatial distribution of the two methanogenic populations corresponded well to the methane production rates determined by incubation with either [14C]acetate or [14C]bicarbonate. Methanogenesis from acetate accounted for almost all of the total methane production, which concurs with the predominance of acetoclastic Methanosaeta spp. that represented on average 91% of the archaeal population. Significant hydrogenotrophic methanogenesis was found only in the organically enriched upper 2 cm of the sediment, where the probably hydrogenotrophic relatives of the methanogenic endosymbiont of P. nasuta, accounting on average for 7% of the archaeal population, were also detected.  相似文献

In suspension-cultured cells of tomato (Lycopersicon esculentum Mill.), the activity of 1-aminocyclopropane-1-carboxylate synthase (ACC-S) rapidly increases in response to fungal elicitors. The effect of inhibitors of protein kinases and protein phosphatases on the regulation of ACC-S was studied. K-252a, an inhibitor of protein kinases, prevented induction of the enzyme by elicitors and promoted its apparent turnover in elicitor-stimulated cells, causing a 50% loss of activity within 4 to 8 min in both the presence and absence of cycloheximide. Calyculin A, an inhibitor of protein phosphatases, caused a rapid increase of ACC-S in the absence of elicitors and an immediate acceleration of the rate of ACC-S increase in elicitor-stimulated cells. In the presence of cycloheximide there was no such increase, indicating that the effect depended on protein synthesis. Cordycepin, an inhibitor of mRNA synthesis, did not prevent the elicitor-induced increase in ACC-S activity but strongly reduced the K-252a-induced decay and the calyculin A-induced increase of its activity. In vitro, ACC-S activity was not affected by K-252a and calyculin A or by treatments with protein phosphatases. These results suggest that protein phosphorylation/dephosphorylation is involved in the regulation of ACC-S, not by regulating the catalytic activity itself but by controlling the rate of turnover of the enzyme.  相似文献

Zusammenfassung In den Sommern 1955 und 1956 wurden in dem Seevogelschutzgebiet Wangerooge-West 352 ad. Küstenseeschwalben gefangen. Unter Hinzuziehung von 9 Totfunden ergaben sich 100 Wiederfunde von Ex., die in den Jahren 1935 bis 1953 beringt worden waren. Für die als juv. Beringten wurde ein Durchschnittsalter von etwa 12 Jahren ermittelt, während sich für die als ad. Beringten ein solches von x + 6,5 ergab.Ein Vergleich mit den Flußseeschwalben der Cape Cod-Kolonien zeigt, daß die Wangerooger Küstenseeschwalben ein erheblich höheres Durchschnittsalter aufweisen, andererseits aber auch eine geringere Nachwuchsquote erzielen. Dagegen ergeben sich zum Austernfischer nach Feststellungen auf Mellum weitgehende Übereinstimmungen.  相似文献
