Local Conformation and Dynamics of Isoleucine in the Collagenase Cleavage Site Provide a Recognition Signal for Matrix Metalloproteinases

The mechanism by which enzymes recognize the “uniform” collagen triple helix is not well understood. Matrix metalloproteinases (MMPs) cleave collagen after the Gly residue of the triplet sequence Gly∼[Ile/Leu]-[Ala/Leu] at a single, unique, position along the peptide chain. Sequence analysis of types I-III collagen has revealed a 5-triplet sequence pattern in which the natural cleavage triplets are always flanked by a specific distribution of imino acids. NMR and MMP kinetic studies of a series of homotrimer peptides that model type III collagen have been performed to correlate conformation and dynamics at, and near, the cleavage site to collagenolytic activity. A peptide that models the natural cleavage site is significantly more active than a peptide that models a potential but non-cleavable site just 2-triplets away and NMR studies show clearly that the Ile in the leading chain of the cleavage peptide is more exposed to solvent and less locally stable than the Ile in the middle and lagging chains. We propose that the unique local instability of Ile at the cleavage site in part arises from the placement of the conserved Pro at the P₃ subsite. NMR studies of peptides with Pro substitutions indicate that the local dynamics of the three chains are directly modulated by their proximity to Pro. Correlation of peptide activity to NMR data shows that a single locally unstable chain at the cleavage site, rather than two or three labile chains, is more favorable for cleavage by MMP-1 and may be the determining factor for collagen recognition.     

How Multidisciplinary Are the Multidisciplinary Journals Science and Nature?

Interest in cross-disciplinary research knowledge interchange runs high. Review processes at funding agencies, such as the U.S. National Science Foundation, consider plans to disseminate research across disciplinary bounds. Publication in the leading multidisciplinary journals, Nature and Science, may signify the epitome of successful interdisciplinary integration of research knowledge and cross-disciplinary dissemination of findings. But how interdisciplinary are they? The journals are multidisciplinary, but do the individual articles themselves draw upon multiple fields of knowledge and does their influence span disciplines? This research compares articles in three fields (Cell Biology, Physical Chemistry, and Cognitive Science) published in a leading disciplinary journal in each field to those published in Nature and Science. We find comparable degrees of interdisciplinary integration and only modest differences in cross-disciplinary diffusion. That said, though the rate of out-of-field diffusion might be comparable, the sheer reach of Nature and Science, indicated by their potent Journal Impact Factors, means that the diffusion of knowledge therein can far exceed that of leading disciplinary journals in some fields (such as Physical Chemistry and Cognitive Science in our samples).     

Introgressive hybridization of tilapias in Zimbabwe

Three species of tilapia native to Zimbabwe, Oreochromis mossambicus, O. mortimeri and O. macrochir are not naturally sympatric, but because of transplantation they have been brought into sympatry. Allozymes of fish examined from Lakes Kariba, Kyle and Chivero showed evidence of introgressive hybridization. The data showed non-random mating populations in Lakes Kariba and Kyle. All three reservoirs displayed individuals with intermediate principal component scores and hybrid index scores indicating hybridization.     

The production of floral oils byMonttea (Scrophulariaceae) and the function of tarsal pads inCentris bees

Plant species that secrete oil as their primary floral reward are rare and sporadically found in the angiosperms. We report here thatMonttea, a genus previously unsuspected of being an oil-plant, produces lipids from trichome elaiophores on the inside of the lower (anterior) lip. The discovery of the production of oils by species of this S. American genus explains the occurrence of unusual dual-function collecting structures in ArgentineCentris (Hymenoptera: Anthophoridae) and explains the presence of oil-collecting bees in regions where oil-secreting flowers were previously thought to be absent. The behavior of these centridine pollinators onMonttea flowers parallels that of oil-collecting bees onDiascia (Scrophulariaceae) in S. Africa.     

Secretion of the sweet-tasting plant protein thaumatin byBacillus subtilis

Summary With the -amylase promoter and ribosome binding site,Bacillis subtilis was used to express the sweet plant protein thaumatin II cDNA fused in the correct reading frame to the -amylase leader peptide. The r-thaumatin was purified from the medium on a S-Sepharose column and detected with western blots by sheep -thaumatin antibodies. The r-thaumatin and authentic thaumatin were the same size when reduced by 2-ME and the same size when not reduced.     

From the population to society: The cooperative metaphors of W.C. Allee and A.E. Emerson

Conclusion John Greene has dismissed the evolutionary ethics of Simpson as a case in which science was only a tool, a weapon, in defense of positions that were essentially religious and philosophical.⁵⁷ This position adopts an amorphous view of science, in which a scientific theory can be construed to support practically any rhetorical position. The relationship between theory and rhetoric, however, is more complex; it is interactive, with the theory and the rhetoric influencing and supporting one another. It is no coincidence that Allee, Emerson, and Simpson all arrived at a biological basis of democracy and a naturalistic ethics during a period when the future of world politics and man's own morality were in question. Allee's commitment to world peace certainly antedated his theory of sociality, just as Simpson's commitment to democracy undoubtedly preceded his evolutionary views.By adopting a particular theory, however, the biologist necessarily imposes constraints on the corresponding rhetoric. Allee and Emerson could not, for example, have stressed the importance of the individual in democracy, given the orientation and framework of their biological research. There were differences among the social philosophies of Allee, Emerson, and Simpson; these differences depended, in part, on the specific evolutionary metaphors to which they subscribed. Allee's ideas with respect to cooperation were distinct from Emerson's, and both men differed strongly from Simpson on the role of the individual in evolution. The Chicago school was united by a conceptual framework that emphasized the population as the unit of selection, and the importance of cooperation in nature. But cooperation could play many roles: as a unifying principle for a theory of sociality, as an integrating mechanism in physiological functionalism, and as a biological source of hope for a society in the grips of a world war.     

The response of an established line of rat liver cells to thyroid hormone

The response of an established line of non-transformed adult rat liver epithelial cells (ARL 15) to thyroid hormone (T3) (3,5,3'-triiodothyronine) was characterized. Exposure of confluent monolayers to 1.10(-8) M T3 for 3 days increased O2 consumption (QO2) between 14-58%, ouabain-sensitive Rb+ uptake 26%, (Na+ + K+)-ATPase activity 32%, alpha-glycerophosphate dehydrogenase activity 103% and cytochrome oxidase activity 208%. The ARL 15 cells, maintained in continuous culture, therefore, exhibit the hallmarks of an authentic physiological response to thyroid hormone.
