Seed germination traits of Ailanthus altissima,Phytolacca americana and Robinia pseudoacacia in response to different thermal and light requirements

Invasion of alien plant species (IAS) represents a serious environmental problem, particularly in Europe, where it mainly pertains to urban areas. Seed germination traits contribute to clarification of invasion dynamics. The objective of this research was to analyze how different light conditions (i.e., 12-hr light/12-hr darkness and continuous darkness) and temperature regimes (i.e., 15/6°C, 20/10°C and 30/20°C) trigger seed germination of Ailanthus altissima (AA), Phytolacca americana (PA) and Robinia pseudoacacia (RP). The relationship between seed germination and seed morphometric traits was also analyzed. Our findings highlight that temperature rather than light was the main environmental factor affecting germination. RP germinated at all tested temperatures, whereas at 15/6°C seeds of AA and PA showed physiological dormancy. RP had a higher germination capacity at a lower temperature, unlike AA and PA, which performed better at the highest temperatures. Light had a minor role in seed germination of the three species. Light promoted germination only for seeds of PA, and final germination percentage was 1.5-fold higher in light than in continuous darkness. Seed morphometric traits (thickness [T], area [A] and volume [V]) had a significant role in explaining germination trait variations. The results highlight the importance of increasing our knowledge on seed germination requirements to predict future invasiveness trends. The increase in global temperature could further advantage AA and PA in terms of germinated seeds, as well as RP by enhancing the germination velocity, therefore compensating for a lower germination percentage of this species at the highest temperatures.     

Differences between ranamargarin and other tachykinins in the stimulation of ion transport in frog skin

In frog skin, tachykinins stimulate ion transport by interaction with NK1-like receptors. The structural requirements of the peptide are the presence of the C-terminal sequence Phe-X-Gly-Leu-Met-NH₂ and at least one Pro residue in the N-terminal sequence. In this paper, we demonstrate that the C-terminal amino acid must be amidated but it can be different from Met, and that the sequence cannot be longer or shorter than 11–12 amino acids. Unexpectedly, Ranamargarin (14 amino acids, no Pro residue) increased the short circuit current value by 48 ± 0.3%. On the basis of considerable experimental evidence, we suggest that Ranamargarin interacts with a receptor different from those of other tachykinins.     

The ubiquitin C‐terminal hydrolase UCH‐L1 promotes bacterial invasion by altering the dynamics of the actin cytoskeleton

Invasion of eukaryotic target cells by pathogenic bacteria requires extensive remodelling of the membrane and actin cytoskeleton. Here we show that the remodelling process is regulated by the ubiquitin C‐terminal hydrolase UCH‐L1 that promotes the invasion of epithelial cells by Listeria monocytogenes and Salmonella enterica. Knockdown of UCH‐L1 reduced the uptake of both bacteria, while expression of the catalytically active enzyme promoted efficient internalization in the UCH‐L1‐negative HeLa cell line. The entry of L. monocytogenes involves binding to the receptor tyrosine kinase Met, which leads to receptor phosphorylation and ubiquitination. UCH‐L1 controls the early membrane‐associated events of this triggering cascade since knockdown was associated with altered phosphorylation of the c‐cbl docking site on Tyr1003, reduced ubiquitination of the receptor and altered activation of downstream ERK1/2‐ and AKT‐dependent signalling in response to the natural ligand Hepatocyte Growth Factor (HGF). The regulation of cytoskeleton dynamics was further confirmed by the induction of actin stress fibres in HeLa expressing the active enzyme but not the catalytic mutant UCH‐L1_C90S. These findings highlight a previously unrecognized involvement of the ubiquitin cycle in bacterial entry. UCH‐L1 is highly expressed in malignant cells that may therefore be particularly susceptible to invasion by bacteria‐based drug delivery systems.     

Fluctuations of grass pollen content in the atmosphere of East Perugia and meteorological correlations (year 1989)

Summary Gramineae pollination from a pollen monitoring station located in the eastern suburb of Perugia and meteorological correlations are reported. The data refers to the year 1989. Grass pollen peak pollination was from May to July; in this period the influence of relative humidity and of temperature on pollen concentration was very high. Phenological observations, to identify the time of maximum stamen extension in the most common genera in the area, are also reported. During the period of investigation the counts of pollen grains over four-hour periods showed a regular diurnal rhythm with peaks of concentration in the four-hour period 16.00–20.00. Aerosporological data and meteorological data related to four-hour periods were correlated following different criteria.     

Manipulation of Tissues for Improved Aceto-Carmine Staining

The importance of the study of the structural-mechanic properties of any particular material in order to improve the technic for aceto-carmine smears and to obtain better preparations of that material has not been, perhaps, sufficiently emphasized in the large number of papers on such cytological technic. The usefulness of such a study will be shown here in two cases met by the writer.
