The use of carboplatin in cancer chemotherapy is limited by the emergence of drug resistance. To understand the molecular basis for this resistance, a chemogenomic screen was performed in 53 yeast mutants that had previously presented strong sensitivity to this widely used anticancer agent. Thirty-four mutants were responsive to carboplatin, and from these, 21 genes were selected for further studies because they have human homologues. Sixty percent of these yeast genes possessed human homologues which encoded proteins that interact with cullin scaffolds of ubiquitin ligases, or whose mRNA are under the regulation of Human antigen R (HuR) protein. Both HuR and cullin proteins are regulated through NEDDylation post-translational modification, and so our results indicate that inhibition of this process should sensitise resistant tumour cells to carboplatin. We showed that treatment of a tumour cell line with MLN4924, a NEDDylation inhibitor, overcame the resistance to carboplatin. Our data suggest that inhibition of NEDDylation may be a useful strategy to resensitise tumour cells in patients that have acquired carboplatin resistance.  相似文献

After total hepatectomy in the rat, the presence of conjugated bilirubin in the plasma has been demonstrated using an extraction technique. This is particularly striking after loading the animals with unconjugated bilirubin. Arachnids of the order Opiliones (harvestmen), which includes around 6000 specìes, have a pair of scent glands that open at the sides of the body, producing substances used as defence. Several types of behavioural, morphological and chemical defensive mechanisms have been identified in the order as a whole, although some of these tactics were restricted to particular groups. Only around 60 species have been studied from this perspective so far, more than half of which belong to the largest harvestman family within the order Laniatores, the Gonyleptidae, and have only recently been studied in an evolutionary perspective, showing the usefulness of defensive characters in taxonomy and evolutionary biology. Within Laniatores, the Grassatores clade includes the Gonyleptidae and 20 additional families, mostly poorly or not previously studied. We describe the morphology of the structures involved in fluid displacement during chemical defence in 15 of these families (data on two additional families are available from the literature) and discuss the evolution of such traits based on an available phylogenetic hypothesis of relationships within Grassatores, using the representatives of Triaenonychidae (a non‐Grassatores family of Laniatores) for comparison. We conclude that most non‐gonyleptoid Grassatores share (maybe plesiomorphically) a series of characteristics, mostly strongly different from what is observed within the gonyleptoids, and that smaller groups seem to share diagnostic features related to chemical defence, as is the case of stygnids, cosmetids and triaenonychines, and especially of manaosbiids and cranaids, whose defensive morphologies largely resemble those of derived gonyleptids. The following main synapomorphies were detected: (a) Grassatores: the presence of a deep and well‐defined descending channel; (b) Samooidea+Zalmoxoidea+Assamioidea+Gonyleptoidea: lateral pegs along the lateral channel; (c) Samooidea+Zalmoxoidea: deep channels forming an H on the dorsal scute; (d) Gonyleptoidea: ozopore cutting dorsally (reversing in Agoristenidae and Stygnidae to a laterally placed oval ozopore), a wide and smooth lateral channel, reversing to a lateral channel whose bottom is covered with either small plates (Agoristenidae) or high tubercles (Stygnidae), and apophyses of coxa II close to or covering the ozopore.  相似文献

A bee colony is often compared to a multicellular organism, mainly because of its spatial organization. We propose that a temporal organization of equal importance is also present. To support this view, we studied the reproductive processes of two closely related species of stingless bees. Stingless bees enable observations of daily rhythms that are performed by distinct social classes. The emergent process, POP, is cyclic and consists of the building and provisioning of brood cells by the worker bees and egg‐laying by the queen. Colonies were kept in the laboratory under constant conditions with the exit tube opening to the environment; thus, foragers had direct access to environmental cycles. At a later stage of the experiment, the exit tube was closed by a sieve; in this case, bees had their own stock of food, but the environmental LD cycle could still be detected when they were inside the exit tube. Daily POP rhythms were present and showed distinct temporal patterns in each species. A third condition was imposed on one of the species only: the exit tube was closed by a sieve and maintained inside a box that was provided with constant illumination. In this colony, the POP rhythm was perturbed by the destruction of the brood cells. Restoration of POP consisted of a rapid reconstruction of cells followed by a late oviposition in the same day. As different rhythmic patterns were detected, but showed regular timings with respect to one another, an interpretation based upon the concept of an internal temporal order is suggested.  相似文献

Subterranean rodents spend most of the day inside underground tunnels, where there is little daily change in environmental variables. Our observations of tuco-tucos (Ctenomys aff. knighti) in a field enclosure indicated that these animals perceive the aboveground light-dark cycle by several bouts of light-exposure at irregular times during the light hours of the day. To assess whether such light-dark pattern acts as an entraining agent of the circadian clock, we first constructed in laboratory the Phase Response Curve for 1 h light-pulses (1000lux). Its shape is qualitatively similar to other curves reported in the literature and to our knowledge it is the first Phase Response Curve of a subterranean rodent. Computer simulations were performed with a non-linear limit-cycle oscillator subjected to a simple model of the light regimen experienced by tuco-tucos. Results showed that synchronization is achieved even by a simple regimen of a single daily light pulse scattered uniformly along the light hours of the day. Natural entrainment studies benefit from integrated laboratory, field and computational approaches.  相似文献

This study investigated the exposure of jaguar populations and domestic animals to smooth Brucella, Leptospira spp. and Toxoplasma gondii in the Cerrado, Pantanal and Amazon biomes of Brazil. Between February 2000 and January 2010, serum samples from 31 jaguars (Panthera onca), 1,245 cattle (Bos taurus), 168 domestic dogs (Canis lupus familiaris) and 29 domestic cats (Felis catus) were collected and analysed by rose bengal test for smooth Brucella, microscopic agglutination test for Leptospira spp. and modified agglutination test for T. gondii. Cattle populations from all sites (9.88%) were exposed to smooth Brucella, but only one jaguar from Cerrado was exposed to this agent. Jaguars captured in the Cerrado (60.0%) and in the Pantanal (45.5%) were seropositive for different serovars of Leptospira spp., cattle (72.18%) and domestic dogs (13.1%) from the three sites and one domestic cat from Pantanal were also seropositive for the agent. The most prevalent serotype of Leptospira spp. identified in jaguars from the Cerrado (Grippotyphosa) and the Pantanal (Pomona) biomes were distinct from those found in the domestic animals sampled. Jaguars (100%), domestic dogs (38.28%) and domestic cats (82.76%) from the three areas were exposed to T. gondii. Our results show that brucellosis and leptospirosis could have been transmitted to jaguars by domestic animals; and jaguars probably play an important role in the maintenance of T. gondii in nature.  相似文献

The effect on tissue differentiation and growth in vitro of certain of the factors implicated in collagen synthesis (ascorbic acid, α-ketoglutarate and oxygen) and the influence of hydrocortisone was studied using organ cultures of fetal mouse mandible as a mixed epithelial and connective tissue system. Using serum-free Waymouth’s MB 752/1 chemically-defined medium, addition of high levels of ascorbic acid (300 μg per ml), hydrocortisone (1 μg per ml) and oxygen (95%) enhanced differentiation in a number of tissues, in particular skin and appendages, tooth germs and bone, while osteoid and dentine production were noticeably promoted. It is suggested that an essential aspect of media design for organ culture involves the incorporation of collagen-promoting factors to the in vitro environment particularly with regard to the controlling role implicated for collagen in a variety of biological processes. Endemic and endangered species are highly vulnerable to habitat perturbations and may be subject to variations in their population size. Management plan for these species is crucial to avoid population decline, loss of genetic variability, inbreeding and ultimately extinction. The sand lizard, Liolaemus lutzae, is endemic to a habitat of sandy coastal plain (restinga). Its geographical distribution extends for only 200 km stretch of the coast of Rio de Janeiro state, Brazil, one of South America’s most densely populated regions. Extensive development and degradation of the beaches where the species inhabits, have led to the species becoming critically endangered. We used mitochondrial DNA sequences and microsatellite loci to resolve patterns of population connectivity and genetic variation within the species in order to provide a platform for a species management plan. Our results indicate the existence of three main populations, separated from each other by the Guanabara Bay and by the Arraial do Cabo Peninsula. The low microsatellite genetic variation and heterozygosity witnessed in each of the three populations, together with high levels of inbreeding and low effective population sizes suggest that the species is in urgent need of intensive management. Based on the results of this study we propose strong measures to protect existing restinga fragments and the implementation of programmes of captive breeding and reintroduction of individuals from the heavily threatened regions to protected refugia. Such measures may be the only way of ensuring the continuity of the species.  相似文献
