A new purification procedure for isocitrate lyase from Pinus pinea is reported. The final preparation shows charge homogeneity and a purity degree higher than 95%. It is possible to remove catalase completely by exploiting the high hydrophobicity of isocitrate lyase. The enzyme has a Mr of 264,000 and is likely composed of four subunits, each with a Mr of 66,000. The binding of radioactively labeled oxalate revealed four catalytic sites per oligomer. These data suggest that isocitrate lyase subunits are similar, if not identical. The Michaelis constant for isocitrate is equal to 33 microM; molecular activity is about 2670 mol X min-1 X mol of enzyme-1. The amino acid composition of the enzyme was also determined. Isocitrate lyase appears resistant to proteolysis by carboxypeptidase A. Hydrazinolysis, Edman degradation, and dansyl chloride treatment indicate that both carboxy and amino terminals are probably inaccessible or blocked. 相似文献

The use of sodium selenite as a catalyst in the presence of oxygen was a suitable technique to obtain in good yield an interchain S-S dimeric form of horse muscle acylphosphatase. The dimer so obtained possesses kinetic properties very similar to those of the native enzyme. On the other hand the dimer has shown a generally lower stability in respect of the thermal inactivation, particularly in the acidic environment, to the lyophilization and to the proteolytic attack. As regards the 8 M urea inactivation, the dimer is not able to completely regain its activity by dilution, showing a behaviour quite different from that of the native enzyme. 相似文献

The effect of different benzodiazepines on food intake was studied in cats which had been trained to eat their daily food within 3 hours, under conditions in which emotional factors were not present. When benzodiazepines were administered (at a dose ranging from 0.1 to 3 mg/Kg) before feeding, they increased both the total amount of food eaten and also the rate at which food was injested. When they were administered at the end of the feeding period, these compounds made the animals resume eating voraciously. The order of decreasing potency of the benzodiazepines tested was: oxazepam, N-methyl-lorazepam, diazepam, chlordiazepoxide, pinazepam, medazepam. Oxazepam (3 mg/Kg) stimulated maximally the food intake, even when administered up to 12 hours before feeding. Finally, oxazepam antagonized the anorexigenic effect of d-amphetamine, but did not influence amphetamine-induced hyperactivity and stereotyped behavior. 相似文献

We have predicted the secondary structure of horse muscle acylphosphatase by the statistical method of Chou and Fasman. In addition, we have studied the circular dichroism spectra of the enzyme, obtaining values for comparison to the predicted results. Therefore, it is important to assess them with regard to the longevity of the relined denture. This study investigated the effect of thermal cycling on the microleakage at the interface of three hard chairside reline resins and three denture base resins. Material and methods: Rectangular bars (12 mm × 3 mm × 3 mm) of Lucitone 550, Acron MC and QC 20 were made and relined with Kooliner, Tokuyama Rebase Fast II and Ufi Gel Hard, Lucitone 550, Acron MC and QC 20 resins. Specimens were divided into one control and two test groups (n = 10). In specimens of the control group, the microleakage was performed after the reline procedure. In Test Group 1, the specimens were stored for 24 h in distilled water at room temperature and in Test Group 2; the specimens were thermal cycled from 5 to 55°C for 5000 cycles with a 30‐s dwell time. Subsequently, all specimens were immersed in 50% silver nitrate solutions for 24 h. All specimens were sectioned longitudinally into three fractions and the lateral sections were examined (n = 20). Silver nitrate stain penetration was examined under a stereoscopic lens with ×30 magnification, and the images were captured. Leica Qwin image analysis software was used to determine microleakage at the interface of the materials. Data were analysed using the Kruskal–Wallis test at a 95% level of significance. Results: For all cycles, there were no statistically significant differences between thermal cycled and non‐thermal cycled groups (p > 0.05). Background
The identification of the adipocyte-derived obesity gene product, leptin (Ob), and subsequently its association with reproduction in rodents and humans led to speculations that leptin may be involved in the regulation of oocyte and preimplantation embryo development. In mice and pigs, in vitro leptin addition significantly increased meiotic resumption and promoted preimplantation embryo development in a dose-dependent manner. The influence of local (i.e. physico-chemical constraints) and regional (i.e. dispersal) processes in determining the structure of zooplankton communities in lentic waters was analysed in a mountain district of 4500 km2 (northern Apennines, Italy). In the 47 lakes and ponds considered in this study, a total of 139 taxa were found. Surface area (SA) and maximum depth (Zmax) significantly influenced species richness and nestedness, stressing the importance of habitat availability for zooplankton diversity and composition. Species richness and nestedness did not correlate with site isolation, indicating low influence of dispersal for these community properties. A negative relationship between the similarity in species composition among sites and their mutual geographic distance was also found. The relative importance of different sources of compositional variation was quantified by direct ordination. Local environmental conditions (i.e. Zmax, altitude, pH and conductivity) accounted for 65.9 % of the variation in rotifer assemblages. The importance of environmental constraints in structuring zooplankton communities was best documented by the microcrustacean distribution in the study area, which was mainly correlated (73.4 % of the variation in the species data) with water ionic content, surface area and hydroperiod of the habitats.Although previous studies on processes shaping zooplankton assemblages that operate at different scales have yielded contrasting results, our data provide clear evidence for a major role of local constraints. 相似文献
