The regulatory role of Arg283 in the autoinhibitory domain of Ca2+/calmodulin-dependent protein kinase II was investigated using substituted inhibitory synthetic peptides and site-directed mutation of the expressed kinase. In the synthetic peptide corresponding to the autoinhibitory domain (residues 281-309) of Ca2+/calmodulin-dependent protein kinase II, substitution of Arg283 by other residues increased the IC50 values of the peptides in the following order: Arg much less than Lys much less than Gln much less than Glu. Site-directed mutations of Arg283 to glutamic acid and glutamine in the kinase alpha subunit cDNA were transcribed and translated in vitro. The expressed enzymes had the same total kinase activities, determined in the presence of Ca2+/CaM, but the Glu283 mutant had a slightly higher Ca2(+)-independent kinase activity (5.46 +/- 0.88%) compared to the wild-type Arg283 (1.86 +/- 0.71%) and the Gln283 mutant (2.15 +/- 0.60%). When the expressed kinases were subjected to limited autophosphorylation on ice to monitor generation of the Ca2(+)-independent activity, the Arg283 kinase attained maximal Ca2(+)-independent activity (about 20%) within 30 s, whereas the Gln283 and Glu283 mutants attained maximal Ca2(+)-independence only after about 40 min of autophosphorylation. The results indicate that Arg283 is a very important determinant for the regulatory autophosphorylation of Thr286 that generates the Ca2(+)-independent activity but is not essential for the other multiple autophosphorylations within Ca2+/calmodulin-dependent protein kinase II, and that Arg283 is only one of several important residues for the inhibitory potency of the autoinhibitory domain. 相似文献

We have cloned the gene for human interstitial retinol-binding protein (IRBP) and compared its nucleotide sequence with that of the corresponding cloned cDNA. The human IRBP gene is approximately 9.5 kilobase pairs (kbp) in length and consists of four exons separated by three introns. The introns are 1.6-1.9 kbp long. The gene is transcribed by photoreceptor and retinoblastoma cells into an approximately 4.3-kilobase mRNA that is translated and processed into a glycosylated protein of 135,000 Da. The amino acid sequence of human IRBP can be divided into four contiguous homology domains with 33-38% identity, suggesting a series of gene duplication events. In the gene, the boundaries of these domains are not defined by exon-intron junctions, as might have been expected. The first three homology domains and part of the fourth are all encoded by the first large exon, which is 3,180 base pairs long. The remainder of the fourth domain is encoded in the last three exons, which are 191, 143, and approximately 740 base pairs long, respectively. This unusual structure is shared with the bovine IRBP gene. A large (1.7 kbp) fragment appears to have been lost from the 3'-noncoding region of the last human exon. Separation of dopamine (DA) agonist and antagonist receptors was attempted by means of a covalently-bound fluphenazine-agarose (Flu-agarose). Incubation of striatal membranes with Flu-agarose resulted in loss of 3H-spiroperidol (3H-SPI) binding sites, while incubation with non-coupled agarose did not cause any change. The loss of 3H-SPI binding to the Flu-agarose treated membranes was not attributed to the release of fluphenazine from Flu-agarose as justified by several criteria. Flu-agarose adsorbed more effectively striatal membranes with 3H-SPI binding sites than those with 3H-DA binding sites. Following incubation of the membranes with Flu-agarose (2.5 ml beads/100 mg membrane protein), the density of 3H-SPI binding sites in the resulting membranes was reduced to 29%, whereas the density of 3H-DA binding sites to the same membranes was not changed. In addition, the potencies of DA antagonists to inhibit 3H-N-propylnorapomorphine binding to the membranes were decreased more than a hundred times, while the potencies of DA agonists were little affected. Severe adverse reactions to propylthiouracil occur in 1-5% of patients. Three major side effects, namely agranulocytosis, hepatotoxicity and drug-induced hypersensitivity, have been described though these syndromes are not distinct entities and there can be overlaps in the clinical manifestations. The drug-induced hypersensitivity may be an immune-mediated reaction with multiorgan involvement in which a combination of polyarthritis, cutaneous vasculitis and fever is common. We report a patient with propylthiouracil-induced hypersensitivity with an unusual combination of high spiking fever, migratory polyarthritis, reversible sensorineural deafness, normochromic normocytic anaemia, leucocytosis and hepatotoxicity associated with polyclonal activation of multiple autoantibodies. This case illustrates the highly variable clinical manifestations of the syndrome. The prompt recovery upon withdrawal of the drug indicates the importance of early diagnosis. 相似文献

We have reported nine distinct karyotypes for Aotus, of four pelagic phenotypes, and suggest that this single species has undergone extensive subspeciation. We reconstruct the mechanism of chromosomal evolution and propose a hypothesis about the events of subspeciation in Aotus. We speculate that isolated groups of ancestral individuals living in several confined areas have separately accumulated a fusion or inversion pair as a result of inbreeding. A subsequent reassociation of descendants from these individuals led to the formation of offspring with mixtures of fusion or inversion pairs in their complements. They, in turn, radiated into different ecological niches accompanied by adaptive genetic changes and eventually gave rise to the present forms of Aotus distinguishable by their karyotypes, but not easily recognizable by ordinary taxonomic criteria. 相似文献

Pathogenesis of nonalcoholic fatty liver disease (NAFLD) is not clear. In this study we aimed to identify proteins involved in NAFLD development in free fatty acids (FFA)‐induced hepatosteatotic cells and in human liver biopsies. Steatosis was induced by incubating a normal human hepatocyte‐derived cell line L‐02 with FFA. Differentially expressed proteins in the steatotic cells were analyzed by two‐dimensional gel electrophoresis‐based proteomics. Involvement of one of the up‐regulated proteins in steatosis was characterized using the RNA interference approach with the steatotic cells. Protein expression levels in liver biopsies of patients with NAFLD were assessed by immunohistochemistry. Proteomic analysis of L‐02 steatotic cells revealed the up‐regulation of ERp57, a condition not previously implicated in NAFLD. Knockdown of ERp57 expression with siRNA significantly reduced fat accumulation in the steatotic cells. ERp57 expression was detected in 16 out of 17 patient biopsies and correlated with inflammation grades or fibrosis stages, while in 5 normal biopsies ERp57 expression was not detectable in hepatocytes. In conclusion, ERp57 was up‐regulated in FFA‐induced steatotic hepatic cells and in NAFLD patient livers and demonstrated steatotic properties in cultured cells. The shear strength of soil is an important parameter that affects tree stability and can vary depending on the magnitude of the soil’s negative pore-water pressure (matric suction). The surface flux boundary condition affects the matric suction of soil, and therefore is important for tree stability. Field measurements were performed around a roadside tree for 2 years. The instrumentation results show that the matric suction in the soil fluctuated between 0 and 35 kPa. Matric suction changes in the soil could lead to a decrease in the tree resistive moment of up to 80 %. 相似文献
