We sampled macroinvertebrates at 75 locations in the Mondego river catchment, Central Portugal, and developed a predictive
model for water quality assessment of this basin, based on the Reference Condition Approach. Sampling was done from June to
September 2001. Fifty-five sites were identified as “Reference sites” and 20 sites were used as “Test sites” to test the model.
At each site we also measured 40 habitat variables to characterize water physics and chemistry, habitat type, land use, stream
hydrology and geographic location. Macroinvertebrates were generally identified to species or genus level; a total of 207
taxa were found. By Unweighted Pair Group Method with Arithmetic mean (UPGMA) clustering and analysis of species contribution
to similarities percentage (SIMPER), two groups of reference sites were established. Using Discriminant Analysis (stepwise
forward), four variables correctly predicted 78% of the reference sites to the appropriate group: stream order, pool quality,
substrate quality and current velocity. Test sites’ environmental quality was established from their relative distance to
reference sites, in MDS ordination space, using a series of bands (BEAST methodology). The model performed well at upstream
sites, but at downstream sites it was compromised by the lack of reference sites. As with the English RIVPACS predictive model,
the Mondego model should be continually improved with the addition of new reference sites. The adaptation of the Mondego model
methodology to the Water Framework Directive is possible and would consist mainly of the integration of the WFD typology and
increasing the number of ellipses that define quality bands.

A predictive model for diatoms based on an adaptation of the River Invertebrate Prediction and Classification System/Australian River Assessment System approaches was evaluated as an effective tool for measuring stream ecological quality. This type of model was originally developed in UK and later in Australia and is extensively used to obtain ecological quality assessments with macroinvertebrates. The first step for the model construction was the definition of six consistent reference biological groups (ANOSIM: Global R = 0.77; P < 0.001) after classification (UPGMA) and ordination (nMDS) of 120 reference sites containing 254 different diatom taxa (species and infra-specific rank). A set of five environmental variables (slope, hydrological regime, mean annual temperature, mean annual precipitation and alkalinity) correctly discriminated 67% of reference sites (stepwise forward discriminant analysis, Jackknifed classification). The model was statistically accurate (slope = 1.07, intercept = −0.68, R 2 = 0.65) and was validated by an independent set of reference data (13 reference sites; 70% correct answers). In addition, the model was tested by running data from 113 potentially disturbed sites. The model (DIATMOD) was well correlated with a general abiotic degradation gradient (Spearman correlations, R 2 = 0.53, P < 0.001; and PCA analysis) and also with several specific pressure variables such as nitrates, phosphates, urban area, connectivity and land use (P < 0.001). Most diatom indices assess chemical contamination and we showed here that through predictive modelling the potential of diatoms as bioindicators increases as they also responded to hydromorphological changes. Further investigation on model potential consists in: testing different probability levels for taxa inclusion (here it was >0.5 as the most common models); comparing with alternative classification systems; assessing the influence of substrate type and seasonal variation in assessments. 相似文献

We report the characterization of the molecular properties and EPR studies of a new formate dehydrogenase (FDH) from the sulfate-reducing organism Desulfovibrio alaskensis NCIMB 13491. FDHs are enzymes that catalyze the two-electron oxidation of formate to carbon dioxide in several aerobic and anaerobic organisms. D. alaskensis FDH is a heterodimeric protein with a molecular weight of 126±2 kDa composed of two subunits, =93±3 kDa and =32±2 kDa, which contains 6±1 Fe/molecule, 0.4±0.1 Mo/molecule, 0.3±0.1 W/molecule, and 1.3±0.1 guanine monophosphate nucleotides. The UV-vis absorption spectrum of D. alaskensis FDH is typical of an iron-sulfur protein with a broad band around 400 nm. Variable-temperature EPR studies performed on reduced samples of D. alaskensis FDH showed the presence of signals associated with the different paramagnetic centers of D. alaskensis FDH. Three rhombic signals having g-values and relaxation behavior characteristic of [4Fe-4S] clusters were observed in the 5–40 K temperature range. Two EPR signals with all the g-values less than two, which accounted for less than 0.1 spin/protein, typical of mononuclear Mo(V) and W(V), respectively, were observed. The signal associated with the W(V) ion has a larger deviation from the free electron g-value, as expected for tungsten in a d1 configuration, albeit with an unusual relaxation behavior. The EPR parameters of the Mo(V) signal are within the range of values typically found for the slow-type signal observed in several Mo-containing proteins belonging to the xanthine oxidase family of enzymes. Mo(V) resonances are split at temperatures below 50 K by magnetic coupling with one of the Fe/S clusters. The analysis of the inter-center magnetic interaction allowed us to assign the EPR-distinguishable iron-sulfur clusters with those seen in the crystal structure of a homologous enzyme.Abbreviations AOR

The thermal unfolding of the plastocyanin from Phormidium laminosum, a thermophilic cyanobacterium, is herein described. The main objective of this work is to identify structural factors responsible for the higher stability observed in proteins from thermophilic organisms. With the aid of fluorescence spectroscopy, EPR, and NMR, the factors influencing the unfolding process of the protein were investigated, and procedures for its study have been standardized. The different spectroscopic techniques used provided consistent results showing that the thermal unfolding of plastocyanin is irreversible under all the conditions investigated and that this irreversibility does not appear to be related to the presence of oxygen. The oxidized plastocyanin species has proven to be more stable than the reduced one, with respect to both the required temperature for protein unfolding (up to a 9 degrees C difference between the two forms) and the kinetics of the process. The behavior of this plastocyanin contrasts with that of other cupredoxins whose unfolding had previously been studied. The unfolding pH dependence and kinetic studies indicate a process with a tight control around the physiological pH in which plastocyanin plays its redox role and the protein's isoelectric point (5.2), suggesting a close compromise between function and stability. 相似文献

The effect of changing dilution rate (D) on Bacillus sp. CCMI 1051 at dilution rates between 0.1 and 0.55 h−1 in a glucose-limited medium was studied. Biomass values varied between 0.88 and 1.1 g L−1 at D values of 0.15–0.35 h−1. Maximal biomass productivity was found to be 0.39 g L−1 h−1, obtained at D = 0.35 h−1 and corresponding to a 54.4% conversion of the carbon into cell mass. The highest rate of glucose consumption was 4.45 mmol g−1 h−1 occurring at D = 0.4 h−1. The glucose concentration inside the chemostat was below the detection level starting to accumulate around 0.4 h−1. Growth inhibition of fifteen strains of fungi by the broth of the steady-state cell-free supernatants was assessed. Results showed that the relative inhibition differ among the target species but was not influenced by the dilution rate changing. 相似文献

Aims: The aim of this study was to evaluate the influence of environmental conditions on the antifungal activity of the Bacillus sp. CCMI 1053 cultures. Methods and Results: The electrospray ionization mass spectra (ESI‐MS) analysis was used to detect the active peptides produced by Bacillus amyloliquefaciens CCMI 1051 cultures in a glucose‐containing medium to which four different nitrogen sources were added. The cultures produced different patterns of Bacillus sporulation and distinct antifungal activity of the cell‐free culture broths. Conclusions: The highest sporulation obtained corresponds to higher antifungal activity when it is formed after 3 days of microbial growth. The antifungal activity against Trichoderma harzianum CCMI 783 is more influenced by the concentration on the nitrogen source than the culture time of incubation. The association of nitrogen concentration and the time of incubation is particularly relevant in the expression of the antifungal activity. Significance and Impact of the Study: The present findings allow the reduction of the use of chemical pesticides and to limit some plant diseases. The association of the nitrogen source and the time of incubation is a novelty, which would improve the production of secondary metabolites. The caddisfly Sericostoma vittatum Rambur (Trichoptera: Sericostomatidae) is an endemic species of the Iberian Peninsula. Under laboratory conditions, larvae of S. vittatum had a higher activity and metabolism during the night. Besides consuming particulate allochthonous organic matter, young stages are also able to feed and grow on faecal pellets from adults. Daily growth rates varied from 0.02 mg (0.8–3.7 mg size class animals) to 0.31 mg dry mass (10.6–22.8 mg size class animals). Due to the high densities of this species (annual mean of 25 individuals m–2; maximum of 96 individuals m–2) and high consumption rates (0.47 mg leaf dry mass mg animal–1 d–1for small larvae), this species has a potential key role on the fragmentation of allochthonous organic matter of streams in central Portugal. 相似文献

To understand whether seasons influence the ecological quality assessment of streams on the basis of diatoms, a study was
undertaken in two lowland water courses located in northwest Portugal, between autumn 2008 and summer 2009. Temporal variation
in the chemical pollution of these streams was small as revealed by a number of physical and chemical parameters analyzed.
PERMANOVA global test revealed significant variation in water temperature among seasons. The diatom communities also showed
a temporal variation although not all the seasons were statistically different. The multidimensional scaling analysis showed
that the main differences in the diatom communities were between two groups of seasons: autumn/winter and spring/summer. Species
such as Cocconeis pseudolineata, Gomphonema parvulum var. exilissimum, Fragilaria vaucheriae, Encyonema minutum, and Nitzschia recta were more abundant in spring/summer, while species such as Mayamaea atomus and Nitzschia pusilla were more abundant in autumn/winter. The BIOENV routine confirmed that the biological and temperature patterns are highly
correlated. Despite the effects observed on diatom communities, these differences were buffered by the EQR (IPS) values which
do not reflect seasonal differences. Therefore, the use of the index IPS seems to allow the monitoring of the streams’ ecological
quality throughout the year without the interference of the natural temporal variability of diatom communities. 相似文献

(-)-Agelasidine A was identified from the methanol extract of the marine sponge Agelas clathrodes for the first time together with zooanemonin, 1-carboxymethylnicotinic acid, hymenidin, mukanadins A and C, monobromodispacamide, agelasidine D, 2-amide-4-bromopyrrole, O-methyltryptophan and an agelasines mixture. The structures were characterized by spectroscopic methods. (-)-Agelasidine A was tested for antibacterial and antifungal activities and shown to act as a bacteriostatic agent as it inhibited the growth of Staphylococcus aureus and partially the growth of other bacteria. 相似文献

AIMS: To study the antifungal activity of methyl cis-7-oxo-deisopropyldehydroabietate (MCOD) against phytopathogenic fungi, Botrytis cinerea and Lophodermium seditiousm. The effect of the compound was studied by transmission electron microscopy (TEM) and the composition of sterols on both treated and untreated cultures was determined. METHODS AND RESULTS: MCOD was tested at concentrations in the range 0.003-0.5% by the agar plate dilution method. The radial growth of the colonies treated with MCOD was measured against colonies from untreated cultures. The radial growth of colonies of both fungi and the spore germination of B. cinerea were partially or completely inhibited. Fragments of active growing colonies treated and untreated with MCOD were submitted to the conventional procedure for ultrastructural observation by TEM. Observations by TEM on colonies of B. cinerea and L. seditiosum under 0.1% MCOD revealed several autophagic-like vacuoles, morphological alterations on lomasome and lipid accumulations in the apical zone of hyphae of both fungi. Observations on spore germination of B. cinerea revealed the presence of strongly stained lipid accumulations retained by vacuoles at the cell periphery of young hyphae. The sterol composition of B. cinerea and L. seditiosum was determined on MCOD treated and untreated cultures by gas-chromatography/mass-spectrometry (GC-MS) with molecular ions and fragmentation patterns characteristics of ergosterol (M+396) and dihydroergosterol (M+398) in both fungi. CONCLUSIONS: The morphological alterations are consistent with an unspecific mode of action of MCOD causing inhibition of normal growth or damaging the fungi cells. TEM observations suggest a mechanism of resistance based on the retention of MCOD by the lipid accumulation. SIGNIFICANCE AND IMPACT OF THE STUDY: The results obtained in the present work afforded a better understanding of the mode of action of a resin acid derivative on phytopathogenic fungi. The inhibition growth of both fungi by MCOD demonstrates the antifungal activity of this compound and the interest on further in vivo studies, in order to evaluate its potential as a benign alternative to conventional fungicides. 相似文献
