Sensory neurons provide important feedback to pattern-generating motor systems. In the crustacean stomatogastric nervous system (STNS), feedback from the anterior gastric receptor (AGR), a muscle receptor neuron, shapes the activity of motor circuits in the stomatogastric ganglion (STG) via polysynaptic pathways involving anterior ganglia. The AGR soma is located in the dorsal ventricular nerve posterior to the STG and it has been thought that its axon passes through the STG without making contacts. Using high-resolution confocal microscopy with dye-filled neurons, we show here that AGR from the crab Cancer borealis also has local projections within the STG and that these projections form candidate contact sites with STG motor neurons or with descending input fibers from other ganglia. We develop and exploit a new masking method that allows us to potentially separate presynaptic and postsynaptic staining of synaptic markers. The AGR processes in the STG show diversity in shape, number of branches and branching structure. The number of AGR projections in the STG ranges from one to three simple to multiply branched processes. The projections come in close contact with gastric motor neurons and descending neurons and may also be electrically coupled to other neurons of the STNS. Thus, in addition to well described long-loop pathways, it is possible that AGR is involved in integration and pattern regulation directly in the STG. 相似文献

There is an increasing need for more efficient generation of transgenic constructs. Here we present a universal multi-site Gateway vector for use in recombineering reactions. Using transgenic mouse models, we show its use for the generation of BAC transgenics and targeting vectors. The modular nature of the vector allows for rapid modification of constructs to generate different versions of the same construct. The presence of Francisella-like endosymbionts in tick species known to transmit tularemia poses a potential diagnostic problem for laboratories that screen tick samples by PCR for Francisella tularensis. Tick samples initially considered positive for F. tularensis based on standard 16S rRNA gene PCR were found to be positive only for Francisella-like endosymbionts using a multitarget F. tularensis TaqMan assay (ISFtu2, tul4, and iglC) and 16S rRNA gene sequencing. Specificity of PCR-based diagnostics for F. tularensis should be carefully evaluated with appropriate specimen types prior to diagnostic use. 相似文献

Pogonophora, also known as Siboglinidae, are tube-dwelling marine annelids. They rely on endosymbiotic chemoautotrophic bacteria
for nutrition and their anatomy and physiology are adapted to their need to obtain both oxygen and reduced sulphur compounds.
Frenulate pogonophores are generally long and slender, sediment-living tubeworms; vestimentiferans are stouter, inhabitants
of hydrothermal vents and cool seeps; and moniliferans or sclerolinids are very slender inhabitants of decaying wood and sulphidic
sediments. The anatomy and ultrastructure of the three groups are compared and recent publications are reviewed. Annelid characters
are the presence of chaetae and septa, concentrated at the hind end. The adaptations to a specialised way of life include,
in particular, the chitinous tube; the anterior appendages that function as gills; the internal tissue called the trophosome,
where the endosymbiotic bacteria live; and the blood vascular system that transports oxygen, sulphide and carbon dioxide to

Biotic indirect effects: a neglected concept in invasion biology 总被引:7,自引:2,他引:5
Indirect effects involve more than two species and are defined as how one species alters the effect that another species has on a third. These complex interactions are often overlooked in studies of interactions between alien and native species, and their role in influencing biological invasions has been rarely considered. Based on a comprehensive review of the invasion biology literature, we examine the evidence for the occurrence of four of the most commonly documented indirect effects (apparent competition, indirect mutualism/commensalism, exploitative competition, and trophic cascades) in the invasion process. Studies investigating indirect effects in the context of invasion biology are relatively rare, but have been increasing in recent years, and there are sufficient examples to indicate that this kind of interaction is likely to be more common than is currently recognized. Whether indirect interactions are mediated by an alien or a native species, and whether they occur between ecologically similar or dissimilar alien and native species, depends in part on the type of interaction considered and no predictable patterns were detected in the literature. Further empirical studies will help to elucidate such patterns. At this stage, the inherent unpredictability of indirect interactions means that their impacts in relation to invasions are particularly challenging for land managers to deal with, and their role in invasions is a complex, but is a valuable area of investigation for researchers. 相似文献

Shared and Separate Knowledge among Eight Cultural Groups Based on Ethnobotanical Uses of Rhododendron (Ericaceae) in Yunnan Province, China. Yunnan, a province in southwest China, is known for its cultural diversity of 25 ethnic minorities and its vast Himalayan biodiversity, especially of Rhododendron. Previous literature has shown that some cultural groups share ethnobotanical knowledge while other cultural groups keep their knowledge separate. We investigated factors that may lead to the sharing of knowledge based on the uses of rhododendron among seven cultural minorities (the Bai, Dulong, Lisu, Naxi, Nu, Tibetan, and Yi) and the Han majority. Semistructured interviews about rhododendrons were conducted with approximately 30 individuals in each cultural group. Cluster analyses and a new analysis method were conducted to determine the within-group homogeneity of knowledge of rhododendron uses to test hypotheses related to strength of cultural traditions. The Dulong, Lisu, and Nu were compared with each other as these groups share villages and languages. The Naxi, Tibetan, and Yi live predominantly with members of their cultural group, and are often monolingual; thus, these three cultural minorities were compared. The Bai and Han compose the final comparison as the Bai are increasingly interacting with the local Han majority as tourism grows in that area. The Bai, Dulong, Han, Lisu, and Nu had variable answers within each group, while the Naxi, Tibetan, and Yi have homogeneous knowledge of uses of rhododendron within their cultural group. Among the eight cultural groups compared for this study, factors such as sharing of language, overlap of living situation, and sharing of markets leads to non-homogenous knowledge of rhododendron uses among members of the same cultural group. 相似文献

The 5-HT2A receptor (5-HT2AR) is implicated in psychotropic changes within the central nervous system (CNS). A number of polymorphisms have been reported in the 5-HT2AR gene; one of these results in a non-synonymous change, H452Y, in the carboxy-terminal tail of the receptor protein. The minor allele (9% occurrence) has been statistically associated with CNS dysfunction such as impaired memory processing and resistance to neuroleptic treatment in schizophrenic patients. We investigated the impact of H452Y mutation of the 5-HT2AR expressed in COS7 cells on distinctly coupled intracellular signalling pathways from the receptor, focusing on the heterotrimeric G protein-independent phospholipase D (PLD) pathway, compared to the conventional Gq/11-linked phospholipase C (PLC) pathway. The H452Y mutation selectively attenuated PLD signalling, which as in the wild-type receptor, was mediated by a molecular complex involving PLD1 docked to the receptor's carboxy-terminal tail domain. Co-immunoprecipitation and GST-fusion protein experiments revealed that the H452Y mutation selectively reduced PLD1 binding to the receptor. Experiments with blocking peptides to mimic short sections of the 5-HT2AR tail sequence revealed that the peptide spanning residue 452 strongly reduced PLD but not PLC responses of the receptor. Similar observations were made when assessing both PLD responses and PLD-dependent cellular proliferation elicited by activation of 5-HT2ARs natively expressed in MCF-7 cells. Overall these findings indicate that the H452Y polymorphic variant of the 5-HT2AR displays selective disruption of its PLD signalling pathway. This may potentially play a role in the CNS dysfunction associated with the H452Y allele of the 5-HT2AR. 相似文献
