The formin protein formin-like 1 (FMNL1) is highly restrictedly expressed in hematopoietic lineage-derived cells and has been previously identified as a tumor-associated antigen. However, function and regulation of FMNL1 are not well defined. We have identified a novel splice variant (FMNL1γ) containing an intron retention at the C terminus affecting the diaphanous autoinhibitory domain (DAD). FMNL1γ is specifically located at the cell membrane and cortex in diverse cell lines. Similar localization of FMNL1 was observed for a mutant lacking the DAD domain (FMNL1ΔDAD), indicating that deregulation of autoinhibition is effective in FMNL1γ. Expression of both FMNL1γ and FMNL1ΔDAD induces polarized nonapoptotic blebbing that is dependent on N-terminal myristoylation of FMNL1 but independent of Src and ROCK activity. Thus, our results describe N-myristoylation as a regulative mechanism of FMNL1 responsible for membrane trafficking potentially involved in a diversity of polarized processes of hematopoietic lineage-derived cells.  相似文献

Zusammenfassung 1. Auf Grund der Untersuchung von zahlreichen Stämmen von Drosophila subobscura, die aus Köderfängen in der Natur an verschiedenen Orten in Österreich, Oberitalien, der Schweiz, England und einigen außereuropäischen Ländern abgeleitet worden waren, wurde ein Verzeichnis aller bisher bei dieser Art festgestellten natürlichen Inversionen gegeben, wobei die Anordnung des strukturell homozygoten Stammes Küsnacht als Standardanordnung diente.2. Durch den Vergleich von Stämmen, durch deren Kreuzung mit dem Standardstamm, durch die Ableitung von Inzuchtlinien aus verschiedenem Ausgangsmaterial und deren Kreuzung mit bestimmten Stämmen wurde die Zahl und der Bau der chromosomalen Strukturtypen ermittelt, auf deren Koexistenz in der Population die Mannigfaltigkeit der beobachteten Inversionsbilder beruht.3. Die Verteilung der Inversionen auf die verschiedenen Chromosomen des Satzes ist nicht zufällig. Auch die relative Länge der Inversionen scheint nicht rein zufallsmäßig zu sein.4. Bei den Bruchstellen der Inversionen finden sich auffallend oft Koinzidenzen, bis zu 5 Bruchstellen an einem Ort eines Chromosoms. Wie eine statistische Überlegung zeigt, können diese nicht zufallsmäßig bedingt sein. Es werden einige hypothetische Deutungsmöglichkeiten dieses Phänomens vorgelegt.5. Catabolism of flavonol glucosides was investigated in plant cell suspension cultures using kaempferol 3-O-β-d-glucoside and kaempferol 7-O-β-d-glucoside labelled with 14C either in the glucose or in the flavonol moiety. Catabolic rates of glucosides were compared with those of free glucose and kaempferol. All substrates were degraded efficiently by cell cultures of mungbean, soybean, garbanzo bean and parsley. Based on 14CO2-formation, glucose from position 3 of kaempferol is 3–5 times more rapidly metabolized than that from position 7. The flavonol nucleus from both isomers is, however, oxidized to the same extent with a considerable portion of the flavonol being incorporated into insoluble polymeric cell material.  相似文献

Objective: Severe obesity is a clear indication for appropriate, effective weight loss therapy. One option is operative intervention, e.g., gastric banding. Risks of the operation and therapeutic alternatives need to be comprehensibly presented to the patient. The literature has shown that better informed consent is obtained using information presented in a multimedia/video‐based format. The current study developed and evaluated a multimedia program aimed at obtaining informed consent from obese patients before gastric banding. Research Methods and Procedure: An interactive multimedia program was developed with information about preoperative examinations, the operation itself, hospital stay, operative risks, alternative therapies, and the pathophysiology and health risks of obesity. Two groups (Group 1, n = 20, mean age 38 years, informed consent attained with conventional document information; Group 2, n = 20, mean age 37 years, informed consent attained with additional multimedia information) were interviewed regarding comprehensibility of the information presented, personal satisfaction, and anxiety levels during the informed consent process. Results: Group 2 showed significantly better (p < 0.05) understanding of the presented information and higher levels of satisfaction with the informed consent process. Anxiety levels did not significantly differ between the two groups. Discussion: Because patient satisfaction with the informed consent process and understanding of the presented information significantly improved, the multimedia program clearly benefits both surgeons and patients. Personal contact from the surgeon remains essential. High volumes of information presented in multimedia format do not alleviate patient anxiety, and personal contact may be beneficial.  相似文献

Short- and long-chain 1-O-alkyl-2-acylaminodeoxyglycero- and alkoxy-alkylphosphonic acid p-nitrophenyl esters were synthesized as inhibitors for analytical and mechanistic studies on lipolytic enzymes. The respective compounds contain perylene or nitrobenzoxadiazole as reporter fluorophores covalently bound to the omega-ends of the respective 2-acylamino- and alkoxy- residues. Their inhibitory effects on the activities of three selected lipases showing different substrate preferences were determined, including the lipases from Rhizopus oryzae, Pseudomonas species, and Pseudomonas cepacia. R. oryzae lipase reacted much better with the single-chain inhibitors than the two-chain deoxyglycerolipids. In contrast, P. cepacia lipase was inactivated by perylene-containing two-chain phosphonate (XXII) to a larger extent as compared to the other inhibitors whereas Pseudomonas species lipase interacted efficiently and without any preferences with all inhibitors used in this study. In summary, the different lipases show a very characteristic reactivity pattern not only with respect to triacylglycerol substrates but also to their structurally related inhibitors. Thus, the novel phosphonates might be useful tools not only for analysis and discrimination of known lipolytic enzymes but also for discovery of yet unknown lipases/esterases in biological samples.  相似文献

Several structurally different glucans (alpha- and beta-) and galactomannans were characterized as components of four species of the genus Ramalina, namely R. dendriscoides, R. fraxinea, R. gracilis and R. peruviana. Freeze-thawing treatment of hot aqueous extracts furnished as precipitates (PW) linear alpha-D-glucans of the nigeran type, with regularly distributed (1-->3)- and (1-->4)-linkages in a 1:1 ratio. The supernatants (SW) contained alpha-D-glucans with (1-->3)- and (1-->4)-linkages in a molar ratio of 3:1. The lichen residues were then extracted with 2% aq. KOH, and the resulting extracts submitted to the freeze-thawing treatment, giving rise to precipitates (PK2) of a mixture of alpha-glucan (nigeran) and beta-glucan, which were suspended in aqueous 0.5% NaOH at 50 degrees C, dissolving preferentially the beta-glucan. These were linear with (1-->3)-linkages (laminaran). The mother liquor of the KOH extractions (2% and 10% aq. KOH) was treated with Fehling's solution to give precipitates (galactomannans). The galactomannans are related, having (1-->6)-linked alpha-D-mannopyranosyl main chains, substituted at O-4 and in a small proportion at O-2,4 by beta-D-galactopyranosyl units. Despite the different habitats of these lichenized fungi, all species studied in this investigation have a similar pool of polysaccharides.  相似文献
