Unconventional Sequence Requirement for Viral Late Gene Core Promoters of Murine Gammaherpesvirus 68

Infection with the human gammaherpesviruses, Epstein-Barr virus (EBV) and Kaposi''s sarcoma-associated herpesvirus (KSHV), is associated with several cancers. During lytic replication of herpesviruses, viral genes are expressed in an ordered cascade. However, the mechanism by which late gene expression is regulated has not been well characterized in gammaherpesviruses. In this study, we have investigated the cis element that mediates late gene expression during de novo lytic infection with murine gammaherpesvirus 68 (MHV-68). A reporter system was established and used to assess the activity of viral late gene promoters upon infection with MHV-68. It was found that the viral origin of lytic replication, orilyt, must be on the reporter plasmid to support activation of the late gene promoter. Furthermore, the DNA sequence required for the activation of late gene promoters was mapped to a core element containing a distinct TATT box and its neighboring sequences. The critical nucleotides of the TATT box region were determined by systematic mutagenesis in the reporter system, and the significance of these nucleotides was confirmed in the context of the viral genome. In addition, EBV and KSHV late gene core promoters could be activated by MHV-68 lytic replication, indicating that the mechanisms controlling late gene expression are conserved among gammaherpesviruses. Plantlets of Chrysanthemum x morifolium were grown from nodal sections in cellulose plugs which were saturated with liquid rooting medium containing 1 mg l–1 paclobutrazol in culture vessels that maintained relative humidities (RH) of 100%, 96% and 94%, respectively. After 4 weeks, plantlets were transferred to compost and exposed to 40% RH at 29°C. Marked differences in wilting were observed that were directly related to the RH of the vessel in which the plantlets had been grown. Thicker leaves, improved closure of stomata and increased thickness of the cuticle were associated with increased resistance to wilting. Reduced RH of the culture vessel was also associated with significantly higher concentrations of chlorophyll in the leaves. 相似文献

Serum from larvae of Lacanobia oleracea L. (Lepidoptera; Noctuidae) parasitized by Eulophus pennicornis (Hymenoptera; Eulophidae) and from normal non‐parasitized larvae is capable of agglutinating rabbit, sheep, calf, goat, chicken, horse and human erythrocytes, but not yeast. Studies with a range of inhibitory carbohydrates showed that serum lectins(s) had specificity for sugars containing galactose and for rhamnose, and for the glycosubstances fetuin and asialofetuin. Lectin activity is heat‐labile and is not dependent on calcium. Parasitism by E. pennicornis caused an increase in the agglutination titre of the serum from larvae of L. oleracea but not an increase in specific activity (titre per mg protein per ml). However, when venom from the venom gland of female wasps was injected into L. oleracea larvae, both the agglutinating activity and the specific activity of the larval serum increased. The possible causes of this increase are discussed. It is suggested that venom contains antigenic components which, when injected into the haemocoel of the L. oleracea larva, may be increasing lectin synthesis and/or release into the serum. 相似文献

Pseudomonas aeruginosa UFPEDA 614 produced a rhamnolipid biosurfactant when grown on sugarcane bagasse impregnated with a solution containing glycerol. Biosurfactant levels reached 40 g of rhamnolipid per kilogram of dry initial substrate after 12 days. On the basis of the volume of liquid used, the biosurfactant levels were similar to those obtained in submerged liquid culture of a medium identical to the impregnating solution. The properties of the biosurfactant were very similar to those obtained with rhamnolipids produced in submerged culture, with a critical micelle concentration of 46.8 mg/L and an emulsification index at 24 h of over 90% against gasoline. The surface properties were maintained after autoclaving of the fermented solids, meaning that it is possible to minimize safety risks by killing the producing organism with a heat treatment of the solids prior to product extraction. The biosurfactant was used in the washing of soils contaminated with gasoline. An aqueous biosurfactant solution was 3.2-fold more efficient than water in leaching organic material from the soil, demonstrating the viability of application of rhamnolipids in the bioremediation of soils contaminated with gasoline. 相似文献

White‐sand forests are patchily distributed ecosystems covering just 5% of Amazonia that host many specialist species of birds not found elsewhere, and these forests are threatened due to their small size and human exploitation of sand for construction projects. As a result, many species of birds that are white‐sand specialists are at risk of extinction, and immediate conservation action is paramount for their survival. Our objective was to evaluate current survey methods and determine the relative effect of the size of patches of these forests on the presence or absence of white‐sand specialists. Using point counts and autonomous recorders, we surveyed avian assemblages occupying patches of white‐sand forest in the Peruvian Amazon in April 2018. Overall, we detected 126 species, including 21 white‐sand forest specialists. We detected significantly more species of birds per survey point with autonomous recorders than point counts. We also found a negative relationship between avian species richness and distance from the edge of patches of white‐sand forest, but a significant, positive relationship when only counting white‐sand specialists. Although we detected more species with autonomous recorders, point counts were more effective for detecting canopy‐dwelling passerines. Therefore, we recommend that investigators conducting surveys for rare and patchily distributed species in the tropics use a mixed‐method approach that incorporates both autonomous recorders and visual observation. Finally, our results suggest that conserving large, continuous patches of white‐sand forest may increase the likelihood of survival of species of birds that are white‐sand specialists. 相似文献

Abstract: To investigate the regulation of posttranslational modifications of τ that might be pertinent to the production of the paired helical filament (PHF) of Alzheimer's disease, we incubated human neuroblastoma cells with the protein phosphatase inhibitor okadaic acid. This treatment results in increased immunoreactivity of τ with the monoclonal antibodies Alz-50, PHF-1, T3P, and NP8, a reduction in Tau-1 immunoreactivity, and an elevation in apparent molecular weight of τ. Moreover, our data demonstrate that accumulation of phosphates in τ leads to a decrease in the turnover rate of τ in the neuroblastoma cells. It is suggested that similar build-up of hyperphosphorylated τ in the neuronal perikarya may represent an early event in PHF formation. The present system facilitates the investigation of regulatory mechanisms governing the occurrence of PHF epitopes, their effects on neuronal cell metabolism, and possible pharmacological intervention. 相似文献
