根癌农杆菌Ti质粒的T区DNA带有致瘤基因,其基因1和基因2编码生长素吲哚乙酸生物合成途径中的两个酶。以pGV 354(pBR322质粒中插有Ti质粒C 58 T区DNA的HindⅢ15—HindⅢ22大片段)重组质粒出发,我们分离了基因1和基因2,并构建了带有卡那霉素抗性基因的重组质粒pBZ 692,通过基因载体pGV 3850,我们将基因1和基因2引入了高等植物。结果证明基因1和基因2能促使烟草、向日葵、土豆等转化组织分化长根,转化的根在MS_0培养基上能脱分化形成愈伤组织并自主生长,在转化的组织中有转化标记胭脂碱的存在。  相似文献

本文记述采自新疆南部的裸蝗属Conophyma Zub.二新种.新种“友谊裸蝗”Conophyma amica近似于昭苏裸蝗C.zhaosuensis Huang,以下列特征区别于后者:1)雄性肛上板两后角明显突出,后缘中央锐角形突出;2)后足胫节红褐色.新种“红胫裸蝗”C.rufitibia与C.alajense Misth.相似,其区别为:1)新种雄性尾片显著较大;2)雄性尾须较长,明显长过肛上板后缘中央的突起,3)雌性下产卵瓣下外缘缺一明显的大齿.红胫裸蝗亦近似于C.alamasyi(Kuthy),但可以下述特征区别之:1)雄性肛上板近方形,侧缘中部具一钝瘤状突起;2)雄性腹部背面具二条淡色纵带.模式标本均保存在新疆八一农学院植保系.  相似文献

【背景】大熊猫数量稀少、繁殖困难,在其生长过程中极易感染消化系统疾病甚至死亡。【目的】分析圈养大熊猫肠道可培养乳酸菌的群落结构与功能,筛选具有益生特性的乳酸菌菌株,为大熊猫消化系统疾病的预防和肠道微生物研究提供理论参考及菌种资源。【方法】采用3种培养基分离大熊猫粪便中的乳酸菌,通过革兰氏染色镜检和过氧化氢试验对分离的菌株进行初步鉴定,基于BOXA1R-PCR图谱遗传多样性选取代表菌株进行16S rRNA基因测序分析并进行主成分分析(principal component analysis, PCA),同时分析乳酸菌菌株安全性和益生特性。【结果】通过初步鉴定共分离获得58株乳酸菌,根据BOXA1R-PCR结果挑选20株菌进行测序,结果显示20株菌分属于明串珠菌属(Leuconostoc)、肠球菌属(Enterococcus)、魏斯氏菌属(Weissella)和链球菌属(Streptococcus)这4个属,主成分分析结果表明不同年龄段的大熊猫肠道乳酸菌群落结构存在差异。20株乳酸菌均不溶血,17株乳酸菌对11种抗生素均敏感;11株菌耐pH值2.0酸性条件,14株菌对0.3%的胆盐具有良好...  相似文献

To determine 15 bile acid metabolic products in human serum by liquid chromatography-tandem mass spectrometry (LC/MS/MS) and value their diagnostic outcome in primary biliary cholangitis (PBC). Serum from 20 healthy controls and 26 patients with PBC were collected and went LC/MS/MS analysis of 15 bile acid metabolic products. The test results were analyzed by bile acid metabolomics, and the potential biomarkers were screened and their diagnostic performance was judged by statistical methods such as principal component and partial least squares discriminant analysis and area under curve (AUC). 8 differential metabolites can be screened out: Deoxycholic acid (DCA), Glycine deoxycholic acid (GDCA), Lithocholic acid (LCA), Glycine ursodeoxycholic acid (GUDCA), Taurolithocholic acid (TLCA), Tauroursodeoxycholic acid (TUDCA), Taurodeoxycholic acid (TDCA), Glycine chenodeoxycholic acid (GCDCA). The performance of the biomarkers was evaluated by the AUC, specificity and sensitivity. In conclusion, DCA, GDCA, LCA, GUDCA, TLCA, TUDCA, TDCA and GCDCA were identified as eight potential biomarkers to distinguish between healthy people and PBC patients by multivariate statistical analysis, which provided reliable experimental basis for clinical practice.  相似文献

An insecticidal crystal protein gene of Bacillus thuringiensis was transferred into cabbage genome with the method of Agrobacterium infection. Cotyledons with petioles as explants were cocultivated with Agrobacterial suspension. Calli generated at the basis of petiole were subjected to selection on the MS medium containing 15-30 mg/L kanamycin (Km). About 5% explants produced calli growing continuously on the selective medium. Green shoots appeared on these calli when they were transplanted onto medium with Km and 6-BA for plant differentiation. The shoots were separated and cultivated on medium with kanamycin. About 80% shoots were rooted. Non-transformed control calli could not give normal shoots and roots and brownized and died gradually. Larvae of Pieris rapae showed poisonous symptoms: growth inhibition and mortality when fed with the leaf of the transgenic plants. About 80% of regenerated plants showed positive hybridization bands when their DNA were probed with crystal protein sequence of Bacillu  相似文献

Fruit crops, including apple, orange, grape,banana, strawberry, watermelon, kiwifruit and tomato, not only provide essential nutrients for human life but also contribute to the major agricultural output and economic growth of many countries and regions in the world. Recent advancements in genome editing provides an unprecedented opportunity for the genetic improvement of these agronomically important fruit crops. Here, we summarize recent reports of applying CRISPR/Cas9 to fruit crops,including efforts to reduce disease susceptibility, change plant architecture or flower morphology, improve fruit quality traits, and increase fruit yield. We discuss challenges facing fruit crops as well as new improvements and platforms that could be used to facilitate genome editing in fruit crops, including d Cas9-base-editing to introduce desirable alleles and heat treatment to increase editing efficiency. In addition, we highlight what we see as potentially revolutionary development ranging from transgene-free genome editing to de novo domestication of wild relatives. Without doubt, we now see only the beginning of what will eventually be possible with the use of the CRISPR/Cas9 toolkit. Efforts to communicate with the public and an emphasis on the manipulation of consumerfriendly traits will be critical to facilitate public acceptance of genetically engineered fruits with this new technology.  相似文献

In organic semiconductors, optical excitation does not necessarily produce free carriers. Very often, electron and hole are bound together to form an exciton. Releasing free carriers from the exciton is essential for the functioning of photovoltaics and optoelectronic devices, but it is a bottleneck process because of the high exciton binding energy. Inefficient exciton dissociation can limit the efficiency of organic photovoltaics. Here, nanoscale features that can allow the free carrier generation to occur spontaneously despite being an energy uphill process are determined. Specifically, by comparing the dissociation dynamics of the charge transfer (CT) exciton at two donor–acceptor interfaces, it is found that the relative orientation of the electron and hole wavefunction within a CT exciton plays an important role in determining whether the CT exciton will decompose into the higher energy free electron–hole pair or relax to the lower energy tightly‐bound CT exciton. The concept of the entropic driving force is combined with the structural anisotropy of typical organic crystals to devise a framework that can describe how the orientation of the delocalized electronic wavefunction can be manipulated to favor the energy‐uphill spontaneous dissociation of CT excitons over the energy‐downhill CT exciton cooling.  相似文献
