The effects of water activity (aw, 0.955 to 0.970), pH (4.75 to 5.75), and storage time (up to 60 days) on toxin production by Clostridium botulinum in cooked, vacuum-packed potatoes were studied by using factorial design experiments and most-probable-number methodology. Samples were inoculated with 10(3), 10(4), or 10(5) spores of a mixture of five type A and five proteolytic type B strains, incubated at 25 degrees C, and analyzed for toxin production. Toxin was produced at pH levels of greater than or equal to 4.75 when the aw was greater than or equal to 0.970, pH greater than 5.25 when the aw was 0.965, and pH greater than or equal to 5.75 at an aw of 0.960. No toxin was detected when the aw was 0.955. The probability of toxigenesis was significantly affected (P less than 0.0001) by storage time, aw, pH, and the interactions aw.pH and aw.storage time. The response to a decrease in pH was linear, while the response to a decrease in aw was curvilinear. Using multiple linear regression, equations were derived which could predict the length of time until toxin production and the probability of toxigenesis by a single spore under defined conditions.  相似文献

Pharmaceutical aerosols have been targeted to the lungs for the treatment of asthma and pulmonary infectious diseases successfully. Micafungin (Astellas Pharma US, Deerfield, IL, USA) has been shown to be an effective antifungal agent when administrated intravenously. Pulmonary delivery of micafungin has not previously been reported. In the present pilot study, we characterize the performance of two nebulizers and their potential for delivering micafungin to the lungs as well as the use of multivariate data analysis for mass distribution profile comparison. The concentration of micafungin sodium increased by 21% when delivered by the Acorn II nebulizer and by 20% when delivered by the LC Plus nebulizer, respectively, from the first to the second sampling period. The Acorn II nebulizer delivered a fine particle fraction FPF5.8 (%<5.8 μm) of 92.5 ± 0.8 and FPF3.3 (%<3.3 μm) of 82.3 ± 2.1 during the first sampling period. For the LC Plus nebulizer, FPF5.8 was 92.3 ± 0.1 and FPF3.3 was 67.0 ± 0.7 during the first sampling period. The mass median aerodynamic diameter (MMAD) increased from 1.67 ± 0.05 to 1.77 ± 0.04 μm (Acorn II nebulizer) and from 2.09 ± 0.01 to 2.20 ± 0.01 μm (Pari LC Plus nebulizer) from the first to the second sampling periods. These changes in MMAD were statistically significant by paired t test. Multivariate data analysis showed that this could be explained systematically by greater drug deposition on stages with larger cutoff sizes and reduced drug deposition on stages with smaller cutoff sizes rather than multimodal deposition or other anomalies in size distribution.  相似文献

Arc repressor is a homodimeric protein with a ribbon‐helix–helix fold. A single polar‐to‐hydrophobic substitution (N11L) at a solvent‐exposed position leads to population of an alternate dimeric fold in which 310 helices replace a β‐sheet. Here we find that the variant Q9V/N11L/R13V (S‐VLV), with two additional polar‐to‐hydrophobic surface mutations in the same β‐sheet, forms a highly stable, reversibly folded octamer with approximately half the?α‐helical content of wild‐type Arc. At low protein concentration and low ionic strength, S‐VLV also populates both dimeric topologies previously observed for N11L, as judged by NMR chemical shift comparisons. Thus, accumulation of simple hydrophobic mutations in Arc progressively reduces fold specificity, leading first to a sequence with two folds and then to a manifold bridge sequence with at least three different topologies. Residues 9–14 of S‐VLV form a highly hydrophobic stretch that is predicted to be amyloidogenic, but we do not observe aggregates of higher order than octamer. Increases in sequence hydrophobicity can promote amyloid aggregation but also exert broader and more complex effects on fold specificity. Altered native folds, changes in fold coupled to oligomerization, toxic pre‐amyloid oligomers, and amyloid fibrils may represent a near continuum of accessible alternatives in protein structure space.  相似文献

Expansion of woody vegetation into areas that were historically grass-dominated is a significant contemporary threat to grasslands, including native tallgrass prairie ecosystems of the Midwestern United States. In tallgrass prairie, much of this woody expansion is concentrated in riparian zones with potential impacts on biogeochemical processes there. Although the effects of woody riparian vegetation on denitrification in both riparian soils and streams have been well studied in naturally wooded ecosystems, less is known about the impacts of woody vegetation encroachment in ecosystems that were historically dominated by herbaceous vegetation. Here, we analyze the effect of afforestation and subsequent woody plant removal on riparian and benthic denitrification. Denitrification rates in riparian soil and selected benthic compartments were measured seasonally in naturally grass-dominated riparian zones, woody encroached riparian zones, and riparian zones with woody vegetation removed in two separate watersheds. Riparian soil denitrification was highly seasonal, with the greatest rates in early spring. Benthic denitrification also exhibited high temporal variability, but no seasonality. Soil denitrification rates were greatest in riparian zones where woody vegetation was removed. Additionally, concentrations of nitrate, carbon, and soil moisture (indicative of potential anoxia) were greatest in wood removal soils. Differences in the presence and abundance of benthic compartments reflected riparian vegetation, and may have indirectly affected denitrification in streams. Riparian soil denitrification increased with soil water content and NO3 ?. Management of tallgrass prairies that includes removal of woody vegetation encroaching on riparian areas may alter biogeochemical cycling by increasing nitrogen removed via denitrification while the restored riparian zones return to a natural grass-dominated state.  相似文献
