Abstract: We have identified previously a synaptic membrane-associated protein, PP59, that serves as a substrate for cyclic AMP-dependent protein kinase and is enriched in rat cerebellum. We show here that PP59 can be extracted from synaptic plasma membranes with a combination of 2% Triton X-100 plus 1 M KCl. A 290-fold purification of PP59 was achieved by selective solubilization, followed by continuous-elution preparative gel electrophoresis. To determine the amino acid sequence surrounding the cyclic AMP-dependent protein kinase phosphorylation site within PP59, the partially purified 32 P-phosphorylated protein was digested with chymotrypsin, and radiolabeled peptides were purified by sequential reversed-phase HPLC in two different solvent systems. Automated Edman degradation revealed a single phosphorylation site contained within the sequence Ala-Arg-Glu-Arg-Ser-Asp-Ser(P)-Thr-Gly-Ser-Ser-Ser-Val-Tyr. No strong sequence homology to this peptide fragment with other known peptides or proteins in the SwissProt, PIR, or GenPept databases could be found. A synthetic peptide containing this unique 14-amino acid sequence was used to develop polyclonal anti-peptide antibodies that were affinity-purified and shown to recognize intact PP59 as determined by western blotting. These antibodies specifically inhibited the phosphorylation of PP59 by cyclic AMP-dependent protein kinase in an in vitro phosphorylation assay containing synaptic plasma membranes. 相似文献

Nitrogen Addition Could Shift Plant Community Composition in a Restored California Salt Marsh 总被引:6,自引:0,他引:6
At a salt marsh restoration site, fertilizer trials to improve height growth of Spartina foliosa (a C4 perennial grass that can reach 140 cm) appeared to favor Salicornia bigelovii (an annual C3 succulent under 40 cm tall) where the two species co-occurred on the marsh plain. This observation prompted a field experiment to examine the potential for nitrogen (N) addition to shift community composition. Without N addition, total stem length and stem density of S. foliosa did not respond to the presence or absence of S. bigelovii. But where N was added, S. foliosa growth increased only where S. bigelovii was removed from plots. S. bigelovii responded strongly to fertilizer, with mean heights matching those of S. foliosa and 600% increases in biomass, branching, and seed production (to more than 1 million seeds/m2). Soil N also increased seasonally where S. bigelovii was present, suggesting that this species may aid accumulation of N at restoration sites with poor soils. S. foliosa growth is greatest at lower elevations along tidal creeks where it occurs alone. Beyond creek edges, where S. bigelovii and other potential competitors occur, S. foliosa is unlikely to grow tall even with N addition. Thus, there is little point in trying to force mixed-species stands to provide tall S. foliosa for nesting by an endangered bird, Rallus longirostris levipes (the Light-footed Clapper Rail). A marsh construction design that maximizes tidal creek edges is thus recommended when restoration goals include providing habitat for clapper rails. 相似文献

Summary Cultures of maize (Zea mays L.) endosperm grown in vitro for over 3 years were examined cytologically. Conditions of aneuploidy and polyploidy were noted.
Chromosome numbers ranged from 21 to over 200, with 30 to 60 being observed most often. Although a few extra large cells with
polyploid nuclei were scattered throughout the smear preparation, a large proportion of the interphase nuclei appeared similar
in volume and probably contained a near normal complement of chromosomes. Anaphase bridges were the most commonly observed
chromosome aberration. No cell divisions were observed the first 24 hr after transfer. From 2 to 8 days after transfer the
proportion of cells in division was relatively constant with a mitotic index of approximately 5.5%. The proportion of cells
in division began to decline 8 days after transfer and in the final sample taken after 13 days only 2.6% of the cells were
in division. Examples of localized synchrony were observed and mitotic indices for individual cell clumps ranged from 0 to
17%.

The cave environment is consistently radically different than the surface environment because it lacks light, and animals adapting to cave life are subject to strong selective forces much different than those experienced by their ancestors who evolved in the presence of light. As such, their divergence from surface ancestors and eventual speciation is likely to be driven by the shift in ecology. We report here that hybrids between cave and surface Astyanax mexicanus fishes produce offspring with allelic frequencies that differ significantly from Mendelian expectations both for transmission ratios and for independent assortment of unlinked markers. Comparison of allelic content of DNA from fin clips and sperm pools show that the transmission ratio distortion likely occurs during spermatogenesis. Departures from expectations of independent assortment are essentially epistatic phenomena generating linkage disequilibrium. A novel analysis of the epistatic interactions reveals an apparent network of interactions among genes known or suspected to be involved in cave adaptation, implying that the epistasis arose as a “by product” of the divergence due to cave adaptation. 相似文献

Gender differences in the incidence of symptomatic hip osteoarthritis (OA), changes in hip cartilage volume and hip joint space and rates hip arthroplasty of older people are reported in the literature. As the rate of progression of OA is in part mechanically modulated it is possible that this gender bias may be related to inherent differences (if they exist) in walking mechanics between older males and females. The purpose of this study was to examine potential mechanisms for gender differences in hip joint mechanics during walking by testing the hypotheses that females would exhibit higher hip flexion, adduction and internal rotation moments but not significantly greater normalized ground reaction forces (GRFs). Forty-two healthy subjects (21 male, 21 female), ages 50–79 yr were recruited for gait analysis. In support of the hypotheses, greater external hip adduction and internal rotation along with hip extension moments were found for females compared to males after normalizing for body size for all self-selected walking speeds. Differences in walking style (kinematics) were the main determinants in the joint kinetic differences as no differences in the normalized GRFs were found. As external joint moments are surrogate measures of the joint contact forces, the results of this study suggest the hip joint stress for the female population is higher compared to male population. This is in favor of a hypothesis that the increased joint contact stress in a female population could contribute to a greater joint degeneration at the hip in females as compared with males. 相似文献

'15N signatures of fossil peat were used to interpret past ecosystem processes on tectonically active subantarctic Macquarie Island. By comparing past vegetation reconstructed from the fossil record with present-day vegetation analogues, our evidence strongly suggests that changes in the '15N signatures of fossil peat at this location reflect mainly past changes in the proportion of plant nitrogen derived from animal sources. Associated with uplift above sea level over the past 8,500 years, fossil records in two peat deposits on the island chronicle a change from coastal vegetation with fur and elephant seal disturbance to the existing inland herbfield. Coupled with this change are synchronous changes in the '15N signatures of peat layers. At two sites 15N-enriched peat '15N signatures of up to +17 were associated with a high abundance of pollen of the nitrophile Callitriche antarctica (Callitrichaceae). At one site fossil seal hair was also associated with enriched peat '15N. Less 15N enriched '15N signatures (e.g. -1.9 to +3.9) were measured in peat layers which lacked animal associated C. antarctica and Acaena spp. Interpretation of a third peat profile indicates continual occupation of a ridge site by burrowing petrels for most of the Holocene. We suggest that 15N signatures of fossil peat remained relatively stable with time once deposited, providing a significant new tool for interpreting the palaeoecology. 相似文献

Administration of purified pertussis toxin to rats induced persistent tachycardia, (observed in conscious rats but not after pithing); as little as 0.05 microgram/100 g produced a significant effect. Pertussis toxin-treatment did not affected the pressor response produced in the pithed rats by the alpha 2-adrenergic agonist methoxamine but markedly diminished the pressor effect of the alpha 2-adrenergic agonists clonidine and azepexole. A role of adenylate cyclase inhibition in the action of postsynaptic vascular alpha 2-adrenergic receptors is suggested. 相似文献
