The effects of logging on three species of common skinks were estimated from censuses in four age classes of forest: unlogged, just logged, 1-year logged and 10–15 year regrowth. The effects of topography (ridge and gully) were examined in each age class. Afire in November 1980 occurred fust after the initial census was completed. Another census was taken in December 1980 to assess its immediate effects. Further censuses were carried out each December from 1981 to 1984. An intense drought overlapped from 1980 to 1983 with the census period. Lampropholis guichenoti occurred in about equal numbers in unlogged and recently logged forest, but its numbers were reduced in the 10–15 year regrowth forest. This was attributed to changes in the amount and pattern of sunlight reaching the ground. A similar pattern of response was found for Lampropholis delicata. The numbers of Eulamprus heatwolei, a gully species requiring partial shade, were lowest in the exposed, recently-logged forest, but had increased in the 10–15 year regrowth class to about equal their numbers in unlogged forest. Fire reduced the numbers ofL. guichenoti on ridges but had no immediate impact on numbers of tire other species, while drought markedly depressed numbers of all species. Lampropholis guichenoti recovered more quickly from the drought than did L. delicata, but the numbers of E. heatwolei were still declining 19 months after the drought broke. The drought also revealed a habitat (ridge/gully) difference between the two Lampropholis species. The management of these species in commercial forests requires that some areas, particularly gullies, be reserved and the logging sequence modified to prevent the creation of widespread stands of uniform regrowth.  相似文献

Background aims

Multipotent mesenchymal stromal cell (MSC)-based medicines are extensively investigated for use in regenerative medicine and immunotherapy applications. The International Society for Cell and Gene Therapy (ISCT) proposed a panel of cell surface molecules for MSC identification that includes human leukocyte antigen (HLA)-DR as a negative marker. However, its expression is largely unpredictable despite production under tightly controlled conditions and compliance with current Good Manufacturing Practices. Herein, we report the frequency of HLA-DR expression in 81 batches of clinical grade bone marrow (BM)-derived MSCs and investigated its impact on cell attributes and culture environment.

Methods

The levels of 15 cytokines (interleukin [IL]-1β, IL-4, IL-6, IL-10, IL-17A, IL-17F, IL-21, IL-22, IL-23, IL-25, IL-31, IL-33, interferon-γ, soluble CD40 ligand and tumor necrosis factor-α) were determined in sera supplements and supernatants of BM-MSC cultures. Identity, multipotentiality and immunopotency assays were performed on high (>20% of cells) and low (≤20% of cells) HLA-DR+ cultures.

Results

A correlation was found between HLA-DR expression and levels of IL-17F and IL-33. Expression of HLA-DR did neither affect MSC identity, in vitro tri-lineage differentiation potential (into osteogenic, chondrogenic and adipogenic lineages), nor their ability to inhibit the proliferation of stimulated lymphocytes.

Discussion

Out of 81 batches of BM-MSCs for autologous use analyzed, only three batches would have passed the ISCT criteria (<2%), whereas 60.5% of batches were compliant with low HLA-DR values (≤20%). Although a cause–effect relationship cannot be drawn, we have provided a better understanding of signaling events and cellular responses in expansion culture conditions relating with HLA-DR expression.  相似文献

Abstract: Despite its importance for wildlife, most forests in the Pacific Northwest contain low volumes of large downed wood compared to fine woody debris (FWD). We used a replicated experiment to compare short-term responses of deer mice (Peromyscus maniculatus) and western red-backed voles (Clethrionomys californicus) among 3 arrangements of FWD: piled, lopped and scattered, and pile burning, a commonly used method of fuel reduction in commercial Douglas fir (Pseudotsuga menziesii) forests in southwest Oregon, USA. We assessed habitat use, density, and survival of mice and voles during 2 consecutive summers (Jun—Aug 1999 and 2000). Both mice and voles used FWD cover disproportionately from its availability, and they differed in their responses to specific FWD arrangements. Mice used piled FWD (proportional use = 37.0%, 90% CI = 33.0–44.0) 43% more than expected (26.0). Number of mice captured (x̄ = 1.9 mice, 90% CI = 1.5–2.5) and index of home range size (x̄ = 4.8 m, 90% CI = 0.7–8.9) at individual FWD piles decreased up to 16% and increased up to 50%, respectively, for each 1-m increase in distance from piles. Voles used all FWD cover classes in proportion to availability, but number of voles captured increased slightly (x̄ = 0.016 voles/m, 90% CI = 0.001–0.031) for each 1-m increase in distance from piles. Piled FWD had no discernable effect on population density and apparent survival of mice, but analyses had low power (0.25, 0.67). Our results suggest that piling FWD would benefit deer mice, whereas lopped and scattered FWD might benefit voles. Abstract: We chemically restrained free-ranging coyotes (Canis latrans), red foxes (Vulpes vulpes), and raccoons (Procyon lotor) using medetomidine antagonized by atipamezole. All coyotes and 80% of red foxes were sedated with mean ± standard deviation doses of 0.12 ± 0.02 mg/kg and 0.14 ± 0.02 mg/kg medetomidine, respectively. Seventy-seven percent of raccoons were sedated with 0.21 ± 0.05 mg/kg medetomidine. In all species we observed occasional movement, muscle rigidity, and partial-arousal during sedation. Animals were alert within 4.3–8.6 ± 3.5–8.4 min following atipamezole at 0.4 mg/kg. Medetomidine and atipamezole provided safe handling in most animals and rapid recovery without use of a controlled substance. At these doses, biologists in the field should be prepared to administer a supplementary dose of medetomidine to some animals depending on ambient conditions and the objectives of the restraint event.  相似文献

Abstract: The marbled murrelet (Brachyramphus marmoratus) is a small Pacific seabird with a breeding range that extends from the Aleutian Islands to central California. Throughout most of its breeding range, it uses mature and old-growth coniferous forests as nesting habitat. Although most murrelets seem to nest within 60 km of the coast, occupied nesting habitat has been identified as far as 84 km from the ocean in Washington state. Due to the extensive inland distances within which birds are known to breed, the area requiring surveys to identify breeding sites can be enormous. Therefore, the standard 2-year survey protocol can be expensive and time-consuming for forest management agencies and companies to administer. We developed a logistic regression model to determine whether a suite of forest structural characteristics could be used to reliably predict occupancy of a forest patch by marbled murrelets. We tested the performance of the final model using cross-validation procedures and a sample of independent sites. We used 50 sites surveyed for marbled murrelets to estimate the model, and 48 independent sites were available to test model performance. All 50 sites were on private forestland owned by Rayonier located in the western lowlands of Olympic Peninsula within the Sitka spruce and western hemlock transition zones. We sampled forest habitat at each site, and we collected information on 15 explanatory variables. The best-fitting logistic regression model contained variables that measured number of canopy layers (P<0.001, approx. F test) and mistletoe (Arceuthobium sp.) abundance (P = 0.031, approx. F test). The model misclassified 2 of 33 (94% correct) unoccupied sites as occupied using a classification cut-off (c) of c=0.25. In the other direction, under cross-validation the final model misclassified 2 of 17 (88% correct) occupied sites as unoccupied. On a test of the model against an independent sample, using a classification cut-off value of c = 0.25, the final model correctly classified 36 of 48 sites (75% correct). The final model misclassified 3 of 31 occupied sites as unoccupied (90% correct). Use of predictive models could greatly reduce the amount of forest that requires surveys by screening out those sites with little probability of use and by focusing remaining effort on higher probability sites, resulting in a higher likelihood of identifying occupied sites and thereby more efficiently conserving marbled murrelet nesting habitat.  相似文献

Laboratory experiments were conducted to measure the feeding rates, preferences, survival rates, maturation times, and weight gain of four common species of grasshoppers (Orthoptera: Acrididae) on four lines of safflower (Carthamus tinctorius). When safflower lines were presented individually, all were readily consumed although consumption differed significantly according to grasshopper species, age and sex, and to safflower line. Consumption was greatest on a safflower line devoid of spines. However, grasshoppers also fed readily on spiny lines, and results of experiments in which all safflower lines were presented together indicated preferences that did not necessarily correspond to the results of the single-line feeding trials. Camnula pellucida, a graminivorous species, did feed on safflower, but restricted feeding almost exclusively to stem cambium, whereas Melanoplus bivittatus, M. packardii and M. sanguinipes fed mainly on the heads, flowers and leaves. Highly significant differences in grasshopper maturation time and weight gain were apparent among grasshopper species, and among safflower lines. Seedtec-5, the line which was least digestible, least consumed and least preferred nevertheless yielded rapid development and the greatest body weights of grasshoppers. All species of Melanoplus developed more quickly and gained more weight on safflower than on wheat. One species, M. packardii, was more able to utilise safflower than its congeners, and may gain competitive advantage if safflower becomes widely grown.  相似文献

SYNOPSIS. Carcinonemertes errans is a nemertean worm, the juvenilesof which are found as epibionts on the Dungeness crab, Cancermagister, in close association with the arthrodial membranesof the crabs. The juvenile nemerteans appear to have no meansof taking in particulate food but survive for many months onthe surface of the host. We show that the juvenile C. erransare capable of removing amino acids from dilute solution insea water, that the water near the arthrodial membranes wherethe worms are found contains high concentrations of primaryamines, and that there is a low resistance pathway for low molecularweight amino acids across the arthrodial membrane examined invitro.  相似文献
