Abstract

Opportunistic sightings and strandings of Caperea marginata (n=196) from the vicinity of Australia and New Zealand (1884 to early 2007) were used to relate geographic and temporal patterns to oceanographic and broad-scale climatic variability. Records were not uniformly distributed along the coast and more (69%) were from Australia than New Zealand. Seven coastal whale ‘hotspots’ were identified which accounted for 61% of records with locality data. Half of the hotspot records were from southeast (37) and northwest (20) Tasmania—others each had 9–15 events. Upwelling and/or high zooplankton abundance has been documented near all whale hotspots. Records of C. marginata occurred in all months, with 75% in spring and summer. Inter-annual variability showed broad agreement between increased whale records (usually in spring/summer) and strongly positive ‘Niño 3.4’ during 1980–1995 but not thereafter. Coastal upwelling and productivity increase during climatic phenomena such as El Niño and are likely to be quickly beneficial to plankton-feeding whales such as C. marginata.  相似文献

Background  

Tenascins are a family of glycoproteins found primarily in the extracellular matrix of embryos where they help to regulate cell proliferation, adhesion and migration. In order to learn more about their origins and relationships to each other, as well as to clarify the nomenclature used to describe them, the tenascin genes of the urochordate Ciona intestinalis, the pufferfish Tetraodon nigroviridis and Takifugu rubripes and the frog Xenopus tropicalis were identified and their gene organization and predicted protein products compared with the previously characterized tenascins of amniotes.  相似文献

Background  

Trichomonosis caused by Trichomonas vaginalis is the number one, non-viral sexually transmitted disease (STD) that affects more than 250 million people worldwide. Immunoglobulin A (IgA) has been implicated in resistance to mucosal infections by pathogens. No reports are available of IgA-reactive proteins and the role, if any, of this class of antibody in the control of this STD. The availability of an IgA monoclonal antibody (mAb) immunoreactive to trichomonads by whole cell (WC)-ELISA prompted us to characterize the IgA-reactive protein of T. vaginalis.  相似文献

Aims: When subjected to dynamic temperatures surpassing the expected maximum growth temperature, Escherichia coli K12 MG1655 shows disturbed growth curves. These irregular population dynamics were explained by considering two subpopulations, i.e. a thermoresistant and a thermosensitive one ( Van Derlinden et al. 2010a ). In this paper, the influence of the initial cell concentration on the subpopulations’ dynamics is evaluated. Methods and Results: Experiments were performed in a bioreactor with the temperature increasing from 42 to 65·2°C (1 and 4°C h?1) with varying initial cell concentrations [6, 12 and 18 ln(CFU ml?1)]. When started from the highest cell concentration, the population was characterized by a higher overall maximum growth temperature and a higher inactivation temperature. For all experimental set‐ups, resistant cells were still growing at the final temperature of 65·2°C. Conclusions: The initial cell concentration had no effect on temperature resistance. The increase in temperature resistance of the sensitive subpopulation was because of the change of the physiological state to the stationary phase. Significance and Impact of the Study: A higher initial cell concentration leads to higher heat stress adaptation when cultures reach a maximum cell concentration. The observed growth at a temperature of 65·2°C is very important for food safety and the temperature treatment of micro‐organisms.  相似文献

The ability to cultivate the cyanobacterium Arhtrospira platensis in artificially lightened photobioreactors using high energetic efficiency (quasi‐monochromatic) red LED was investigated. To reach the same maximal productivities as with the polychromatic lightening control conditions (red + blue, P/2e? = 1.275), the need to work with an optimal range of wavelength around 620 nm was first established on batch and continuous cultures. The long‐term physiological and kinetic behavior was then verified in a continuous photobioreactor illuminated only with red (620 nm) LED, showing that the maximum productivities can be maintained over 30 residence times with only minor changes in the pigment content of the cells corresponding to a well‐known adaptation mechanism of the photosystems, but without any effect on growth and stoichiometry. For both poly and monochromatic incident light inputs, a predictive spectral knowledge model was proposed and validated for the first time, allowing the calculation of the kinetics and stoichiometry observed in any photobioreactor cultivating A. platensis, or other cyanobacteria if the parameters were updated. It is shown that the photon flux (with a specified wavelength) must be used instead of light energy flux as a relevant control variable for the growth. The experimental and theoretical results obtained in this study demonstrate that it is possible to save the energy consumed by the lightening device of photobioreactors using red LED, the spectral range of which is defined according to the action spectrum of photosynthesis. This appears to be crucial information for applications in which the energy must be rationalized, as it is the case for life support systems in closed environments like a permanent spatial base or a submarine. © 2009 American Institute of Chemical Engineers Biotechnol. Four amino acids in the major light-harvesting chlorophyll (Chl) a/b complex (LHCII) that are thought to coordinate Chl molecules have been exchanged with amino acids that presumably cannot bind Chl. Amino acids H68, Q131, Q197, and H212 are positioned in helixes B, C, A, and D, respectively, and, according to the LHCII crystal structure [Kühlbrandt, W., et al. (1994) Nature 367, 614-621], coordinate the Chl molecules named a(5), b(6), a(3), and b(3). Moreover, a double mutant was analyzed carrying exchanges at positions E65 and H68, presumably affecting Chls a(4) and a(5). All mutant proteins could be reconstituted in vitro with pigments, although the thermal stability of the resulting mutant versions of recombinant LHCII varied significantly. All complexes reconstituted with the mutant proteins contained fewer chlorophyll molecules per two lutein molecules than complexes reconstituted with the wild-type protein. However, the chlorophyll-binding amino acids could not be unambiguously assigned to binding either chlorophyll a or b, as in most cases more than one chlorophyll molecule was lost due to the mutation. The changes in Chl stoichiometries suggest that in LHCII some chlorophyll positions can be filled with either Chl a or b. Only some of the point mutations in LHCII affected the ability of the apoprotein to assemble into trimeric LHCII upon insertion into isolated thylakoid membranes. Among these were exchanges of H68 with either F or L, suggesting that the stability of the LHCII trimer significantly depends on this amino acid or the Chl molecule named a(5) that is attached to it and is located close to the center of the trimeric complex. The ion pair bridge between E65 and R185 in LHCII does not appear to be essential for the proper folding of the protein.  相似文献

Cell size, cell growth, and cell cycle events are necessarily intertwined to achieve robust bacterial replication. Yet, a comprehensive and integrated view of these fundamental processes is lacking. Here, we describe an image‐based quantitative screen of the single‐gene knockout collection of Escherichia coli and identify many new genes involved in cell morphogenesis, population growth, nucleoid (bulk chromosome) dynamics, and cell division. Functional analyses, together with high‐dimensional classification, unveil new associations of morphological and cell cycle phenotypes with specific functions and pathways. Additionally, correlation analysis across ~4,000 genetic perturbations shows that growth rate is surprisingly not predictive of cell size. Growth rate was also uncorrelated with the relative timings of nucleoid separation and cell constriction. Rather, our analysis identifies scaling relationships between cell size and nucleoid size and between nucleoid size and the relative timings of nucleoid separation and cell division. These connections suggest that the nucleoid links cell morphogenesis to the cell cycle.  相似文献
