The phenological development of nine Chilean accessions of Medicagopolymorpha, collected along a northsouth aridity gradient,and of two commercial cultivars of the same species, were comparedin 12 sequential outdoor sowings at Cauquenes (35°58'S,72°17'W, elev. 177 m), in the sub-humid Mediterranean climatezone of Chile. A glasshouse experiment was also conducted toevaluate the effect of photoperiod on phenophase timing. Therewas a clear gradient in precocity among the Chilean accessionsin both experiments: accessions MPO-9-88 and MPO-7-88, fromthe arid zone, were the earliest-flowering accessions, whereasMPO-36-88 from the humid Mediterranean zone was the latest.Both experiments revealed significant variation among the Chileanaccessions in the response of flowering time to variation inphotoperiod regime. Differences in days to flowering betweenthe least- (8 h) and the most- (16 h) inductive photoperiodswere lower in precocious accessions from arid and semi-aridzones, than in late-flowering accessions from more humid zones.Rate of progress to flowering, defined as the inverse of timefrom emergence to first flower appearance (1/ f), was relatedto mean diurnal temperature, or to both mean diurnal temperatureand mean photoperiod. In two early-flowering accessions fromthe arid zone, and in the Australian cultivar CircleValley, 1/ f was affected significantly (P < 0.05)by both temperature and photoperiod. Leupeptin is a peptide which inhibits several of the lysosomal proteases. When this compound was added in low concentrations to a perfused liver, the degradation of 125I-asialo-fetuin by the liver was dramatically slowed. When 5 mg leupeptin were added to the perfusate 1 h prior to the radioactive glycoprotein, the liver retained from 70 to 90% or the radioisotope 60 min after infusing 125I-asialo-fetuin. However, untreated livers contained less than 20% of the radioactivity at that time. Subcellular fractionation experiments showed that the radioactivity accumulated in the heavy and light mitochondrial fractions (ML) of the homogenate. At 80 min after the glycoprotein was added, almost 40% of the radioactivity was still located with these fractions. Very similar inhibitory effects were seen upon treating rats intravenously with 5 mg of leupeptin 60 min prior to injection of 125I-labelled asialo-fetuin. A 7 fold increase in liver radioactivity was observed 6 hrs after the glycoprotein had been given to the treated animals. Purified human liver cathepsin B digested fetuin to about 3% of total hydrolysis and the major peptide fragment produced had an SDS-electrophoretic mobility equivalent to that of ovalbumin. 相似文献

An insect larval toxin designated CryII is produced by several subspecies of Bacillus thuringiensis and differs from the other major delta-endotoxins in these bacteria in its size, toxicity profile and presence as part of an operon with three open reading frames (ORF). Such an operon from a novel B. thuringiensis isolate has been cloned and differs from one previously characterized in the following ways: (a) the size and number of amino acid repeats in one of the ORFs; (b) the smaller size of the CryII protoxin and the presence of a unique 110-kDa CryII-related antigen; and (c) high larvicidal activity for a particular Lepidopteran but low activity for a Dipteran. Various subclones of this operon were introduced into a plasmid-free B. thuringiensis strain and only the cryII gene was found to be necessary for protoxin accumulation. 相似文献

The major function of the Haptoglobin (Hp) protein is to control trafficking of extracorpuscular hemoglobin (Hb) thru the macrophage CD163 receptor with degradation of the Hb in the lysosome. There is a common copy number polymorphism in the Hp gene (Hp 2 allele) that has been associated with a severalfold increased incidence of atherothrombosis in multiple longitudinal studies. Increased plaque oxidation and apoptotic markers have been observed in Hp 2-2 atherosclerotic plaques, but the mechanism responsible for this finding has not been determined. We proposed that the increased oxidative injury in Hp 2-2 plaques is due to an impaired processing of Hp 2-2-Hb complexes within macrophage lysosomes, thereby resulting in redox active iron accumulation, lysosomal membrane oxidative injury, and macrophage apoptosis. We sought to test this hypothesis in vitro using purified Hp-Hb complex and cells genetically manipulated to express CD163. CD163-mediated endocytosis and lysosomal degradation of Hp-Hb were decreased for Hp 2-2-Hb complexes. Confocal microscopy using lysotropic pH indicator dyes demonstrated that uptake of Hp 2-2-Hb complexes disrupted the lysosomal pH gradient. Cellular fractionation studies of lysosomes isolated from macrophages incubated with Hp 2-2-Hb complexes demonstrated increased lysosomal membrane oxidation and a loss of lysosomal membrane integrity leading to lysosomal enzyme leakage into the cytoplasm. Additionally, markers of apoptosis, DNA fragmentation, and active caspase 3 were increased in macrophages that had endocytosed Hp 2-2-Hb complexes. These data provide novel mechanistic insights into how the Hp genotype regulates lysosomal oxidative stress within macrophages after receptor-mediated endocytosis of Hb. 相似文献
