Summary In this investigation, ammonium hydroxide and acetic acid were used as pH control agents during Bacillus thuringiensis (Bt) fermentation in a pilot scale fermentor (150-l) employing two secondary wastewater sludges from two different wastewater treatment plants (CUQS and JQS) and semi-synthetic soybean meal medium as raw materials. Regardless of the cultivation medium, a substantial increase in total cell count, spore count, protease activity and entomotoxicity was achieved when the pH of the culture was controlled using NH4OH/CH3COOH. At harvest, total cell count increased by almost 17%, 33% and 25%; protease activity was enhanced by 12%, 33% and 53% and maximal spore count augmented by almost 28%, 48% and 33% in CUQS, JQS and soybean medium, respectively. Entomotoxicity potency was improved by 22%, 21% and 14% in CUQS, JQS and soybean medium, respectively compared to results obtained with NaOH/H2SO4 as pH control agents. A higher entomotoxicity was also observed using sludge compared to the soybean medium. This improvement of the Bt process performance was a consequence of the addition of rapidly utilizable carbon and nitrogen source through pH control, which stimulated endotoxin production in the crystal and enhanced sporulation.  相似文献

Vitamin K is involved in the γ-carboxylation of the vitamin K-dependent proteins, and vitamin K epoxide is a by-product of this reaction. Due to the limited intake of vitamin K, its regeneration is necessary and involves vitamin K 2,3-epoxide reductase (VKOR) activity. This activity is known to be supported by VKORC1 protein, but recently a second gene, VKORC1L1, appears to be able to support this activity when the encoded protein is expressed in HEK293T cells. Nevertheless, this protein was described as being responsible for driving the vitamin K-mediated antioxidation pathways. In this paper we precisely analyzed the catalytic properties of VKORC1L1 when expressed in Pichia pastoris and more particularly its susceptibility to vitamin K antagonists. Vitamin K antagonists are also inhibitors of VKORC1L1, but this enzyme appears to be 50-fold more resistant to vitamin K antagonists than VKORC1. The expression of Vkorc1l1 mRNA was observed in all tissues assayed, i.e. in C57BL/6 wild type and VKORC1-deficient mouse liver, lung, and testis and rat liver, lung, brain, kidney, testis, and osteoblastic cells. The characterization of VKOR activity in extrahepatic tissues demonstrated that a part of the VKOR activity, more or less important according to the tissue, may be supported by VKORC1L1 enzyme especially in testis, lung, and osteoblasts. Therefore, the involvement of VKORC1L1 in VKOR activity partly explains the low susceptibility of some extrahepatic tissues to vitamin K antagonists and the lack of effects of vitamin K antagonists on the functionality of the vitamin K-dependent protein produced by extrahepatic tissues such as matrix Gla protein or osteocalcin.  相似文献

Bemisia tabaci is a complex of putative species that exhibit a strong geographical pattern. Crossing experiments have revealed various degrees of reproductive isolation between these nascent species, ranging between fertile first‐generation hybrids (F1) and no F1 at all. However, the relevance of these results under natural conditions is generally not known. The worldwide invasion of the putative species Middle East‐Asia Minor 1 (MEAM1) has caused secondary contacts between allopatric species, which in turn provide an opportunity to detect potential hybrids in nature. A total of 346 female B. tabaci were collected in 2003 and 2005 in the North East of Morocco and assigned to MEAM1 (119), Mediterranean (Med) (225) and a new putative species (2) using mitochondrial cytochrome oxidase (mtCOI) gene sequences. MEAM1 and Med individuals were characterized at seven microsatellite loci. MEAM1 and Med were found to be sympatric in 11 of 12 samples (6 fields/year). As previously reported from Spain, MEAM1 frequency decreased over time. The genetic data are consistent with a recent introduction of MEAM1. A Bayesian clustering analysis (Structure ) distinguished two groups, which were 100% consistent with the mtCOI groups. From several lines of evidence, two individuals were identified as hybrids. Assignment profiles using NewHybrids and allele composition indicated that they were not F1 hybrids. The results are discussed in relation to the secondary endosymbiont infection status determined on a sample of individuals, and the contrasting outcomes of the reported crossing experiments between MEAM1 and Med.  相似文献

Terrestrial gastropods occur in many North African localities in Eocene continental deposits. Here we analyse the faunal assemblage from the Hamada de Méridja Formation in southwestern Algeria, dated as Early to Middle Eocene on the basis of charophytes. The assemblage consists of three closely related species that to date have been classified either in the extant Madagascan genus Leucotaenius v. Martens, 1860, or in the SW European Eocene genera Romanella Jodot, 1957 and Vicentinia Jodot, 1957. This is rejected for shell morphological and phylogeographical reasons, and a new classification as Maghrebiola gen. nov. is proposed. Maghrebiola is tentatively placed in the South American family Strophocheilidae, as species from the Early Eocene Itaboraí Basin of Brazil, currently placed in the genus Eoborus Klappenbach and Olazarri, 1970 in the family Strophocheilidae, superfamily Acavoidea, have a very similar shell habitus. This record possibly extends the known geographical range of the Strophocheilidae into the African continent during the Eocene. Immigration of this stock into North Africa during the Cretaceous via a still existing plate connection is assumed. An attribution of Maghrebiola to the African family Achatinidae is unlikely for shell morphological reasons despite certain habitus similarities, although the Priabonian genera Arabicolaria and Pacaudiella from Oman most likely belong into this family, and not to the Vidaliellidae as originally proposed. Possible causes for the very low diversity of the assemblage are mainly unfavourable living conditions, i.e. a relatively dry climate resulting in sparse vegetation and only occasional presence of water bodies, which may have had increased salinities, accounting for the lack of freshwater mollusks. The absence of any competing large gastropods may possibly have facilitated high intraspecific variability leading to sympatric occurrence of three closely related species, due to the animals occupying a wide range of available ecological niches. As the species discussed here have also been attributed to the genera Romanella and Vicentinia in the Vidaliellidae, we provide an appendix with annotated characterisations of most genera of the Vidaliellidae and list the nominal species assigned to them. This family is tentatively placed in the South American superfamily Orthalicoidea; its stock would have similarly immigrated from South America, but have successfully colonized mainly SW Europe, with only one Eocene species [Romanella kantarensis (Jodot, 1936)] recognized in Algeria.  相似文献

DNA undergoes a helix-to-coil transition (also called denaturation transition) upon heating. This transition can also be facilitated by using solvent mixtures (for example water–alcohol). An increase in the hydrophobic tail of the second solvent molecule first decreases then increases the melting temperature appreciably. Measurement on 4% DNA in a series of water–alcohol mixtures shows that the helix-to-coil melting transition is driven by the solvent ability to cross the hydrophobic sugar-rich region. A new non-marine ostracod fauna from the Paleogene “hamadian deposits” outcropping west of Bechar (southwestern Algeria) has been recovered from lacustrine to fluvial deposits of the Oued Méridja section and fluvial deposits on the southern edge of the Hamada de Méridja section. Recently, these sections have been dated as late Thanetian – early Ypresian (latest Paleocene to earliest Eocene) and Ypresian – earliest Lutetian (early to earliest middle Eocene), respectively, based on charophytes. The associated ostracod fauna recovered consists of relatively mostly moderately to badly preserved specimens and comprises 14 taxa, none of which could be identified to species level in view of its poor state of preservation; we have nevertheless been able to identify and describe the following taxa: Herpetocypris sp., Cyprinotus? sp., Heterocypris? sp. 1 and sp. 2, Cypris? sp., Ilyocypris sp., Cytheroidea indet. sp. 1 and sp. 2, Limnocytheridae indet. sp. 1, Cypridoidea indet. sp. 1, Cyprididae indet. sp. 1, and Ostracoda indet. sp. 1, 2 and 3. Only Heterocypris sp. 1 occurs in both sections. Although the fauna can as yet not be related to the few other contemporaneous faunas reported from the wider palaeogeographic area, it adds important new information to our poor knowledge on Eocene non-marine ostracods in North Africa and southern Europe. The Méridja sections and area are promising regarding the discovery of more, better preserved material and further studies, and one main limitation to the correlation of the fauna is the hitherto insufficient taxonomic knowledge on many faunal elements of Eocene non-marine ostracods to which our section contributes considerably.  相似文献

Summary Egyptian cotton seeds var. Menoufi were soaked in water for 24 hours and then subjected to X-irradiation doses of 250, 500, and 10000 r. The seeds were then sown and observations on growth, flowering, and fruiting were recorded. A stimulatory effect was observed with the three radiation doses. Biocompatible and biodegradable peptide hydrogels are drawing increasing attention as prospective materials for human soft tissue repair and replacement. To improve the rather unfavorable mechanical properties of our pure peptide hydrogels, in this work we examined the possibility of creating a double hydrogel network. This network was created by means of the coassembly of mutually attractive, but self-repulsive oligopeptides within an already-existing fibrous network formed by the charged, biocompatible polysaccharides chitosan, alginate, and chondroitin. Using dynamic oscillatory rheology experiments, it was found that the coassembly of the peptides within the existing polysaccharide network resulted in a less stiff material as compared to the pure peptide networks (the elastic modulus G' decreased from 90 to 10 kPa). However, these composite oligopeptide-polysaccharide hydrogels were characterized by a greater resistance to deformation (the yield strain γ grew from 4 to 100%). Small-angle neutron scattering (SANS) was used to study the 2D cross-sectional shapes of the fibers, their dimensional characteristics, and the mesh sizes of the fibrous networks. Differences in material structures found with SANS experiments confirmed rheology data, showing that incorporation of the peptides dramatically changed the morphology of the polysaccharide network. The resulting fibers were structurally very similar to those forming the pure peptide networks, but formed less stiff gels because of their markedly greater mesh sizes. Together, these findings suggest an approach for the development of highly deformation-resistant biomaterials.  相似文献

Vision substitution by electro-stimulation has been studied since the 60s beginning with P. Bach-y-Rita. Camera pictures or movies encoded in gray levels are displayed using an electro-stimulation display device on the surface of a body part, such as the skin or the tongue. Medical-technical devices have been developed on this principle to compensate for sensory-motor disabilities such as blindness or loss of balance, or to guide specific actions, such as surgery. However, the electrical signals of stationary or moving slowly moving objects, displayed on a Tongue display unit (TDU), are quickly lost due to saturation of receptors undergoing electrostimulation. We propose to add random saccades or sparkle to the displayed visual scene to increase the quality of pattern recognition by the subjects. In the present experimental trimodal study (normal vision, TDU vision substitution, or both), we show that the presence of a moderate sparkle level enhances the perception of the direction of lines drawn on a TDU and reduces the response time.  相似文献
