The effect of altering normal cell associations and interactions on the synthesis of 5S RNA and transfer RNA (tRNA) was studied in cleaving embryos of the sea urchin, Arbacia punctulata. Cell interactions were altered: (1) by culturing cleaving embryos in the animalizing agent, Evans Blue, and in the vegetalizing agent, Li+ as LiCl and (2) by culturing dissociated cells. Control and experimental embryos each were labeled from 3 h to 6 h post fertilization with [8-3H]-guanosine. Sixteen-cell embryos, whose GTP precursor pools had been preloaded, were dissociated, labeled and cultured under conditions which prevent reaggregation. Quantitative measurements of rates of accumulation of newly synthesized 5S RNA and tRNA showed that these rates are similar in cleaving sea urchin embryos and in corresponding embryos cultured in the presence of Evans Blue and of Li+. In addition, cells dissociated from cleavage embryos and maintained under conditions which prevent reaggregation retained the ability to synthesize 5S RNA and tRNA. These results suggest that normal cell associations and interactions are not necessary for the synthesis of 5S RNA and tRNA to occur in cleaving sea urchin embryos.  相似文献

1. To study the bottom‐up linkages in arctic lakes, we treated one side of a partitioned lake with inorganic nitrogen and phosphorus for a 6‐week period each summer for 6 years starting in the summer of 1985. We took a variety of weekly measurements to determine the impact of the nutrient loading on the lake and continued weekly measurements for 2–6 years after the cessation of nutrient loading to observe the recovery of the treated side. The loading rates (2.91 mmol N m?2 day?1 and 0.23 mmol P m?2 day?1) were five times the calculated loading rates for Toolik Lake, located nearby. 2. In all 6 years of nutrient addition, phytoplankton biomass and productivity were greater in the treated sector than the reference sector. In the first 4 years of nutrient addition there was no flux of phosphorus from the mineral‐rich sediments. This changed in the last 2 years of nutrient addition as phosphorus was released to the lake. 3. The response of the animal community to increased plant production was mixed. One of the four macro‐zooplankton species (Daphnia longiremis) increased in number by about twofold in the first 5 years. However, the copepod Cyclops scutifer showed no response during the treatment phase of the study. The benthic invertebrate response was also mixed. After a 2‐year lag time the snail Lymnaea elodes increased in the treated lake sector but chironomids did not. 4. Ecosystem response to fertilisation was not controlled solely by nutrient addition because phosphorus was not recycled from the sediments until the last 2 years of nutrient addition. Phytoplankton still showed the effects of nutrient addition in the recovery period and the hypolimnion of the treated sector was still anaerobic starting at 6 m in 1996.  相似文献

A modification to Schaefer's agglutination method for serotyping mycobacteria within the Mycobacterium avium-M. intracellulare-M. scrofulaceum complex is described. The antigens are screened against polyvalent sera and subsequently a reduced range of absorbed antisera. This expedites the serotyping procedure and conserves expensive antiserum stocks.  相似文献

1. Digital video recording (DVR) devices, such as the GoPro Hero, have the potential to greatly benefit pollination ecology, but the advantages of digitally recording pollinator activity over direct human observation have not been formally assessed. 2. Two plant taxa, Lavandula angustifolia and Canna ‘sp.’, with differing floral morphology, were used to compare the value of DVR and direct observations in estimating honeybee (Apis mellifera) visitation, flower density and number of flowers visited per foraging bout. 3. The two methods yielded identical results when observing the structurally simple L. angustifolia at both high (10.54 ± 0.52 per plant) and low honeybee density (2.24 ± 0.20 per plant). However, DVR underestimated the number of flowers scored in the field of view (28.7 ± 1.8 direct vs. 22.7 ± 0.9 DVR), the number of honeybees observed (5.3 ± 0.8 direct vs. 3.7 ± 0.7 DVR) and the number of flowers visited during foraging bouts (8.3 ± 1.2 direct vs. 5.5 ± 1.0 DVR) on the more complex Canna ‘sp.’ 4. It is concluded that portable weatherproof DVR devices such as the GoPro Hero are valuable tools for pollination biologists, allowing a single researcher to make simultaneous observations of multiple plants in one or more sites, whilst also allowing the footage to be reviewed. However, DVR devices are limited by their depth and field of view when target plants are large or structurally complex.  相似文献

Summary

The distribution of serotonin-like immunoreactivity was studied in the central nervous system and the gonad of Pecten maximus. Cerebral and pedal ganglia contain a well developed serotonin-immunoreactive neuronal subpopulation, whereas positive neurons are scarce in the visceral ganglion. The distribution pattern of immunoreactive elements in the gonad indicate that serotonin is involved in peripheral neurotransmission of this organ. Seasonal variations of monoamines (serotonin, dopamine and noradrenaline) have been investigated in the nervous system using HPLC. Lower concentrations of serotonin are observed during winter in the central nervous system; dopamine levels of the visceral ganglion are correlated to gonadal growth.  相似文献
