High level expression of the major auxin-binding protein (ABP1) from maize (Zea maysL.) has been used to demonstrate that the machinery for retaining proteins in the endoplasmic reticulum (ER) of insect cells functions efficiently throughout the baculovirus infection cycle. Immuno-localization showed wild-type ABP1 (ABP1-KDEL) to be targeted to the lumen of the ER, in accordance with its signal peptide and carboxyterminal KDEL ER-retention signal. The protein accumulated in dilations of the ER, and none was detected at the cell surface. Immunoblotting of concentrated culture medium confirmed that ABP1-KDEL was not secreted at a detectable level. In contrast, when the carboxyterminus was mutated to KEQL, secretion of the baculovirus-expressed protein was readily detected. Immunolocalization and immunoblotting demonstrated that a high proportion of the ABP1-KEQL protein was secreted at the cell surface and into the culture medium. The data demonstrate that the ER of insect cells has a great capacity to retain proteins and that this property is largely unaffected by the cellular disruption caused by baculovirus replication. 相似文献

Use of lichen secondary metabolites as antifeedants to protect higher plants from damage caused by slug feeding 总被引:1,自引:0,他引:1
Extracts of 15 species of lichen were tested for antifeedant properties against the field slug Deroceras reticulation (Müller) (Mollusca: Pulmonata). All but three showed some activity and two of these were from endolithic species. The most effective extract was from Letharia vulpina. The major active compound from this species, vulpinic acid, was tested for dose response, applied as a foliar spray to turnip plants and as a dressing to wheat seeds in laboratory experiments. The post-mortem history of a prolific Maastrichtian ammonite and nautiloid fauna preserved as phosphatic steinkerns in chalk of the upper Miria Formation of Western Australia is described. Sediment infilling of phragmocones, required for their fossilisation, was accomplished by means of perforations in the shell wall induced by the activity of abundant endoliths. These include borings ascribed to clionid sponges (Entobia), thallophytes, polychaete worms (including Caulostrepis and probable Maeandropolydora), phoronids (Talpina and Gnathichnus) and others of conjectural origin. Sediment infilling by this mechanism is considered to be more applicable to the taphonomy of phragmocones in general than sediment entry through the siphuncle and to be indicative of low sedimentation rates for the hosting strata. Nonheteromorph ammonites, and the nautiloid Cimomia, are preserved almost exclusively as phragmocones in the upper Miria Formation, and are numerically subordinate to the heteromorph Eubaculites which, together with Glyptoxoceras, is preserved predominantly as body chambers. This phragmocone/body chamber preservational contrast is attributed to the influence of shell shape on pre-burial mechanical abrasion. The apparent dominance of Eubaculites is considered to be largely a preservational artifact and ascribed to the ease with which body chambers were infilled, and thereby favoured for steinkern formation, relative to phragmocones. Phragmocones, or parts thereof. not filled with sediment were eliminated from the fossil record by diagenetic aragonite dissolution. Many ammonites with open umbilici have the early whorls missing. This we attribute to the trapping of sediment in the umbilicus prior to burial, preventing endolith attack whereby the early whorls avoided a sediment infilling. However, the protected inner whorls of involute ammonites and Cimomia, which have closed umbilici, and the inner whorls of evolute ammonites where a cemented umbilical plug supported the mouldic cavity left by shell dissolution, were preserved and were commonly infilled with calcite spar later in diagenesis. The dearth of cephalopod fossils in chalk underlying the upper Miria Formation is ascribed to diagenesis in which aragonite dissolution was not preceded by cementation. 相似文献

Owing to the complex interactions among climate, plants, cattle grazing, and land management practices, the impacts of climate change on cattle have been hard to predict. Predicting future grassland ecosystem functioning relies on understanding how changes in climate alter the quantity of forage produced, but also forage quality. Plant protein, which is a function of plant nitrogen concentrations, and digestible energy limit the performance of herbivores when in short supply; moreover, deficiencies can be expensive to mitigate. To better understand how changes in temperature and precipitation would affect forage protein and energy availability, we analyzed over 21 000 measurements of cattle fecal chemistry acquired over 14 years in the continental US. Our analysis of patterns in forage quality among ecologically defined regions revealed that increasing temperature and declining precipitation decreased dietary crude protein and digestible organic matter for regions with continental climates. Within regions, quality also declined with increased temperature; however, the effects of precipitation were mixed. Any future increases in precipitation would be unlikely to compensate for the declines in forage quality that accompany projected temperature increases. As a result, cattle are likely to experience greater nutritional stress in the future. If these geographic patterns hold as a proxy for future climates, agriculture will require increased supplemental feeds or the consequence will be a decrease in livestock growth. 相似文献

Abstract Scavenging bird deterrence frequently fails due to habituation. We demonstrated such habituation by gulls and corvids to blank rounds used in a dawn-to-dusk regime at a landfill site in southern England. We then combined blank rounds with live rounds and shot birds whenever they attempted to land. Gull numbers declined significantly despite only 1.9% of the population being shot. Corvid numbers returned to precontrol levels despite 52.7% of the population being shot. We suggest that shooting reduces gull habituation to blank rounds but is ineffective at reducing habituation by corvids. 相似文献
