Hucho taimen are listed as endangered in China. The population size has declined recently, prompting an increase in the level of listing from grade three in 2002 to grade five in 2006. We analyzed the genetic diversity of wild populations using 17 microsatellite markers to establish a scientific basis for conservation of this species. We collected tissue samples from four populations in the Heilongjiang River basin: Huma River (HM), Hutou (HT), Haiqing (HQ), and Zhuaji (ZJ). A total of 21 loci were amplified, 18 of which were polymorphic. The number of alleles per locus ranged from 2 to 9 (mean: 4.1905). There were 13 highly polymorphic loci and 5 moderately polymorphic loci. Analysis of five genetic diversity parameters (Na, Ne, Ho, He, and PIC) suggested moderate levels of diversity within the populations. The populations were ranked HT > HQ > ZJ > HM, but the differences in diversity were not statistically significant (P > 0.05). A comparison of variation among all four populations suggested Hardy–Weinberg disequilibrium at 20% of the loci. Genetic differentiation (Fst) was 0.0644 and the gene flow among populations was estimated at 3.36 individuals per generation. The majority of diversity (93.88%) occurred among individuals within a population. In contrast, relatively little (6.12%) of the genetic diversity was distributed between the populations. An analysis of genetic differentiation and genetic distance between pairs of populations revealed that both parameters were higher in comparisons of the HM population to the HT, HQ, and ZJ populations than among the three latter populations. This suggests that the HM population has a distinct genetic structure. We hypothesize that habitat degradation and excessive fishing, not low genetic diversity, has caused the decline in H. taimen populations. The aim of this study was to characterize the electropharmacological effects of prostacyclin (PGI2) in human atrial fibers and cardiomyocytes. Atrial tissues obtained from the hearts of 28 patients undergoing corrective cardiac surgery were used. Transmembrane action potentials were recorded using a conventional microelectrode technique, and twitch force by a transducer. Effects of PGI2 (1 nM–10 µM) on action potential characteristics and contraction of atrial fibers were evaluated in normal [K]o (4 mM) and high [K]o (27 mM) in the absence and presence of cardiotonic agents. In addition, atrial and ventricular myocytes were isolated enzymatically from atrial tissues and hearts of 4 patients undergoing cardiac transplant. The effects of PGI2 on Na- and Ca-dependent inward currents (INa and ICa) of cardiomyocytes were tested. In 9 human atrial fibers showing fast-response action potentials (mean dV/dtmax = 101 ± 15 Vs–1) in 4 mM [K]o, PGI2 did not influence dV/dtmax of phase 0 depolarization even at 1 µM. However, at a concentration as low as 10 nM, PGI2 depressed spontaneous rhythms or slow-response action potentials in high-K-depolarized fibers. PGI2 also depressed delayed afterdepolarizations and aftercontractions induced by cardiotonic agents. In isolated cardiomyocytes, PGI2 reduced ICa but not INa. The present findings show that, in human atrial fibers and cardiomyocytes, PGI2 induces greater depressant effects on the slow-response action potential, ICa and triggered activity than on the fast-response action potential. To examine the association between glutathione peroxidase 1 (GPx1) gene Pro198Leu polymorphism with the development and progression of prostate cancer. A comprehensive search was conducted to identify all case-control studies of GPx1 polymorphisms and prostate cancer. Statistical analysis was performed with the software program Stata, version 11.0, and Review Manage, version 4.2. A total of 7 eligible studies relating the GPx1 polymorphism to the risk of prostate cancer were identified. The results indicated no significant association between GPx1 polymorphisms and prostate cancer susceptibility in the dominant model (random effects OR 0.75, 95 % CI 0.48-1.18), recessive model (random effects OR 0.47, 95 % CI 0.22-1.01) and co-dominant genetic model (random effects OR 0.72, 95 % CI 0.43-1.21). For the analysis of GPx1 polymorphism and progression of prostate cancer, no significant association were found in the dominant model (fixed effects OR 1.20, 95 % CI 0.95-1.52), recessive model (fixed effects OR 0.69, 95 % CI 0.48-1.00) and co-dominant genetic model (fixed effects OR 0.95, 95 % CI 0.79-1.15). The study focused on the dynamics of Malondialdehyde (MDA) contents and the activities of protective enzymes in the leaves of alfalfa varieties with various resistances to Aphis medicaginis Koch. The results showed that susceptible varieties always had higher MDA contents than resistant varieties, and the MDA contents tended to rise in both susceptible and resistant varieties in period of the varieties were pierced and sucked by aphids. Superoxide dismutase (SOD), peroxidase (POD) and phenylalanine ammonia-lyase (PAL) activities in susceptible varieties were lower than those in resistant varieties, and in both susceptible and resistant varieties the SOD and POD activities tended to rise at first and then decline, and the PAL activities rose to their peaks and then tended to remain stable. In the susceptible and resistant varieties the catalase (CAT) activities appeared to rise and decline alternatively; the PPO activities in resistant varieties were lower than those in susceptible varieties in early growth, but higher than those in susceptible varieties in later growth. It follows that infested by aphids, susceptible and resistant varieties had the MDA contents, variations of SOD, POD, PAL and PPO activities were closely correlated with their aphid resistances, hence these indexes could be used as physiological indexes for testing aphid resistance of alfalfa, whereas the relations of their CAT activities to their resistances needed to be further studied. 相似文献

Background
Uterine leiomyomas, or fibroids, represent the most common benign tumor of the female reproductive tract. Fibroids become symptomatic in 30% of all women and up to 70% of African American women of reproductive age. Epigenetic dysregulation of individual genes has been demonstrated in leiomyoma cells; however, the in vivo genome-wide distribution of such epigenetic abnormalities remains unknown.Principal Findings
We characterized and compared genome-wide DNA methylation and mRNA expression profiles in uterine leiomyoma and matched adjacent normal myometrial tissues from 18 African American women. We found 55 genes with differential promoter methylation and concominant differences in mRNA expression in uterine leiomyoma versus normal myometrium. Eighty percent of the identified genes showed an inverse relationship between DNA methylation status and mRNA expression in uterine leiomyoma tissues, and the majority of genes (62%) displayed hypermethylation associated with gene silencing. We selected three genes, the known tumor suppressors KLF11, DLEC1, and KRT19 and verified promoter hypermethylation, mRNA repression and protein expression using bisulfite sequencing, real-time PCR and western blot. Incubation of primary leiomyoma smooth muscle cells with a DNA methyltransferase inhibitor restored KLF11, DLEC1 and KRT19 mRNA levels.Conclusions
These results suggest a possible functional role of promoter DNA methylation-mediated gene silencing in the pathogenesis of uterine leiomyoma in African American women. 相似文献9.
The magnetic chitosan nanocomposites have been studied intensively and been used practically in various biomedical and biological applications including enzyme immobilization. However, the loading capacity and the remained activity of immobilized enzyme based on existing approaches are not satisfied. Simpler and more effective immobilization strategies are needed. Here we report a simple catechol modified protocol for preparing a novel catechol-chitosan (CCS)-iron oxide nanoparticles (IONPs) composites carrying adhesive moieties with strong surface affinity. The ω-transaminase (ω-TA) was immobilized onto this magnetic composite via nucleophilic reactions between catechol and ω-TA. Under optimal conditions, 87.5% of the available ω-TA was immobilized on the composite, yielding an enzyme loading capacity as high as 681.7 mg/g. Furthermore, the valuation of enzyme activity showed that ω-TA immobilized on CCS-IONPs displayed enhanced pH and thermal stability compared to free enzyme. Importantly, the immobilized ω-TA retained more than 50% of its initial activity after 15 repeated reaction cycles using magnetic separation and 61.5% of its initial activity after storage at 4°C in phosphate buffered saline (PBS) for 15 days.
