Penetration of radioactive sodium and chloride into cerebrospinal fluid and aqueous humor

1. Experiments were performed on six dogs to determine the rate of penetration of Cl(33) and Na(24) across the blood-aqueous humor and blood-cerebrospinal fluid barriers after intravenous injection of the radioactive ions. The radioactivity measurements were made with an immersion type of Geiger-Müller counter. 2. The concentrations of the labelled ions in the anterior chamber and the cisterna magna increase slowly to approach that of plasma. The rate of penetration k is calculated from a simple exponential equation with the half-value interval t(0.5) or the time required for the labelled-ion concentration in the fluid to reach 50 per cent of that of plasma. The average t(0.5) for Cl(38) and Na(24) in aqueous humor are 34.3 +/- 9 and 27.3 +/- 9 minutes, respectively, while those for cerebrospinal fluid are 90 +/- 6 and 95 +/- 6 minutes, respectively. 3. A study of the radioactivity in plasma was made to determine the per cent remaining after a steady state was reached. By means of this determination the sodium and chloride space was calculated to be 33 +/- 5 per cent.

Proliferation inhibition and apoptosis promotion by dual silencing of VEGF and Survivin in human osteosarcoma

Simultaneous silencing of multiple upregulated genes is an attractive and viable treatment strat-egy for many incurable diseases including cancer.Herein we used a dual gene-targeted siRNA conjugate composed of VEGF and Survivin siRNA sequences in the same backbone to inhibit proliferation and angiogenesis in two human osteosarcoma cell lines.We synthesized siRNA sequences targeting the VEGF and Survivin genes individually (VEGF siRNA and Survivin siRNA) or simultaneously (one-chain-double-target siRNA:dual siRNA).VEGF and Survivin mRNA and protein expression levels in human osteosarcoma MG-63 and Saos-2 cells were detected by qRT- PCR and western blot analysis.VEGF and Survivin protein location and expression were evaluated by immunohistochemistry and immunofluorescence staining.MG-63 and Saos-2 cell migration,proliferation,apoptosis,and angiogenesis were detected by scratch test,MTT assay,flow cytometry,and capillary tube assay respectively.The dual siRNA induced similar downregulation of VEGF and Survivin mRNA and protein levels,compared with VEGF siRNA or Survivin siRNA alone.The dual siRNA caused greater suppression of MG-63 and Saos-2 cell migration,proliferation and angiogenesis,and promoted more cell apoptosis than VEGF siRNA or Survivin siRNA alone,suggesting that the effects of the dual siRNA on inhibiting cell proliferation,migration,and angiogenesis and promoting apoptosis were superior to those of the single-target siRNAs.Simultaneous silencing of VEGF and Survivin using the dual siRNA may be an advantageous alternative for the development of therapeutic strategies against human osteosarcoma.     

Global stability for an SEI model of infectious diseases with immigration and age structure in susceptibility

In this paper, we propose an SEI age-structured model for infectious diseases where the susceptibility depends on the age and with immigration of new individuals into the susceptible, exposed and infectious classes. The existence of a global attractor and the asymptotic smoothness of the solution semi-flow generated by the model are addressed. Using a Lyapunov functional, we show that the unique endemic equilibrium is globally asymptotically stable.     

Databases and Web Tools for Cancer Genomics Study

Publicly-accessible resources have promoted the advance of scientific discovery. The era of genomics and big data has brought the need for collaboration and data sharing in order to make effective use of this new knowledge. Here, we describe the web resources for cancer genomics research and rate them on the basis of the diversity of cancer types, sample size, omics data comprehensiveness, and user experience. The resources reviewed include data repository and analysis tools; and we hope such introduction will promote the awareness and facilitate the usage of these resources in the cancer research community.     

Identification and characterization of Mini1, a gene regulating rice shoot development

The aerial parts of higher plants are generated from the shoot apical meristem(SAM). In this study, we isolated a small rice(Oryza sativa L.) mutant that showed premature termination of shoot development and was named mini rice 1(mini1). The mutant was first isolated from a japonica cultivar Zhonghua11(ZH11) subjected to ethyl methanesulfonate(EMS)treatment. With bulked segregant analysis(BSA) and map-based cloning method, Mini1 gene was finally fine-mapped to an interval of 48.6 kb on chromosome 9. Sequence analyses revealed a single base substitution from G to A was found in the region, which resulted in an amino acid change from Gly to Asp.The candidate gene Os09g0363900 was predicted to encode a putative adhesion of calyx edges protein ACE(putative HOTHEAD precursor) and genetic complementation experiment confirmed the identity of Mini1. Os09g0363900 contains glucose-methanol-choline(GMC) oxidoreductase and NAD(P)-binding Rossmann-like domain, and exhibits high similarity to Arabidopsis HOTHEAD(HTH). Expression analysis indicated Mini1 was highly expressed in young shoots but lowly in roots and the expression level of most genes involved in auxin biosynthesis and signal transduction were reduced in mutant.We conclude that Mini1 plays an important role in maintaining SAM activity and promoting shoot development in rice.     

Sex differences in morphine-induced behavioral sensitization and social behaviors in ICR mice

Gender and genetic strain are two prominent variants that influence drug abuse. Although certain sexrelated behavioral responses have been previously characterized in ICR mice, little is known about the effects of sex on morphine-induced behavioral responses in this outbred strain. Therefore, in this study, we investigated the sex differences of morphine-induced locomotion, anxiety-like and social behaviors in ICR mice. After morphine or saline exposure for four consecutive days(twice daily), increased locomotion, more time spent in the central area, as well as attenuated rearing and self-grooming behaviors were found in morphine-treated females in an open field; no differences were found in locomotion and the time spent in the central area between male and female controls. When interacting with the samesex individuals, female controls were engaged in more social investigation, following, body contacting and self-grooming behaviors than controls; morphine exposure reduced contacting and self-grooming behaviors in females; in contrast, these effects were not found in males. These results indicate that female ICR mice are more prosocial and are more susceptible to morphine exposure than males.
