Activation of STAT3 stimulates AHSP expression in K562 cells

Studies on the chaperone proteinα-hemoglobin stabilizing protein(AHSP)reveal that abundant AHSP in erythroid cells enhance the cells’tolerance to oxidative stress imposed by excessα-hemoglobin in pathological conditions.However,the potential intracellular modulation of AHSP expression itself in response to oxidative stress is still unknown.The present study examined the effect and molecular mechanism of STAT3,an oxidative regulator,on the expression of AHSP.AHSP expression increased in K562 cells upon cytokine IL-6-induced STAT3 activation and decreased in STAT3 knock-down K562 cells.Regulation of AHSP in oxidative circumstance was then examined inα-globin-overloaded K562 cells,and real-time PCR showed strengthened expression of both AHSP and STAT3.ChIP analysis showed binding of STAT3 to AHSP promoter and binding was significantly augmented with IL6 stimulation and uponα-globin overexpression.Dual luciferase reporter assays of the wildtype and mutated SB3 element,an IL-6RE site,in the AHSP promoter in K562 cells highlighted the direct regulatory effect of STAT3 on AHSP gene.Finally,direct binding of STAT3 to SB3 site of AHSP promoter was confirmed with EMSA assays.Our work reveals an adaptive AHSP regulation mediated by the redox-sensitive STAT3 signaling pathway,and provides clues to the therapeutic strategy for AHSP enhancement.

Host plant mediation of diapause induction in the cabbage beetle, Colaphellus bowringi Baly （Coleoptera： Chrysomelidae）

The cabbage beetle, Colaphellus bowringiBaly （Coleoptera： Chrysomelidae）, is a serious pest of crucifers in China, undergoing an imaginal summer and winter diapause in the soil. The effects of host plants on diapause incidence were tested in the beetle. The ratio of adults entering diapause was significantly low when they fed on the mature leaves of Chinese cabbage Shanghaiqin （Brassica chinesis var communis） than those feeding on Chinese cabbage Suzhouqin （Brassica chinesis var communis）, radish （Raphanus sativus var longipinnatus） and stem mustard （Brassicajuncea vat tumida） at 25℃ combined with 13：11 （L： D） h. Fewer adults entered diapause on young leaves compared to physiologically aged and mature radish leaves at 25℃ combined with 13：11 （L： D） h. The effect of host plant species on diapause induction was also evident under continuously dark rearing conditions or at different photoperiods. These experimental results demonstrate that host plant mediation of diapause induction exists in the cabbage beetle. However, at temperatures ≤20℃ or photoperiods of 16：8 （L： D） h combined with 25℃, all individuals entered diapause regardless of the host plants, indicating that the effects of host plants on diapause induction could be expressed only within a limited range of temperatures and photoperiods.     

广州汉族人群DYS19、DYS389Ⅰ/Ⅱ、DYS390多态性及其单体型

用PCR结合PAGE技术观察111例广州汉族男性DYS19、DYS389Ⅰ/Ⅱ、DYS390等位基因及单体型分布状况。结果显示：广州地区汉族男性DYS19基因座观察到5种等位基因,DYS389Ⅰ观察到4种等位基因,DYS389Ⅱ观察到5种等位基因,DYS390观察到5种等位基因;χ2检验表明上述各等位基因频率分布与其他地区人群存在明显的差异。此外,还观察到72种由上述基因座共同构成的单体型,单体型多样性达0.953。Abstract: In order to apply a set of useful and high polymorphic Y?STRs in forensic practice and genetic analysis,we performed a population genetic study from Chinese.The allele distributions of the systems DYS19、DYS389Ⅰ/Ⅱ、and DYS390 were investigated in sample of 111 unrelated males from the area of Guangzhou, China.PCR products were detected using polyacrylamide gel electrophoresis and silver staining.5、4、5、5 alleles were observed in locus DYS19、DYS389Ⅰ、DYS389Ⅱ、DYS390 respectively.Different allele frequency distributions were observed when compared to other population.Haplotype frequency date of 72 different types were obtained.
